The role of VEGF in vascular dementia: impact of aging and cellular senescence.

Vascular Endothelial Growth Factor (VEGF) is a critical element in vascular dementia (VD) pathogenesis and therapeutic development while rem … and oxidative stress to generate adverse vascular symptoms that include atherosclerosis and stroke. Cellular senescence convolutes these processes by causing damaging inflammatory reactions alongside impaired vascular healing abilities. The secretion of pro-inflammatory cytokines from senescent cells (SCs) disrupts VEGF signaling and produces harmful consequences for …

Cellular senescence and its pathogenic and therapeutic implications in autoimmune hepatitis.

INTRODUCTION: Senescent cells are characterized by replicative arrest and phenotypes that produce diverse pro-inflammatory and pro-oxidant mediators. The senescence of diverse hepatic cell types could constitute an unrecognized pathogenic mechanism and prognostic determinant in autoimmune hepatitis. The impact of cellular senescence in autoimmune hepatitis is unknown, and it may suggest adjunctive management strategies. AREAS COVERED: This review describes the molecular mechanisms of cellular senescence, indicates its diagnostic features, suggests its consequences, presents possible therapeut…

Biomarkers of cellular senescence and major health outcomes in older adults.

The geroscience hypothesis proposes that underlying biological processes, such as the accumulation of senescent cells, have deleterious effects on multiple tissues and increase the risk of many chronic conditions with aging. Senescent cells produce heterogenous biomarkers, also called senescence-associated secretory phenotype (SASP). Circulating concentrations of senescence biomarkers may reflect an underlying burden of senescent cells in various tissues. Plasma levels of these proteins have been associated with increased mortality and poorer physical function. The associations of them with th

Effect of peripheral cellular senescence on brain aging and cognitive decline.

We examine similar and differential effects of two senolytic treatments, ABT-263 and dasatinib + quercetin (D + Q), in preserving cognition, markers of peripheral senescence, and markers of brain aging thought to underlie cognitive decline. Male F344 rats were treated from 12 to 18 months of age with D + Q, ABT-263, or vehicle, and were compared to young (6 months). Both senolytic treatments rescued memory, preserved the blood-brain barrier (BBB) integrity, and prevented the age-related decline in hippocampal N-methyl-D-aspartate receptor (NMDAR) function associated with impaired cognition. Cellular senescence in the cholangiopathies: a driver of immunopathology and a novel therapeutic target.

The cholangiopathies are a group of liver diseases that affect cholangiocytes, the epithelial cells that line the bile ducts. Biliary atresia (BA), primary biliary cholangitis (PBC), and primary sclerosing cholangitis (PSC) are three cholangiopathies with significant immune-mediated pathogenesis where chronic inflammation and fibrosis lead to obliteration of bile ducts and eventual liver cirrhosis. Cellular senescence is a state of cell cycle arrest in which cells become resistant to apoptosis and profusely secrete a bioactive secretome. Recent evidence indicates that cholangiocyte senescence …

Cellular senescence in the aging brain: A promising target for neurodegenerative diseases.

Aging is inevitable. Along with reduced ability to maintain the homeostasis of various biological processes, aging gradually deteriorates overall health. Extensive research on the aging brain has identified cellular senescence as a critical risk factor of neurodegeneration and cognitive decline. Associations of cellular senescence with neurodegenerative diseases like Alzheimer's disease, Down syndrome, Parkinson's disease, and multiple sclerosis are evident in an extensive body of literature generated over decades of research on aging. Cellular senescence triggers neurodegeneration via a compl…

Fracture Healing in the Setting of Endocrine Diseases, Aging, and Cellular Senescence.

More than 2.1 million age-related fractures occur in the United States annually, resulting in an immense socioeconomic burden. Importantly, the age-related deterioration of bone structure is associated with impaired bone healing. Fracture healing is a dynamic process which can be divided into four stages. While the initial hematoma generates an inflammatory environment in which mesenchymal stem cells and macrophages orchestrate the framework for repair, angiogenesis and cartilage formation mark the second healing period. In the central region, endochondral ossification favors soft callus devel

Cdkn1a transcript variant 2 is a marker of aging and cellular senescence.

Cellular senescence is a cell fate response characterized by a permanent cell cycle arrest driven primarily the by cell cycle inhibitor and tumor suppressor proteins p16Ink4a and p21Cip1/Waf1. In mice, the p21Cip1/Waf1 encoding locus, Cdkn1a, is known to generate two transcripts that produce identical proteins, but one of these transcript variants is poorly characterized. We show that the Cdkn1a transcript variant 2, but not the better-studied variant 1, is selectively elevated during natural aging across multiple mouse tissues. Importantly, mouse cells induced to senescence in culture by geno…

Cellular senescence in vivo: its relevance in ageing and cardiovascular disease.

In most somatic mammalian cell types extensive replication and various types of cellular insults induce a permanent form of growth arrest called senescence. Senescence has been comprehensively characterised in cell culture, but its occurrence in vivo has only recently started to become appreciated. In this mini-review, we examine the evidence for the occurrence of senescence in vivo, with particular emphasis on the cardiovascular system. We also describe the senescent phenotype and discuss its pathophysiological implications. We examine findings in animal models of ageing and human genetic dis

Neuroendocrine-associated epigenetic factors in cellular senescence: mechanisms and therapeutic implications.

