Coal fires, industrial emissions and motor vehicles as sources of environmental carcinogens.

One of the most widely studied carcinogenic agents in the environment is the polycyclic hydrocarbon, benzo(a) pyrene. As a component of soot from the inefficient combustion of coal, its association with cancer can be traced back 200 years, but its possible relevance to lung cancer as a widely distributed air relevance to lung cancer as a widely distributed air pollutant has been investigated only during the past 25 years. Domestic coal fires have been shown to be important sources, and smaller amounts come from industrial sources and from motor vehicles. There is evidence now that the concentr

Foodborne Mycotoxins as Molecular and Epigenetic Carcinogens: Biochemical Mechanisms and Toxicological Implications.

Mycotoxins are toxic secondary metabolites produced predominantly by fungal genera, such as Aspergillus, Fusarium, and Penicillium, and represent major foodborne contaminants responsible for chronic human exposure worldwide. While aflatoxin B1 (AFB1) is a well-established hepatocarcinogen, increasing evidence indicates that multiple mycotoxins contribute to tumorigenesis across diverse organ systems through shared biochemical and molecular mechanisms. At the molecular level, mycotoxins undergo cytochrome P450-mediated bioactivation, generating reactive intermediates that induce DNA adduct form

Variations in exposure to carcinogens among shift workers and non-shift workers.

BACKGROUND: There is limited information on whether the prevalence of exposure to workplace carcinogens varies among shift workers and non-shift workers. METHODS: This analysis used data from the Australian Work Exposures Study-Cancer, a telephone survey which examined exposure to carcinogens in the workplace. Workers were classified as shift workers if they indicated that their usual roster ever included work between the hours of midnight and 5 am. Modified Poisson regression was used to estimate the adjusted prevalence ratios (aPRs) and 95% confidence internals (CIs). RESULTS: Among the 54…

Lunasin-aspirin combination against NIH/3T3 cells transformation induced by chemical carcinogens.

Carcinogenesis is a multistage process involving a number of molecular pathways sensitive to intervention. Chemoprevention is defined as the … rdic acid on cell proliferation and foci formation of transformed NIH/3T3 cells induced by chemical carcinogens 7,12-dimethylbenz[a]anthracene or 3-methylcholanthrene. The results revealed that lunasin, acting as a single agent, inhibits cell proliferation and foci formation. When combined with aspirin, these effects were significantly increased, indicating t …

Exposure to chemical carcinogens and risk of cancer among Finnish laboratory workers.

BACKGROUND: Laboratory workers have long been suspected of having increased risks of cancer due to their occupation. We evaluated occupational exposure and cancer incidence among Finnish laboratory workers. MATERIALS AND METHODS: The cohort was comprised of 4,722 laboratory workers reported to the Finnish Register of Workers Exposed to Carcinogens in 1979-1988. The standardized incidence ratios (SIR) for cancers and their 95% confidence intervals (CI) were calculated based on data of the Finnish Cancer Registry. RESULTS: The most common carcinogens, to which these workers were potentially ex…

Ethanol interactions with other cytochrome P450 substrates including drugs, xenobiotics, and carcinogens.

Chronic ethanol abuse is associated with increased activity of the microsomal ethanol-oxidizing system. This effect is due primarily to induction by ethanol of a specific cytochrome P450 (CYP2E1) responsible for enhanced oxidation of ethanol and other P450 substrates and, consequently, for metabolic tolerance to these substances. Furthermore, cytochrome 450 induction increases the activation of numerous xenobiotics to toxic metabolites and of chemical carcinogens to reactive metabolites, thereby accelerating their adverse effects. Microsomal enzyme induction has been associated with increased …

Antigen presentation in the skin: modulation by u.v. radiation and chemical carcinogens.

The skin provides an excellent model to investigate antigen presenting cells (APC) particularly using contact hypersensitivity responses. The skin has its own distinct APC of which Langerhans cells (LC) are the best characterized. Fluorescein isothiocyanate (FITC) has proved a useful reagent with which to follow antigen-labeled APC to draining lymph nodes, where they bind to T lymphocytes. Cluster formation appears to be a necessary component of the APC-T cell interaction. Both u.v. radiation and chemical carcinogens are able to alter LC in the skin, resulting in immunological tolerance when a…

Localized hepatocarcinogenesis: the response of the liver and kidney to implanted carcinogens.

Attempts by early workers to induce liver tumours by the local implantation of carcinogens had by and large not been successful, so that the liver came to be viewed as being "resistant" to tumourigenesis by this means. A review of these early studies showed not only that fibrosarcomas could be easily induced by the local application of 3-methylcholanthrene (3-M.C.), but that there were also reasons why the apparently low susceptibility of the liver to the localised induction of hepatocellular tumours should not be accepted as established dogma. In an attempt to re-investigate this problem pell…

Metabolism of tobacco specific carcinogens in cultured rat buccal mucosa epithelial cells.

The metabolism of tobacco carcinogens was studied in a potential target for their carcinogenic effects. Rat buccal mucosa cells metabolized several polycyclic aromatic hydrocarbons, e.g., benzo(a)pyrene, benz(b)- and benz(j)-fluoranthene, dibenz(a,j)acridine, N,N-diethylnitrosamine and protein pyrolysate products, 2-amino-3-methylimidazo(4,5-f)quinoline, and 2-amino-3,4-dimethylimidazo(4,5-f)quinoline, as measured by binding of the carcinogens to cellular DNA. The highest level of binding was seen with the nitrosamine followed by the protein pyrolysate products. There was no significant differ…

Hyperthermia and other factors increasing sensitivity of Euglena to mutagens and carcinogens.

