Mechanisms of hormone-mediated carcinogenesis of the ovary.

Experimental ovarian carcinogenesis has been investigated in inbred and hybrid strains of mice and induced by a diversity of mechanisms including X-irradiation, oocytotoxic xenobiotic chemicals, ovarian grafting to ectopic or orthotopic sites, neonatal thymectomy, mutant genes reducing germ cell populations, and aging. Disruptions in the function of graafian follicles by a variety of mechanisms results in a spectrum of ovarian proliferative lesions, including tumors. The findings in mutant and genetically engineered mice support the concept of a secondary (hormonally mediated) mechanism of ova…

[Genetics and environmental factors in gastric carcinogenesis].

BACKGROUND: Gastric cancer is considered to be the second most common cancer worldwide. Carcinogenesis of the stomach is a multi-stage process. The progression from normal epithelial to tumor cells may involve at least five stages: superficial gastritis, chronic atrophic gastritis, intestinal metaplasia, dysplasia and carcinoma. These sequential changes in the gastric mucosa may occur over a period of many years as a result of exposure to a variety of exogenous and/or endogenous factors which cause genetic alterations. Recent developments in molecular genetics have shown that the accumulation …

Molecular and cellular mechanisms of cadmium carcinogenesis.

Cadmium is a heavy metal, which is widely used in industry, affecting human health through occupational and environmental exposure. In mammals, it exerts multiple toxic effects and has been classified as a human carcinogen by the International Agency for Research on Cancer. Cadmium affects cell proliferation, differentiation, apoptosis and other cellular activities. Cd2+ does not catalyze Fenton-type reactions because it does not accept or donate electrons under physiological conditions, and it is only weakly genotoxic. Hence, indirect mechanisms are implicated in the carcinogenicity of cadmiu

In vivo cell kinetics in breast carcinogenesis.

BACKGROUND: Disruption of the balance between apoptosis and proliferation is considered to be an important factor in the development and progression of tumours. In the present study we determined the in vivo cell kinetics along the spectrum of apparently normal epithelium, hyperplasia, preinvasive lesions and invasive carcinoma, in breast tissues affected by fibrocystic changes in which preinvasive and/or invasive lesions developed, as a model of breast carcinogenesis. MATERIALS AND METHODS: A total of 32 areas of apparently normal epithelium and 135 ductal proliferative and neoplastic lesion…

Mechanistic analysis and chemoprevention of pancreatic carcinogenesis.

Thirteen presentations given at a workshop held in November 1997, under the sponsorship of The Princess Takamatsu Cancer Research Fund in Nara, Japan, are summarized. Topics include studies of initiation, promotion, and chemoprevention in animal models of pancreatic carcinogenesis, molecular and histopathologic studies of human carcinomas, and epidemiologic studies. A recurring focus was to explore ways that knowledge of molecular mechanisms of carcinogenesis and molecular abnormalities in carcinomas can guide approaches for prevention and treatment of pancreatic cancer.

Protective role of lactulose in intestinal carcinogenesis.

Primary bile acids are converted to carcinogenic secondary bile acids by colonic bacteria when the colonic pH is high. Therefore acidification of the luminal contents may reduce the cancer risk. The ability of lactulose to reduce colonic pH, secondary bile acid production, mucosa crypt cell production rate and tumour formation was measured in a rat model of intestinal carcinogenesis. Eighty Wistar rats were divided into four groups receiving normal diet alone or with lactulose, azoxymethane or both azoxymethane and lactulose. The addition of lactulose was associated with a significant fall in …

The role of dietary phytosterols in colon carcinogenesis.

Epidemiological and experimental studies have shown that increased intake of plant foods and decreased meat consumption are correlated with a decreased risk for colon cancer. Many components of plant foods are suggested to mitigate colon carcinogenesis, including vitamins, minerals, and dietary fiber. Phytosterols are a common component of plant foods consumed in relatively large quantities by vegetarians, who are at lower risk for colon cancer development than individuals on a Western diet low in phytosterols. In addition, phytosterols have been shown experimentally to inhibit colon cancer de…

Role of proto-oncogene activation in carcinogenesis.

