Effects on dentin treated with eluted multi-mineral varnish in vitro.

PURPOSE: The purpose of this pilot study was to assess the non-contact effects of a NaF varnish on the morphology and elemental compositions of sound and demineralized dentin in an in vitro pH cycling model. METHODS: 3 mm diameter dentin cores were extracted from bovine teeth, mounted in acrylic rods, and ground and polished. Sound and demineralized (in 10 mL unstirred 50% citric acid (pH = 1.2) for two minutes) specimens were divided into four groups (N=6) corresponding to treatments with either water (control group) or non-contact Vanish® 5% NaF White Varnish with tri-calcium phosphate (3M

Metabolism of branched-chain amino acids and ammonia during exercise: clues from McArdle's disease.

Patients with McArdle's disease (myophosphorylase deficiency) cannot use muscle glycogen as an energy source during exercise. They therefore … of BCAA drains 2-oxoglutarate in the primary aminotransferase reaction and thus reduces flux in the citric acid cycle and impedes aerobic oxidation of glucose and fatty acids. This draining effect is normally counteracted by the anaplerotic conversion of muscle glycogen to citric acid cycle intermediates, a reaction which is severely hampered in these patients due …

3-Nitropropionic acid animal model and Huntington's disease.

Huntington's disease (HD) is a progressive neurodegenerative disorder associated with severe degeneration of basal ganglia neurons, especial … ted that systemic administration of 3-nitropropionic acid (3-NP), an inhibitor of the mitochondrial citric acid cycle, results in a progressive locomotor deterioration resembling that of HD. Furthermore, we observed congruent with other reports, that 3-NP produces a very selective striatal degeneration. It differs mechanistically from excitotoxic lesions in that 3- …

Growth and synthesis of rubratoxin by Penicillium rubrum in a chemically defined medium fortified with organic acids and intermediates of the tricarboxylic acid cycle.

A sterile glucose-mineral salts broth was fortified with equimolar concentrations (10--3 M) of various organic acids and intermediates in the tricarboxylic acid cycle. Appropriate media were neutralized with 2 N NaOH, inoculated with spore suspensions or mycelial pellets of Penicillium rubrum and incubated quiescently for 14 days or with shaking for 5 days. Rubratoxins were recovered from culture filtrates by ether extraction and resolved by thin-layer chromatography. Toxin formation in quiescent cultures was enhanced by malonate but was not markedly affected by ethyl malonate, shikimate, and

Multi-component analysis of tetracyclines, sulfonamides and tylosin in swine manure by liquid chromatography-tandem mass spectrometry.

A multi-component method focussing on thorough sample preparation has been developed for simultaneous analysis of swine manure for three classes of antibiotic-tetracyclines, sulfonamides, and tylosin. Liquid manure was initially freeze-dried and homogenised by pulverization before extraction by pressurised liquid extraction. The extraction was performed at 75 degrees C and 2,500 psig in three steps using two cycles with 0.2 mol L(-1) citric acid buffer (pH 4.7) and one cycle with a mixture of 80% methanol with 0.2 mol L(-1) citric acid (pH 3). After liquid-liquid extraction with heptane to rem

In vitro and in vivo enhancement of colistin activity against Escherichia coli strains by macelignan.

UNLABELLED: Colistin remains a last-line antibiotic against multidrug-resistant Gram-negative pathogens, but its clinical utility is limited … Mechanistic studies revealed that macelignan disrupts bacterial energy metabolism by inhibiting the citric acid cycle, leading to ATP depletion. This energy compromise impairs the bacterial capacity to repair colistin-induced membrane damage, exacerbating membrane disruption and accelerating bacterial death. These findings establish macelignan as a promising adjuva …

Frequency of Application of AmF/NaF/SnCl2 Solution and Its Potential in Inhibiting the Progression of Erosion in Human Dental Enamel - An In Vitro Study.

PURPOSE: To evaluate whether increasing the frequency of its use can enhance the protective effect of AmF/NaF/SnCl2 solution against dental erosion. MATERIALS AND METHODS: Sixty human enamel samples were obtained from sound human third molars, and after the formation of incipient erosive lesions (1% citric acid, pH 4.0, for 3 min), they were divided into five treatment groups (n = 12): G1 - deionised water (negative control); G2 - NaF solution (positive control) once a day; G3 - NaF solution (positive control) twice a day; G4 - AmF/NaF/SnCl2 solution once a day; G5 - AmF/NaF/SnCl2 solution tw

Metabolic profiling and biomarker identification in idiopathic intracranial hypertension with venous sinus stenosis.

