Orai1 expression pattern in tooth and craniofacial ectodermal tissues and potential functions during ameloblast differentiation.

BACKGROUND: Orai1 is a plasma membrane protein that forms the pore of the calcium release activated calcium channel. Humans with mutated Orai1 present with hereditary combined immunodeficiency, congenital myopathy and anhidrotic ectodermal dysplasia. Consistent with the ectodermal dysplasia phenotype, enamel formation and mineralization is also abnormal in Orai1 deficient patients. The expression pattern and potential functions of Orai1 in enamel formation remains unclear. To contribute toward understanding the role of Orai1 in amelogenesis we characterized ORAI1 protein developmental pattern

Decreased levels of Cx43 gap junctions result in ameloblast dysregulation and enamel hypoplasia in Gja1Jrt/+ mice.

Coordinated differentiation of the ameloblast cell layer is essential to enamel matrix protein deposition and subsequent mineralization. It has been hypothesized that this process is governed by Cx43-based gap junctional intercellular communication as oculodentodigital dysplasia (ODDD) patients harboring autosomal-dominant mutations in Cx43 exhibit enamel defects typically resulting in early adulthood tooth loss. To assess the role of Cx43 in tooth development we employ a mouse model of ODDD that harbors a G60S Cx43 mutant, Gja1(Jrt)/+, and appears to exhibit tooth abnormalities that mimic the

Total mandibular reconstruction after resection of rare "honeycomb-like" ameloblastic carcinoma--a case report.

PURPOSE: This case report describes a rare and aggressive ameloblastic carcinoma that infiltrated the mandible in a "honeycomb" pattern. METHODS: A total mandibulectomy with bilateral modified neck dissection was followed by primary reconstruction with a single free vascularised fibula flap. RESULTS: The postoperative course was uneventful. The one year follow-up revealed no signs of recurrent tumour or metastases. Nine months later distant metastases occurred in the lung. CONCLUSION: Ameloblastic carcinoma is a highly malignant lesion, which requires aggressive therapy. Prognosis is poor.

Secondary Ameloblastic Carcinoma With Craniofacial and Skull Base Invasion: En Bloc Resection and Dural Reconstruction.

BACKGROUND: Secondary ameloblastic carcinoma is a rare malignant odontogenic tumor arising through malignant transformation of a pre-existing ameloblastoma and may involve complex craniofacial compartments. When the skull base is involved, management requires integration of oncologic resection with reconstructive strategies that ensure durable intracranial-extracranial separation and structural stability. CASE PRESENTATION: A 27-year-old male with a 13-year history of recurrent mandibular ameloblastoma presented with extensive craniofacial progression involving the temporal bone, masticator a

Ameloblastic Fibroma of the Mandible Presenting as a Dentinoid Formation in an 8-Year-Old Child: A Case Report.

Ameloblastic fibroma is an uncommon, benign, mixed odontogenic tumor characterized by the presence of both epithelial and mesenchymal components, without the formation of hard tissue. It constitutes approximately 1.5%-4.5% of all odontogenic tumors and predominantly affects the posterior mandible. Most cases are observed in individuals under 22 years of age, with a slight predilection for males. The initial presentation typically involves a painless, slow-growing mass, often discovered incidentally during routine radiographic examinations. This lesion carries a risk of recurrence and potential

Ameloblastic fibroma and BRAF V600E immunohistochemistry staining pattern: case report and review of a rare entity.

Ameloblastic fibroma is a rare, benign, biphasic odontogenic tumour that typically presents in young patients and is treated conservatively with an excellent prognosis. An important differential is ameloblastoma, a more common odontogenic neoplasm that shares many of the clinical, radiological, and histological features, but requires more extensive surgery due to its potentially destructive nature. Both tumours demonstrate BRAF V600E mutations. While epithelial staining with BRAF V600E-specific immunohistochemistry is considered a surrogate marker for mutation and is supportive of the diagnosi

Are Ameloblastic Fibroma-related Lesions True Tumors?: Evidence Through CNA and BRAF Mutation Analysis.

Ameloblastic fibroma (AF) and related lesions, namely ameloblastic fibrodentinoma (AFD) and ameloblastic fibro-odontoma (AFO), span a spectrum from true neoplasms to hamartomas. The 2017 World Health Organization classification proposes that AFD and AFO are precursors to odontomas, yet their precise nature remains uncertain. This study examined 19 AF cases, 4 AFD, 15 AFO, 19 odontomas (OD, 14 complex, 5 compound), and 2 ameloblastic fibrosarcomas (AFS), focusing on clinical characteristics, recurrence, and molecular profiles. AF primarily affected individuals under 20 years (60.0% of cases), m

Primary and secondary ameloblastic carcinoma can be classified into not distinct categories but a single entity: A comparative study.