The effects of different factors (temperature, light, enzymic activation) on the ability of selected mutagens and carcinogens to induce hereditary bleaching of Euglena gracilis were investigated. In the resting medium, the elevation of incubation temperature from 25 degrees C to 37 degrees C increased significantly the effect of all compounds tested on the frequency of bleached mutants of E. gracilis. The effect of light is not so unambiguous. While nitrosoguanidine (NG) exhibited practically the same bleaching activity both in the light and dark, the mutagenic effect of sodium azide (SA), nit…

[Comparison of the effect of different carcinogens on the skin of rats].

Solution of N-methyl-N-nitrosourea (MNU), diazoacetic ester (DAAE), N-methyl-N-nitro-N-nitrosoguanidine (MNNG), N-methyl-N,N'-dinitroguanidine, 4-dimethylaminoazobenzene (DAB), o-aminoazotoluene or 7,12-dimethylbenz(a)anthracene (DMBA) were applied to rat skin. Neoplasms (skin tumors) were found only in the experiments with MNU, DAAE, MNNG, DAB and DMBA, MNU, MNNG, DAB and DMBA induced tumours in the sites of skin painting. In experiments with DAAE distant skin neoplasms developed as well. Possible mechanisms of action of these compounds are discussed.

Changes in tumor-specific nuclear antigen activity in carcinogen-treated colon by tumor promoter and carcinogen inhibitors.

Changes in the immunospecificity of the nuclear antigens were demonstrated in the colon chromatin of rats treated with 1,2-dimethylhydrazine. Tumor-specific nuclear antigen appeared in the early stages of chemically induced colon carcinogenesis. Sodium barbiturate, in conjunction with the carcinogen, induced a higher level of nuclear antigen activity than that obtained with carcinogen alone. The rise of immunoactivity in carcinogen-treated colon chromatin can be abolished by simultaneous treatment with disulfiram or butylated hydroxytoluene.

Carcinogenic, non-carcinogenic risk, and attributable cases to organochlorine pesticide exposure in women from Northern Mexico.

This study aimed to estimate the carcinogenic and non-carcinogenic risk as well as the attributable cases due to exposure to organochlorine pesticides (OCPs): hexachlorobenzene (HCB), dichlorophenyltrichloroethane (DDT), hexachlorocyclohexane (HCH), heptachlor, and chlordane. From serum concentrations of pesticides of interest in a sample of 908 women from Northern Mexico, the risk for both cancer and non-cancer health effects was evaluated. The population attributable fraction (PAF) was also calculated based on summary association estimates between exposure to OCPs and different health events

The WHO claims estrogens are 'carcinogenic': is this true?

Estrogens are in the list of carcinogenic chemicals from the World Health Organization (WHO). However, estrogens require additional factors such as stromal factors or progestogens to increase the ratio of proliferation/apoptosis for initiation of replication errors and consequent mutations to occur. These mutations require at least 5-10 years to develop into clinically detectable cancer, whereby this review is focused on breast cancer. The US National Cancer Institute highlighted a second mechanism of carcinogenicity: certain estrogen metabolites are capable of inducing DNA damage, even in low

Carcinogenicity Evaluation of Baclofen in TgrasH2 Mice.

Baclofen is a γ-aminobutyric acid-B receptor agonist used for control of spastic muscle activity and as a treatment for alcohol abuse. The review of the nonclinical database suggested a data gap for potential carcinogenicity following long-term use. Regulatory requirements for pharmaceutical safety testing of cancer-causing potential have historically included 2-year rodent studies in rats and mice. The availability of transgenic models with greater specificity and sensitivity to carcinogens provides safety testing alternatives that align with the 3Rs. The carcinogenicity of baclofen was evalu…

Biocompatibility and Carcinogenicity of Carbon Nanotubes as Biomaterials.

With the development of nanotechnology in recent years, there have been concerns about the health effects of nanoparticles. Carbon nanotubes (CNTs) are fibrous nanoparticles with a micro-sized length and nano-sized diameter, which exhibit excellent physical properties and are widely studied for their potential application in medicine. However, asbestos has been historically shown to cause pleural malignant mesothelioma and lung cancer by inhalation exposure. Because carbon nanotubes are also fibrous nanotubes, some have raised concerns about its possible carcinogenicity. We have reported that

Carcinogenicity of chromium and chemoprevention: a brief update.

Chromium has two main valence states: hexavalent chromium (Cr[VI]) and trivalent chromium (Cr[III]). Cr(VI), a well-established human carcinogen, can enter cells by way of a sulfate/phosphate anion-transport system, and then be reduced to lower-valence intermediates consisting of pentavalent chromium (Cr[V]), tetravalent chromium (Cr[IV]) or Cr(III) via cellular reductants. These intermediates may directly or indirectly result in DNA damage or DNA-protein cross-links. Although Cr(III) complexes cannot pass easily through cell membranes, they have the ability to accumulate around cells to induc