Ageing is a progressive biological process causing a reduction in tissue and cellular function due to accumulated molecular damage over time. Cellular senescence is a stable cellular state characterized by irreversible cell-cycle arrest accompanied by distinct molecular, epigenetic, and secretory alterations. Moreover, neuroendocrine signalling and epigenetic regulation have had a great impact on controlling ageing and senescence. As a part of the neuroendocrine system, the hypothalamic-pituitary-adrenal (HPA) axis and insulin-like growth factor-1 (IGF-1) mediate stress responses, circadian re…

Cellular Senescence in Skin Wound Repair: Dual Roles and Senotherapeutic Interventions.

Skin wound repair is a complex process involving numerous signalling pathways in various cell types. Cellular senescence (CS), described as a proliferative limit, has dual implications in this process. In acute wounds, transient cellular senescence acts as a beneficial mechanism that facilitates tissue repair through the controlled secretion of the senescence-associated secretory phenotype (SASP), promoting immune cell recruitment, myofibroblast differentiation and angiogenesis. In contrast, chronic wound settings, which are frequently associated with pathological conditions including type 2 d…

Orbital hybridization-mediated nanozymes reverse cellular senescence for aged bone regeneration.

The regeneration of aged bone is severely compromised by a deteriorating microenvironment characterized by excessive reactive oxygen species (ROS) and consequential endoplasmic reticulum stress (ERS). Herein, we report a biomimetic nanozyme engineered through synergistic p-d orbital hybridization and youthful membrane camouflage to precisely reverse this degenerative cascade. The designed Cu-Sn dual-atom core exhibits exceptional multi-enzymatic activity, scavenging superoxide anions with a 2.5-fold higher efficiency than Cu single-atom control. Mechanistic studies confirm that p-d hybridizati

High-Salt Diet-Induced Endothelial Dysfunction Is Mediated by Cellular Senescence.

BACKGROUND: High salt (HS) intake is a known cardiovascular risk factor, yet the mechanisms linking salt intake to endothelial dysfunction remain unclear. We investigated whether HS induces vascular senescence and dysfunction, and whether targeting senescent cells could prevent these effects. METHODS: Male C57BL/6J mice were fed a high-salt diet (HSD [8% NaCl]) for 14 or 28 days. Vascular function was assessed in thoracic aortae and mesenteric arteries using wire myography. Senescence was evaluated by p21, p16, and interleukins (IL) 6 and 1β expression in endothelial cells and whole vessels.

Disordered differentiation and cellular senescence in pediatric Hodgkin Reed-Sternberg cells.

Hodgkin lymphoma (HL) is characterized by inflammatory lesions with relatively few (typically <1%) pathogenic Hodgkin Reed-Sternberg (HRS) cells. The scarcity of malignant HRS cells in HL has historically limited characterization of these disease drivers, and pathogenic mechanisms underlying HL are incompletely defined. Using multiparameter flow cytometry to purify HRS cells and immune cell populations from pediatric HL lesions and control tonsils, we sought to define transcriptional programs in pediatric HRS cells. HRS cells are thought to arise from transformed germinal center B cells. Inter

Cellular sensor DAP5 decodes Betacoronaviral NSP5 to drive virus-induced senescence.

BACKGROUND: Viral infection induces host cells to enter a state of "virus-induced senescence (VIS)", which provides a stable cellular enviro … eavage of DAP5 that led to the production of the N-terminal fragment DAP51-451, which initiated the cellular senescence program, achieving an "apoptosis→senescence" fate transformation and thereby promoting viral replication of SARS-CoV-2. Cellular senescence in adult pulmonary hypertension: current state and future challenges.

Pulmonary hypertension (PH) is a life-threatening disease increasingly being diagnosed in the elderly population, marked by vascular injury, excessive vasoconstriction and progressive remodelling of the pulmonary arteries (PAs). These lead to sustained elevation of PA pressure and subsequent development of right ventricular failure. Despite the beneficial effects on disease progression and quality of life, current treatments do not cure PH, highlighting the need for a deeper understanding of the underlying mechanisms. Recently, cellular senescence has gained much attention as a stress response…

Cellular senescence links severe Leptospira infection to chronic kidney disease progression.

AIMS: Leptospirosis, a re-emerging zoonotic disease caused by pathogenic Leptospira species, poses an increasing public health risk, particularly in the context of climate change. Severe leptospirosis has been associated with a heightened risk of chronic kidney disease (CKD), suggesting potential long-term renal consequences. However, the underlying mechanisms remain unclear. This study investigates the impact of severe acute Leptospira infection on kidney function, focusing on cellular senescence and its role in disease progression. MATERIALS AND METHODS: A hamster model of severe leptospiro…

FGF receptors mediate cellular senescence in the cystic fibrosis airway epithelium.

The number of adults living with cystic fibrosis (CF) has already increased significantly because of drastic improvements in life expectancy attributable to advances in treatment, including the development of highly effective modulator therapy. Chronic airway inflammation in CF contributes to morbidity and mortality, and aging processes like inflammaging and cell senescence influence CF pathology. Our results show that single-cell RNA sequencing data, human primary bronchial epithelial cells from non-CF and CF donors, a CF bronchial epithelial cell line, and Cftr-knockout (Cftr-/-) rats all de

Belinostat (PXD101) resists UVB irradiation-induced cellular senescence and skin photoaging.

Belinostat (PXD101), a new histone deacetylase inhibitor, has shown good performance in various cancer treatments and has been approved by the FDA for the treatment of recurrent or refractory peripheral T-cell lymphoma (PTCL) in patients with drugs. PXD101 is considered to have certain anti-allergic and anti-inflammatory properties, but its beneficial effects in UVB-induced skin photoaging have not been reported. In a recent study, HacaT cells and C57BL6 mice were used to study the impact of PXD101 on UVB-induced cellular senescence and skin photoaging and to explore their potential mechanisms…
