This article describes the development of a simple technique by which the numbers of surface immunoglobulin expressing cells can be analysed by dual fluorescence flow cytometry in samples of whole blood. We have compared the results obtained using this procedure with those obtained using samples prepared by traditional density gradient centrifugation, and demonstrate an excellent correlation between the two techniques. Lipid rafts are dynamic assemblies of glycosphingolipids, sphingomyelin, cholesterol, and proteins that can be stabilized into platforms involved in the regulation of various vital cellular processes. Here, we describe small-scale isolation methods of lipid rafts. Detergent-resistant membrane raft fraction is isolated by a volume of 0.8 mL sucrose density gradient centrifugation from blood platelets. Raft fraction is also isolated from cerebellar neurons by co-immunoprecipitation using anti-glycosphingolipid monoclonal antibodies.

Red blood cells from normal subjects and subjects with heterocellular hereditary persistence of fatal haemoglobin and beta-thalassaemia were fractionated according to density by centrifugation on a discontinuous gradient of Stractan II. F-cells were studied by immunofluorescence and their proportion was evaluated in each separated population. This approach has permitted to show that F-cells were preferentially distributed among high density erythrocytes. This phenomenon reflects a peculiar characteristic of F-cells.
