Molecular determinants of two neurotoxins that regulate sodium current inactivation in rat hippocampal neurons.

We studied functional and structural differences between the two neurotoxins, wasp toxin pompilidotoxin (PMTX) and sea anemone toxin (ATXII). Although PMTX and ATXII inhibited inactivation of sodium currents both toxins had distinct actions on the lobster axon and on the rat hippocampal cells. To determine structural basis of the difference we compared arrangement of polar and non-polar amino acids of the two toxins and found that similar sequence of PMTX exist in a discrete position of three-dimensional structure of ATXII. The sequence may be responsible for the binding site in the neuronal N

Binding of an alpha scorpion toxin to insect sodium channels is not dependent on membrane potential.

The insect-specific Lqh alpha IT toxin resembles alpha scorpion toxins affecting mammals by its amino acid sequence and effects on sodium conductance. The present study reveals that Lqh alpha IT does not bind to rat brain membranes and possesses in locust neuronal membranes a single class of high affinity (Kd = 1.06 +/- 0.15 nM) and low capacity (Bmax = 0.7 +/- 0.19 pmol/mg protein) binding sites. The latter are: (1) distinct from binding sites of other sodium channel neurotoxins; (2) inhibited by sea anemone toxin II; (3) cooperatively interacting with veratridine; (4) not dependent on membra

Removal of bound calcium from nematocyst contents causes discharge.

Nematocysts are the stinging organelles of jellyfish, sea anemones and other cnidarians. Each one consists of a closed capsule filled with fluid. In the resting state, part of the surface of the capsule is inverted, forming a tubular thread, which is everted explosively on excitation. The mechanism of explosion is not yet understood, it may be relevant to exocytosis in general, as the nematocyst is a specialized type of exocytotic secretion originating in the Golgi apparatus. Picken and Skaer observed that the capsular fluid showed a large depression of freezing point, suggesting that the osmo

Animal Venoms have Potential to Treat Cancer.

The resistance to chemotherapeutics by the cancerous cells has made its treatment more complicated. Animal venoms have emerged as an alternative strategy for anti-cancer therapeutics. Animal venoms are cocktails of complex bioactive chemicals mainly disulfide-rich proteins and peptides with diverse pharmacological actions. The components of venoms are specific, stable, and potent and have the ability to modify their molecular targets thus making them good therapeutics candidates. The isolation of cancer-specific components from animal venoms is one of the exciting strategies in anti-cancer res

Biology of FGFRL1, the fifth fibroblast growth factor receptor.

FGFRL1 (fibroblast growth factor receptor like 1) is the most recently discovered member of the FGFR family. It contains three extracellular Ig-like domains similar to the classical FGFRs, but it lacks the protein tyrosine kinase domain and instead contains a short intracellular tail with a peculiar histidine-rich motif. The gene for FGFRL1 is found in all metazoans from sea anemone to mammals. FGFRL1 binds to FGF ligands and heparin with high affinity. It exerts a negative effect on cell proliferation, but a positive effect on cell differentiation. Mice with a targeted deletion of the Fgfrl1

Functional characterization of specialized immune cells in a cnidarian reveals an ancestral antiviral program.

Examining early-branching animal phyla can help reconstruct the evolutionary origins of immune cells. Here, we characterize immune-related cell programs in embryos of the sea anemone Nematostella vectensis, a model of Cnidaria, which diverged ~600 million years ago from other animals. Using a transgenic Nematostella reporter line expressing mCherry under the RLRb antiviral promoter, we identify a morphologically and transcriptomically distinct cell population activated by the viral mimic poly(I:C). These cells upregulate immune effector and regulator genes and show increased phagocytic activit

Seabather's eruption.

BACKGROUND: Seabather's eruption is an annoying pruritic dermatitis that appears on the areas covered by the bathing suit as an erythematous macular or papular dermatitis, with or without urticaria. It occurs sporadically in Florida, the Caribbean, and as far north as Bermuda. The cause is not known. METHODS: We collected information in Nassau County, Long Island, New York, about cases of all types of water-related dermatitis reported by beach personnel, health providers, and affected swimmers from 1970 through 1991. Concurrently, we surveyed all Nassau County swimming waters, especially duri

The emergence of the ectodysplasin pathway.

The genes coding for the proteins of the ectodermal dysplasin pathway, EDA, EDAR and EDA2R are found in genomes of animals that long predate vertebrates. We show that EDA, EDAR and EDA2R are expressed in the sea anemone. A fourth gene in the pathway, EDARADD, only appears with the cyclostomes. With the appearance of EDARADD, the ectodermal dysplasin system was now complete, and the skin appendages emerged, first in the form of the cyclostome's keratinized proto-teeth, until finally, feathers and then mammalian hair and eccrine glands evolved. We elaborate on the specific roles of the ectoderma

A self-cleaning photocatalytic membrane loaded with Bi2O2CO3/In(OH)3 S-scheme heterojunction composites for removing tetracycline from aqueous solutions.

Bi2O2CO3/In(OH)3 (BON) photocatalysts were synthesized by a one-pot method and loaded onto polyvinylidene fluoride (PVDF) membranes to obtain a Bi2O2CO3/In(OH)3/PVDF (BON-M) catalytic membrane system. The catalytic membranes demonstrated complete degradation of tetracycline within 40 min under visible light. They demonstrated robust photocatalytic activity across a broad pH range (5-11) and in the presence of coexisting ions. The membranes demonstrated excellent self-cleaning performance. Following exposure to light, the irreversible contamination decreased from 27.1% to 4.7% and the membrane'

Tetanus and botulinum toxins inhibit, and black widow spider venom stimulates the release of methionine-enkephalin-like material in vitro.