BACKGROUND: Ameloblastic carcinoma (AC) was categorized into primary-type and secondary-type AC by 2005 World Health Organization (WHO) classification, whereas it is reclassified as a single entity in the new WHO classification. However, large sample studies are required to further evaluate and reinforce the classification. MATERIALS AND METHODS: The clinical behaviors and prognosis of primary AC (n = 25) and secondary AC (n = 24) were comparatively analyzed in a retrospective study of 49 AC patients with a mean follow-up period of 64.5 months. RESULTS: Differences in all the clinical parame

Immunohistochemical Profile of Ameloblastic Carcinoma Arising from an Amyloid-Producing Odontogenic Tumour in a Miniature Dachshund.

A 13-year-old female miniature dachshund was presented with a centrally-located sublingual mass in the rostral mandibular region. The focally ulcerated growth completely covered the left (305) and right (405) premolar teeth and partially covered the right canine teeth (404). A punch biopsy sample revealed neoplastic proliferation of odontogenic epithelium arranged in irregular cords with frequent comedo-like necrosis. Following the initial diagnosis of ameloblastic carcinoma, a bilateral rostral hemimandibulectomy was performed. Although the detailed examination of the resected mass was consis

Spatio-Temporal Regulation of IGFs in Enamel Development: Molecular Mechanisms From Ameloblast Polarity to Mineralization Homeostasis.

The development of enamel relies on the precise regulation of ameloblast differentiation, enamel matrix secretion, and mineralization. The formation of enamel is crucial for the normal function of dental tissues, and promoting enamel remineralization is of significant importance for the treatment of dental caries. Understanding the underlying mechanisms of enamel development is essential for oral therapy and provides a bridge to tooth regeneration. Among various growth factors, the insulin-like growth factor (IGF) family, including IGF-1 and IGF-2, has been shown to play a key role in enamel f

Differential Expression of Immunohistochemical Markers in Ameloblastoma & Ameloblastic Carcinoma: A Systematic Review and Meta-analysis of observational studies.

BACKGROUND: Differentiating between ameloblastoma (AB) and ameloblastic carcinoma (AC) is difficult, especially when AB has atypical cytological characteristics or an uncommon clinical history. This systematic review and meta-analysis aimed to elucidate the differential expression of immunohistochemical markers between AB and AC. METHODS: We conducted a thorough search of PUBMED and SCOPUS according to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines to identify cross-sectional studies that compared the expression of immunohistochemical markers in AB

Ameloblastic fibro-odontosarcoma with mandibular bone invasion and regional lymph node metastasis in a cat: case report.

Odontogenic tumours are uncommon neoplasms in domestic animals, mostly solitary and locally infiltrative, but rarely metastatic. We report the case of a 13-year-old neutered male cat presented with a mandibular gingival neoformation. A computed tomography scan revealed an irregular neoformation with marked post-contrast enhancement, associated with lysis of the incisive bone and mandibular symphysis. Histologically, the oral mucosa and mandibular bone were infiltrated by a neoplasm consisting of a mixed population of odontogenic epithelium admixed with bundles of odontogenic ectomesenchyme, mu

MR appearance of a rare ameloblastic fibroma with formation of dental hard tissues with histopathologic correlation: a case report.

An ameloblastic fibroma with formation of dental hard tissues, which the classical name is ameloblastic fibro-odontoma (AFO), is a rare type of mixed odontogenic tumor. An 8-year-old boy was diagnosed with AFO, with an inhomogeneous high signal within the lesion shown by T2-weighted magnetic resonance imaging (MRI). Computed tomography (CT) imaging revealed a unilocular low CT value area of 24 × 19 × 26 mm with buccolingual bony expansion and cortical bone thinning on the left side of the mandible including the crown of the mandibular left second molar. In addition, multiple calcified bodies w

Management of ameloblastic fibro-odontoma in a 6-year-old girl preserving the associated impacted permanent tooth.

The ameloblastic fibro-odontoma (AFO) is a rare odontogenic tumor that occurs predominantly in children and is generally associated with unerupted teeth. The choice of treatment for this entity is conservative surgery with enucleation and its prognosis is excellent. However, preserving the associated impacted teeth may make complete removal of the lesion difficult and may explain some cases of recurrence. A case of AFO in a 6-year-old girl treated with enucleation and preservation of an impacted lower left first permanent molar is reported. After two years of follow-up, there were no signs of

Insulin-like growth factor-I receptor in the cell biology of the ameloblast: an immunohistochemical study on the rat incisor.

The distribution of IGF-I receptor is reported in the odontogenic epithelium and mesenchyme of the continuously erupting mandibular incisor of the rat by immunohistochemistry using a polyclonal antibody specific to the IGF-I receptor. Odontogenic epithelium is a unique odontogenic sequence in that all stages of the complex life cycle of the ameloblast are represented along the length of the enamel-forming aspect of the tooth. Pre-ameloblasts become post-mitotic before secreting enamel matrix. When the full thickness of the enamel has been formed, a remarkable transition in phenotype takes plac…
