INTRODUCTION: Localized renal cell carcinoma is conventionally treated surgically. Preoperative and adjuvant external radiation have not improved survival. Total lymphoid irradiation (TLI) is a highly effective modality for inducing immunosuppression and transplantation tolerance. The cellular basis for this immunosuppression is not clear, although T cells have been implicated. Four local anaesthetics (LAs), amethocaine and procaine (esters) and cinchocaine and lignocaine (amides) have been assessed in terms of their ability to induce turbidity increases in non-growing cells of Escherichia coli suspended in a carbohydrate free defined medium (CFM) or distilled water or in 0.2 M phosphate buffer. We present a methodology integrating spatial dosimetry with colony formation assessment and modelling to improve prediction of SFRT-induced responses.