The accumulation of genetic damage in the forms of activated proto-oncogenes and inactivated tumor-suppressor genes is the driving force in the evolution of a normal cell to a malignant cell. For example, both the activation of ras oncogenes and the inactivation of several suppressor genes, including p53, have been observed in the development of human colon and lung tumors. Point mutations in key codons can activate ras proto-oncogenes and inactivate the p53 suppressor gene. Thus, several critical genes for tumorigenesis are potential targets for carcinogens and radiation that can induce point

Animal models for the study of prostate carcinogenesis.

Human prostate carcinogenesis has been viewed as a multi-step process involving progression from low histologic grade, small, latent carcinoma to large, higher grade, metastasizing carcinoma. However, recent data suggest that a variety of pathogenetic pathways may exist. The precise etiology and pathogenesis of human prostate cancer remain largely undefined. It is difficult to investigate stages in the development of human prostate cancer, but some animal models provide opportunities in this regard. Short-term treatment of rats with chemical carcinogens produces a low incidence (5-15%) of pros…

Folate intake and carcinogenesis of the colon and rectum.

Experimental evidence suggests that folate depletion plays a role in carcinogenesis. A case-control study examining folate intake was conducted. Some 428 colon and 372 rectal cancer cases with matched neighbourhood controls were interviewed regarding usual intake of foods, including food preparation. Unadjusted folate was not associated with risk of either cancer. Controlling for kilocalories, odds ratios (ORs) for those with the highest folate intake were 0.5 (95% confidence interval (CI): 0.24-1.03) and 0.31 (95% CI: 0.16-0.59) for females and males for rectal cancer. There was no change in …

Endometrial carcinogenesis and its relation to oestrogens.

The epidemiological evidence suggesting an association between the administration of exogenous oestrogens and an increased risk of developing endometrial adenocarcinoma is critically reviewed and it is concluded that the case for oestrogens being a cause of endometrial cancer is proven. The benefits flowing from preventing postmenopausal osteoporosis by oestrogen replacement therapy are assessed and contrasted with the danger to life posed by endometrial adenocarcinoma: it is pointed out that the type of neoplasm developing in patients taking oestrogens is well differentiated, nonaggressive an

Chromosome aberrations and their relevance to metal carcinogenesis.

Scoring for structural chromosome abnormalities is one of the only practical methods available for detecting visual damage in human genetic material. Cytogenetic tests in vivo and in vitro have shown the clastogenic potential of a number of metals and metal compounds. The difficulties in in vivo studies lie in identifying a specific clastogen in an occupational setting, where simultaneous exposure to a number of organic and inorganic chemicals is a common phenomenon. Metals known to be carcinogens in animals also tend to possess chromosome-damaging properties, even though more extensive studie

Perinatal viral carcinogenesis: the role of immunity.

The perinatal period cumulates conditions favorable to carcinogenesis that comprise high cellular multiplication rates, unstable homeostasis, and immune incompetence. The placenta allows the passage of various antigens including oncogenic viruses that have immunogenic or tolerogenic effects on the fetus. According to the structural type in different species, the placenta also transfers a diversity of maternal antibodies that protect efficiently the unborn progeny. However, in the postnatal period, the infant deprived of maternal antibodies and still incompetent to produce sufficient levels of …

[Mechanisms of carcinogenesis associated with DNA repair].

The paper presents data from the literature and the author's own materials on the correlation between the extent of activity of reparation processes and the development of cell malignization. A review cites the results of investigations on the molecular mechanisms in some hereditary human diseases in which a defect in some stages of the reparation processes has been found (xeroderma, ataxia telangesthesia. Fanconi anemia, Down's syndrome, etc.). These diseases are also characterized by a high rate of neoplasia development. It is emphasized that inhibition of the reparation process is observed