The actions of tetanus toxin, botulinum A toxin, and black widow spider venom on the release of methionine-enkephalin-like immunoreactivity have been studied; a particulate fraction prepared from rat striata was used. Depending on the duration of preincubation, tetanus toxin diminished the release evoked by veratridine (50 microM final concentration), and abolished it at final concentrations between 0.1 and 1 micrograms/ml. Botulinum A toxin was about 10 to 20 times less potent. Heating or pretreatment with antitoxin inactivated the clostridial toxins. The particulate fraction pretreated with

Batrachotoxinin-A 20-alpha-benzoate: a new radioactive ligand for voltage sensitive sodium channels.

Batrachotoxinin-A 20-alpha-benzoate (BTX-B), an analog of the potent depolarizing agent batrachotoxin (BTX), was prepared by selective esterification of naturally occurring batrachotoxin-A with benzoic acid. BTX-B depolarized rat phrenic nerve-diaphragm preparations with a time course and concentration dependence virtually indistinguishable from that of BTX. A specific, saturable component of equilibrium binding of [3H]BTX-B to mouse cerebral cortex homogenates was measured, described by an equilibrium dissociation constant of 0.7 microM and a maximum number of binding sites of 90 pmol per gra

Increase of cGMP and accumulation of 45Ca2+ evoked by drugs acting on sodium or potassium channels.

Cyclic guanosine monophosphate (cGMP) and 45Ca2+ accumulation were measured in mouse cerebellar slices after treatment with compounds known to affect ion channels in excitable membranes. Scorpion toxin and sea anemone toxin II raise cGMP and 45Ca2+ contents. Both toxins are known to keep the activated sodium channel open. 4-Aminopyridine and tetraethylammonium, which block potassium conductance, also increase cGMP and promote dose-dependently the 45Ca2+ accumulation. The effects of these 4 drugs but not the effects of depolarizing K+-concentrations on the accumulation of cGMP and 45Ca2+ are in

Simulation Studies and Dynamic Interaction of Venom Peptides with Ion Channels.

BACKGROUND: Arthropods such as scorpion, snake, insects, and spider as well as the marine animals like sea anemone and cone snails are venomous animals producing venoms with a complex mixture of peptide, poly peptides and small proteins. The disulfide rich peptides isolated from these animals are potent substances which specifically and selectively modulate different ion channels. The significant characteristics of these distinctive pharmacologically potent compounds highlights the molecular details of their peptide-ion channels interactions as well as provides the opportunities for the develo

Anaphylaxis: a history with emphasis on food allergy.

In the century since Paul Portier and Charles Richet described their landmark findings of severe fatal reactions in dogs re-exposed to venom after vaccination with sea anemone venom, treatment for anaphylaxis continues to evolve. The incidence of anaphylaxis continues to be difficult to measure. Underreporting due to patients not seeking medical care as well as failure to identify anaphylaxis affects our understanding of the magnitude of the disease. Treatment with intramuscular epinephrine continues to be the recommended first-line therapy, although studies indicate that education of both the

Lack of effect of a neurotoxin from the scorpion Buthus martensi Karsch on nerve fibers of this scorpion.

A neurotoxin (BmK I) was purified from the venom of the scorpion species Buthus martensi Karsch. Effects of this toxin on the excitability of the abdominal nerve fibers of the same scorpion were examined. The toxin had no effect at all on the action and resting potentials recorded intracellularly even at a concentration as high as 100 microM. A similar result was obtained through optical measurements of the action potential using a potential sensitive dye. Sea anemone toxin II (8 microM) had no effect on nerve excitability either. However, tetrodotoxin (50 nM) reversibly suppressed the action

Binding of scorpion toxin to receptor sites associated with sodium channels in frog muscle. Correlation of voltage-dependent binding with activation.

Purified scorpion toxin (Leiurus quinquestriatus) slows inactivation of sodium channels in frog muscle at concentrations in the range of 17-170 nM. Mono[125I]iodo scorpion toxin binds to a single class of sites in frog sartorius muscle with a dissociation constant of 14 nM and a binding capacity of 13 fmol/mg wet weight. Specific binding is inhibited more than 90% by 3 microM sea anemone toxin II and by depolarization with 165 mM K+. Half-maximal inhibition of binding is observed on depolarization to -41 mV. The voltage dependence of scorpion toxin binding is correlated with the voltage depend

Enhancing the therapeutic potential of peptide toxins.

Peptide toxins are potent and often exquisitely selective probes of the structure and function of ion channels and receptors, and as such are of significant interest to the pharmaceutical and biotech industries as both therapeutic leads and pharmacological tools. Their progression as clinical candidates, however, faces many of the challenges that are common to peptide drugs generally. Areas covered: The attributes of peptide toxins as therapeutic leads are outlined, as well as some of the limiting factors that have hampered the clinical development of many promising candidates. [Land and marine fauna constituting a threat for recreational divers in the tropics].

Due to intensively growing international tourism, increasing numbers of people leave for countries with hot climates, where various threats for human health and life exist. Besides climatic and sanitary conditions, a rich fauna, represented by predators and venomous animals, can be included. Based on available world literature and their own observations, the authors present the threats that a tourist can possibly encounter whilst relaxing on the beach or during recreational diving in tropical waters. When staying in water, a large threat is posed by marine fish of prey (sharks, barracuda, mura

Venom evolution through gene duplications.

Venoms contain highly complex mixtures that typically include hundreds of different components and have evolved independently in a diverse range of animals including platypuses, shrews, snakes, lizards, fishes, echinoderms, spiders, wasps, centipedes, sea snails, cephalopods, jellyfish and sea anemones. Many venom genes evolved through gene duplication. Gene duplication occurs in all domains of life and provides the raw substrate from which novel function arise. In this review, we focus on the role that gene duplication has played in the origin and diversification of venom genes. We outline th
