Rabbits (Oryctolagus cuniculus) increase caecal calcium absorption at increasing dietary calcium levels.

Hindgut fermenting herbivores from different vertebrate taxa, including tortoises, and among mammals some afrotheria, perissodactyla incl. equids, several rodents as well as lagomorphs absorb more calcium (Ca) from the digesta than they require, and excrete the surplus via urine. Both proximate and ultimate causes are elusive. It was suggested that this mechanism might ensure phosphorus availability for the hindgut microbiome by removing potentially complex-building Ca from the digesta. Here we use Ussing chamber experiments to show that rabbits (Oryctolagus cuniculus) maintained on four diffe…

Mitochondrial calcium uniporter complex activation protects against calcium alternans in atrial myocytes.

Cardiac alternans, defined as beat-to-beat alternations in action potential duration, cytosolic Ca transient (CaT) amplitude, and cardiac contraction is associated with atrial fibrillation (AF) and sudden cardiac death. At the cellular level, cardiac alternans is linked to abnormal intracellular calcium handling during excitation-contraction coupling. We investigated how pharmacological activation or inhibition of cytosolic Ca sequestration via mitochondrial Ca uptake and mitochondrial Ca retention affects the occurrence of pacing-induced CaT alternans in isolated rabbit atrial myocytes. Cytos…

Tetrabenazine Facilitates Exocytosis by Enhancing Calcium-Induced Calcium Release through Ryanodine Receptors.

Vesicular monoamine transporter-2 is expressed in the presynaptic secretory vesicles membrane in the brain. Its blockade by tetrabenazine (T … te delayed. Facilitation of exocytosis was not due to Ca2+ entry blockade through voltage-activated calcium channels (VACCs) because, in fact, TBZ mildly blocked the whole-cell Ca2+ current. However, TBZ mimicked the facilitatory effects of exocytosis elicited by BayK8644 (L-subtype VACC agonist), an effect blocked by nifedipine (VACC antagonist). On the …

[Cognitive Function and Calcium. Cognitive improvement through T type calcium channel stimulation].

Low-threshold Ca2+ spikes are mediated by T-type Ca2+ channels, which have fast inactivation and slow deactivation kinetics (transient) , and single channel conductance. The activation are triggered by -60 to -65 mV. T-type Ca2+ channels are predominantly expressed in the brain and heart pacemaker cells. Three subtypes of T-type Ca2+ channels Cav3.1 (α1G), Cav3.2 (α1H), Cav3.3 (α1I) encoding by CACNA1G, CACNA1H, CACNA1I genes have been cloned. Although high-threshold voltage-gated Ca2+ channels have auxiliary α2δ, β, γ subunits, T-type Ca2+ channels are composed only by α1 subunit. Although T-

Individualization of dialysate calcium concentration according to baseline pre-dialysis serum calcium.

BACKGROUND: A positive calcium balance may contribute to vascular calcification, while a negative balance increases iPTH. We explored the impact of different dialysate calcium concentrations on bone and mineral metabolism parameters according to pre-dialysis serum calcium levels. RESULTS: Fifty-six hemodialysis patients were dialyzed with 3.0 or 2.5 mEq/l dialysate [calcium] in a crossover study of two weeks. Bone mineral metabolites were measured prior to and following the hemodialysis session. A 3.0 mEq/l dialysate [calcium] increased more post-dialysis total calcium and ionized calcium tha…

Aluminum hydroxide, calcium carbonate and calcium acetate in chronic intermittent hemodialysis patients.

BACKGROUND AND METHODS: Prevention of secondary hyperparathyroidism in uremia necessitates correction of hyperphosphatemia and hypocalcemia. In order to avoid aluminum toxicity, calcium containing phosphate binders are used increasingly, instead of aluminium hydroxide. Recent studies have shown that calcium acetate has many characteristics of an ideal phosphate binder. It is, for instance, a more readily soluble salt compared with calcium carbonate. This advantage might, however, disappear if calcium carbonate is taken on an empty stomach, a few minutes before meals. We examined the efficacy o…

Calcium acetate versus calcium carbonate as phosphate-binding agents in chronic haemodialysis.

Hyperphosphataemia plays a key role in the pathogenesis of renal osteodystrophy, and phosphate-binding agents are required in many chronic dialysis patients. Aluminium hydroxide and calcium carbonate are well-established phosphate binders, but their use is associated with toxicity or poor efficacy. Calcium acetate is known to be a potent phosphate binder, and has recently been used successfully in chronic dialysis patients. In this randomized cross-over trial in 31 chronic haemodialysis patients, equimolar doses of calcium acetate and calcium carbonate were administered for 6 weeks each. Compl…

Calcium regulating hormones in healthy elderly men: relation to intestinal calcium absorption.

Aged male subjects often have osteoporosis and it has been suggested that the cause is an age-related lack of vitamin D. We evaluated the calcium regulating endocrine mechanism in healthy aged males in order to verify this hypothesis. We studied serum levels of PTH and 1,25(OH)2D in relation to intestinal calcium absorption, radial bone mass density (BMD) and osteocalcin plasma levels in 30 healthy elderly men (61-88 yr.). 1,25(OH)2D levels and calcium absorption, assessed by oral strontium test, did not change with age with respect to a young control group. PTH was higher (p < 0.02) in the el…

Calcium-independent GABA release from striatal slices: the role of calcium channels.

