Endobody: Genetically Encodable Nanobody-CPP Chimeras for Degradation of Membrane and Extracellular Proteins.

Membrane protein degraders (MPDs) like LYTACs are emerging tools for targeting disease-relevant membrane proteins, but their reliance on specific endocytic receptors limits their applicability across diverse cell types. Here, we present a structurally concise and genetically encodable class of degraders, nanobody-cell-penetrating peptide (CPP) chimeras, termed endobodies. We demonstrated that a genetically fused CPP is sufficient to mediate the internalization and subsequent degradation of membrane or extracellular proteins recognized by the nanobody. Using this platform, we achieved targeted …

Design and functional characterization of recombinant chimeras as vaccine candidates against Lawsonia intracellularis.

Proliferative enteropathy is a gastrointestinal disease caused by the intracellular bacterium Lawsonia intracellularis, affecting swine populations worldwide and leading to significant economic losses due to reduced growth performance, increased mortality, and treatment expenses. Current vaccines present limitations related to production processes, stability, and overall cost. This study aimed to design and evaluat chimeric proteins containing immunogenic epitopes of L. intracellularis. A recombinant chimera, designated Li_full, was rationally designed in silico by integrating predicted B-cell…

Caged Glycan-Antibody Conjugates for Tumor-Selective Activation of Lysosome-Targeting Chimeras.

Lysosome-targeting chimeras (LYTACs) induce lysosomal degradation of extracellular and membrane proteins by bridging target proteins with lysosomal trafficking receptors. However, conventional LYTACs often suffer from off-tissue effects, whereas reported tissue-specific LYTACs typically display limited degradation efficiency. To address these challenges, we report a chemically activatable LYTAC platform that leverages tumor microenvironment-specific cues to achieve precise and safe protein degradation in vivo. We designed a glutathione (GSH)-responsive caged mannose-6-phosphate glycan, GSH-pM6…

An inducible germ cell ablation chicken model for high-grade germline chimeras.

Chicken embryos are a powerful and widely used animal model in developmental biology studies. Since the development of CRISPR technology, gene-edited chickens have been generated by transferring primordial germ cells (PGCs) into recipients after genetic modifications. However, low inheritance caused by competition between host germ cells and the transferred cells is a common complication and greatly reduces production efficiency. Here, we generated a gene-edited chicken, in which germ cells can be ablated in a drug-dependent manner, as recipients for gene-edited PGC transfer. We used the nitro

Assessing species-specific contributions to craniofacial development using quail-duck chimeras.

The generation of chimeric embryos is a widespread and powerful approach to study cell fates, tissue interactions, and species-specific contributions to the histological and morphological development of vertebrate embryos. In particular, the use of chimeric embryos has established the importance of neural crest in directing the species-specific morphology of the craniofacial complex. The method described herein utilizes two avian species, duck and quail, with remarkably different craniofacial morphology. This method greatly facilitates the investigation of molecular and cellular regulation of

Imaging the cell entry of the anthrax oedema and lethal toxins with fluorescent protein chimeras.

To investigate the cell entry and intracellular trafficking of anthrax oedema factor (EF) and lethal factor (LF), they were C-terminally fused to the enhanced green fluorescent protein (EGFP) and monomeric Cherry (mCherry) fluorescent proteins. Both chimeras bound to the surface of BHK cells treated with protective antigen (PA) in a patchy mode. Binding was followed by rapid internalization, and the two anthrax factors were found to traffic along the same endocytic route and with identical kinetics, indicating that their intracellular path is essentially dictated by PA. Colocalization studies …

[Residual neurovirulence of the chimera of Langat and Denge-4 flaviviruses in intracerebral infection of monkeys].

The residual neurovirulence of the previously obtained chimeric virus Tp21/DEN4, containing the RNA region encoding the pre M and E structural enzymes, strain Tp21, virus Langat, and the remaining part of the genome from the Denge 4 virus, was studied in experiments with monkeys Cercopithecus aethiops involving the intracerebral administration of the virus. The tick-borne encephalitis virus, strain Absettarov, was used as positive control. A comparative analysis of the experimental and published data showed the chimeric virus to be less virulent by its degree of morphological affection in the

The origin of the subepicardial mesenchyme in the avian embryo: an immunohistochemical and quail-chick chimera study.

It has been proposed that the subepicardial mesenchymal cells (SEMC) originate from the primitive epicardium and also from migration of extracardiac mesenchyme from the liver area. We have studied the possibility of an origin of SEMC through transformation of the proepicardial mesothelium, as well as the potential of the early proepicardium to generate epicardium and SEMC in quail-chick chimeras. The study was carried out in quail and chick embryos between HH16 and HH29 stages. Most proepicardial cells, mesothelial as well as mesenchymal, were cytokeratin and vimentin immunoreactive, suggestin…

Antiviral immune responses of fully allogeneic irradiation bone marrow mouse chimeras.