We have investigated the role of Ca2+ and Ca2+ channels in the modulation of GABA release. Brain slices prepared from rat striatum were preincubated with [3H]GABA, superfused with Krebs bicarbonate buffer, and exposed to electrical field stimulation (2 Hz for 3 min). Tritium efflux was measured as an index of GABA release. Both resting and evoked efflux were greatly accelerated by deleting Ca2+ from the medium and adding EGTA (1 mM). However, when the concentration of Mg2+ in the buffer was elevated to 10 mM, no effect of the Ca2(+)-deficiency was observed on resting release and its impact on

Calcium uptake and inositol trisphosphate-induced calcium release from parathyroid gland membranes.

The manner in which changes in Ca2+ are translated by the parathyroid gland into a signal for secretion of its major secretory proteins, parathormone and secretory protein-I, is not understood. In order to gain insight into this mechanism, Ca2+ transport was evaluated in porcine parathyroid gland membranes prepared by homogenization and differential centrifugation. The membranes were considered to be intact and appropriately 'sided' since they contained secretory protein-I and exhibited ATP-dependent uptake of [45Ca2+]. About 1 mol of ATP was hydrolyzed per mol of Ca2+ transported, signifying

Calcium chelation and calcium-channel blockade in anesthetized acute renovascular hypertensive dogs.

Acute animal studies were conducted to explore the effect of calcium manipulation on blood pressure and in the blood pressure response to angiotensin blockade under conditions of renovascular hypertension in anesthetized mongrel dogs. Ethylenediaminetetraacetic acid (EDTA) was used to lower serum calcium (SCa) levels, while nifedipine was infused intravenously to antagonize calcium at the cellular level. Angiotensin blockade was accomplished by infusion of the competitive inhibitor of angiotensin, saralasin. Following renal artery clamping, mean arterial pressure (MAP) increased a mean of 21 +…

Distinct calcium-independent and calcium-dependent adhesion systems of chicken embryo cells.

Three criteria have been used to distinguish among different systems of embryonic cell adhesion: dependence on Ca2+, involvement of particular cell-surface molecules, and binding specificity. The characterization of the adhesion with respect to cell-surface molecules was carried out by using specific antibodies against the neural and liver cell adhesion molecules (N-CAM and L-CAM) and antibodies raised against retinal cells prepared by limited trypsinization in the presence of Ca2+ (called "T/Ca cells"). Aggregation of cells prepared from retina or brain without Ca2+ did not require Ca2+ and w

Interactions between a "calcium channel agonist", Bay K 8644, and calcium antagonists differentiate calcium antagonist subgroups in K+-depolarized smooth muscle.

The proposal that calcium antagonists have different sites of action has been tested by attempting to reverse their inhibitory effects with a dihydropyridine which augments Ca2+ entry into cells, Bay K 8644. Bay K 8644 (1-1000 nmol/l) increased the sensitivity to Ca2+ of K+-depolarized taenia preparations from the guinea-pig caecum. Thus Bay K 8644 augmented established submaximal Ca2+-induced contractions and also shifted cumulative concentration-response curves to Ca2+ to the left, even in the presence of an optimal K+-depolarization. The inhibitory effects of nifedipine (10 nmol/l), verapam…

Calcium balance in dialysis is best managed by adjusting dialysate calcium guided by kinetic modeling of the interrelationship between calcium intake, dose of vitamin D analogues and the dialysate calcium concentration.

Calcium mass balance (Ca(MB)) is determined by the difference between Ca absorbed between dialyses (Ca(Abs)) and the Ca removed during dialysis (J(d)Ca(2+)). A mathematical model to quantify (1) Ca(Abs) as a function of Ca intake (Ca(INT)) and the doses of vitamin D analogues, and (2) J(d)Ca(2+) as a function of Ca(2+) dialysance, the mean plasma Ca(2+) ((M)C(pi)Ca(2+)) minus dialysate Ca(2+) (C(di)Ca(2+)), ultrafiltration rate (Q(f)) and treatment time is developed in this paper. The model revealed a basic design flaw in clinical studies of Ca-based as opposed to non-Ca-based binders in that …

Urine calcium and serum ionized calcium, total calcium and parathyroid hormone concentrations in the diagnosis of primary hyperparathyroidism and familial benign hypercalcaemia.

A relationship between serum concentrations of ionized calcium and parathyroid hormone (PTH) in persons without parathyroid overactivity was defined by performing an oral calcium load test. As a result, a serum PTH concentration greater than 2.6 pmol/L in a hypercalcaemic patient was regarded as suggestive of hyperparathyroidism. Fasting serum PTH concentrations in 58 patients with surgically and histologically proven primary hyperparathyroidism (PHPT) were all above 2.7 pmol/L (range 3.2-84.5). Thirteen of 20 patients with familial benign hypercalcaemia (FBH) had fasting serum PTH concentrati…

[Disorders of calcium metabolism].

The majority of clinical complaints derive from disorders of calcium metabolism and are associated with a wide variety of clinical symptoms caused by numerous diseases with entirely different types of pathophysiology. The prognosis varies from favorable to fatal depending on the pathophysiology of the underlying disorder of calcium metabolism; therefore, the diagnostic work-up aims to quickly identify the underlying disease causing the disturbance in calcium homeostasis. Every clinical situation with a diminished state of calcium absorption is treated with calcium and vitamin D in varying dose…

Calcium-sensing in the kidney.

PURPOSE OF REVIEW: Changes in extracellular calcium concentration affect several functions of the renal tubule. The calcium-sensing receptor (CaSR), initially identified in the parathyroid gland cells, is also expressed in the kidney and was assumed to mediate all effects of extracellular calcium on the renal tubule. The purpose of this review is to critically review the evidence supporting this assumption. RECENT FINDINGS: Recent results confirm that, in the kidney, the CaSR is mainly expressed in the thick ascending limb of the loop of Henle. There, it is involved in the control of calcium …