In (B10.BR----B10) chimeras infected with lymphocytic choriomeningitis (LCM) virus higher titers were attained in spleens and livers than in organs of the mice used for their construction, and the subsequent elimination was retarded, but eventually the virus was cleared. The numbers of LCM virus-specific CTL and their precursors as quantitated with chromium-release assay and limiting dilution method, respectively, were lower in chimeras than in B10.BR or C57BL/10J mice, and fewer were restricted for the haplotypes of the donors than of the recipients. The same was true with regard to antiviral…

Mouse chimeras composed of trisomy 16 and normal (2N) cells: preliminary studies.

Many humans with trisomy 21 (Down Syndrome (DS)) have psychomotor and cognitive retardation, congenital heart disease, and hematologic abnormalities. Partial genetic homology exists between mouse chromosome 16 and human chromosome 21; thus, studies of the development of mice with trisomy 16 (Ts16) may provide important insights into the pathogenesis of these defects and into the mechanisms by which they arise in humans. Since Ts16 mice do not survive the late fetal period, chimeras have been formed between Ts16 and normal (2N) mouse embryos to rescue the Ts16 cells for postnatal studies. In th…

Rewiring Oncogenic Transcriptional Complexes with Domain-ALTeration Chimeras (DALTACs) in Prostate Cancer.

Transcriptional addiction to the androgen receptor (AR) underlies metastatic castration-resistant prostate cancer (mCRPC), where AR maintains oncogenic enhancer programs through dynamic, domain-specific interactions with the lysine acetyltransferases p300/CBP and associated cofactors. Here, we describe a mechanistically distinct therapeutic modality, Domain-ALTeration Chimeras (DALTACs), designed to rewire endogenous protein complexes by enforcing non-native domain-domain interactions rather than degrading or inhibiting individual components. Our first-in-class molecule, AR-p300/CBP DALTAC-1, …

Polymeric lysosome-targeting chimeras for extracellular α-synuclein degradation in Parkinson's disease.

Disease progression in Parkinson's disease has been driven by extracellular α-synuclein prion-like seeding throughout the course of the disease and therefore not just by the intracellular accumulation of the protein in isolated aggregates. Current therapies utilizing PROTACs cannot address the extra-cellular effects of α-synuclein spreading in this manner. This article proposes PolyTACs (Polymeric Lysosome-Targeting Chimeras) as hybrid antibody-polymer conjugates which use neuronal exofacial thiol groups produced because of DJ-1/GSH dysregulation to capture α-synuclein pathological conformers …

AAV delivered lysosome-targeting chimeras mediate sustained antibody depletion in vivo.

Immunoglobulin G (IgG) is a critical effector of the adaptive immune system, but IgG autoantibodies against self-antigens drive pathology across a wide range of autoimmune diseases. Lowering circulating IgG is a validated and promising therapeutic avenue. Here we developed genetically-encoded lysosome targeting chimeras (GELYTACs) that target circulating IgGs for clearance and degradation. The GELYTACs comprised two protein modules derived from insulin-like growth factor 2 (IGF2) and an IgG-binding nanobody, respectively, and mediated clearance of plasma IgG via the lysosomal trafficking recep…

Multimodal supramolecular targeting chimeras enable spatiotemporally resolved protein degradation in vivo.

Targeted protein degradation (TPD) has transformed strategies for modulating protein function in both basic biology and therapeutic development. However, current strategies often lack the spatial and temporal precision required for in vivo applications. Herein, we report supramolecular targeting chimeras (SupTACs), a modular and programmable platform that enables tissue-specific and temporally controlled protein degradation in vivo. SupTACs self-assemble into supramolecular nanoparticles (SNPs) that co-localize target-binding ligands and E3 ligase recruiters, thereby facilitating proteasomal d…

Enediyne Chimeras for Targeted NAMPT Degradation: Enhancing Cytotoxicity via a Radical Clock.

Based on a novel targeted protein degradation platform, Protein-Radical-Oxidation Targeting Enediyne Chimera (PROTEC), a series of chimeric enediyne compounds targeting the oncogenic protein nicotinamide phosphoribosyltransferase (NAMPT) were designed and synthesized. These compounds feature different linker lengths and cyclopropane modifications at the alkyne terminus. Computational studies showed that incorporating the cyclopropane group at the propargyl position facilitates the "radical clock" mechanism, achieving greater spatial separation and weaker orbital interactions between the two ra…

Inhibition of equilibrative nucleoside transporters 1 and 2 by proteolysis-targeting chimeras (PROTACs).

Proteolysis-targeting chimeras (PROTACs) are gathering considerable interest due to their ability to address previously undruggable targets. We were keen to understand the potential for these very large molecules to interact with transporters that may influence absorption, distribution, metabolism, and excretion or toxicity properties and to what extent this may be predictable using machine learning models. Consequently, we tested a set of PROTACs against several human drug transporters, namely the equilibrative nucleoside (ENT) family transporters ENT1 and ENT2, which have been directly impli…