Idiopathic intracranial hypertension (IIH) with venous sinus stenosis (VSS) may lead to a vicious cycle of elevated intracranial pressure (ICP) and subsequent brain damage. This study aimed to characterize metabolic alterations and identify potential biomarkers in IIH with VSS compared to individuals with normalized ICP after stent treatment. Patients with trans-stenotic pressure gradient (TSPG) ⩾8 mmHg were defined as the new-onset IIH group (NIG), while those with TSPG <8 mmHg after stenting formed the post-stent follow-up group (PFG). After propensity score matching, 19 participants were in

Global serum metabolomics profiling of colorectal cancer.

Accurate diagnosis of colorectal cancer (CRC) relies on the use of invasive tools such as colonoscopy and sigmoidoscopy. Non-invasive tools … protein biosynthesis, urea cycle, ammonia recycling, alanine metabolism, glutathione metabolism and citric acid cycle. The metabolomics analysis revealed levels of metabolites of glycolysis, tricarboxylic acid cycle, anaerobic respiration, protein, lipid and glutathione metabolism were significantly different between cancer and control samples. Although the majorit …

Loss of TCA cycle turning promotes stem cell function.

The citric acid cycle (TCA cycle) is the common terminal pathway for the oxidation of all nutrients. Citrate oxidation to oxaloacetate produces CO 2 , and citrate synthase (CS) uses nutrient-derived acetyl groups to regenerate citrate and fuel cycle turning. However, the essentiality of cycle fueling and turning in vivo remains unclear. Here, we use hematopoiesis, the most proliferative system in the body, as a model to show that, contrary to common assumptions, TCA cycle turning is dispensable for respiration, survival, and proliferation of stem and progenitor cells in vivo and its loss promo…

Crosstalk between the tricarboxylic acid cycle and peptidoglycan synthesis in Caulobacter crescentus through the homeostatic control of α-ketoglutarate.

To achieve robust replication, bacteria must integrate cellular metabolism and cell wall growth. While these two processes have been well characterized, the nature and extent of cross-regulation between them is not well understood. Here, using classical genetics, CRISPRi, metabolomics, transcriptomics and chemical complementation approaches, we show that a loss of the master regulator Hfq in Caulobacter crescentus alters central metabolism and results in cell shape defects in a nutrient-dependent manner. We demonstrate that the cell morphology phenotype in the hfq deletion mutant is attributab

Metabolic Dysregulation in Parkinson's Disease: Non-Oxidative Phosphorylation and Its Role in Brain Energy Metabolism.

Parkinson's disease (PD) is a progressive neurodegenerative condition affecting around 1-2% of the population over the age of 60. The lack of disease-modifying therapies highlights the need for insights into the etiology and pathogenesis of PD. Mitochondrial dysfunction is recognized to be a significant contributor to disease pathogenesis, resulting in bioenergetic deficits and subsequent neurodegeneration. Research indicates that changes in non-oxidative phosphorylation (non-OXPHOS) metabolism in PD may serve as an adaptive response to mitochondrial dysfunction, compensating for energetic fai

Diagnosis of prostate cancer by desorption electrospray ionization mass spectrometric imaging of small metabolites and lipids.

Accurate identification of prostate cancer in frozen sections at the time of surgery can be challenging, limiting the surgeon's ability to best determine resection margins during prostatectomy. We performed desorption electrospray ionization mass spectrometry imaging (DESI-MSI) on 54 banked human cancerous and normal prostate tissue specimens to investigate the spatial distribution of a wide variety of small metabolites, carbohydrates, and lipids. In contrast to several previous studies, our method included Krebs cycle intermediates (m/z <200), which we found to be highly informative in distin

[Changes in the levels of glycolysis and tricarboxylic acid cycle metabolites in cat tissues during periodontosis].

The predominance of restored substrates in the bone system and liver of cats under chronic parodontosis evidence for intensification of reproduction properties of NAD-pairs and causes inhibition of glycolysis and activation of gluconeogenesis. A high content of alpha-ketoglutarate and isocitrate in tissues in a result of lipogenesis inhibition. Under the lower rate of glycolysis, activation of proteolysis and transamination reactions amino acids are the major contributors to pyruvate. Under the acute course of the disease with the development of the inflammatory process the quantity of pyruvat

Studies on the energetic metabolism of Shigella flexneri x Escherichia coli hybrids devoid of penetration ability. IV. Growth characteristics in the presence of some tricarboxylic acid cycle intermediates.

The dynamics of avirulant lac+KCP- hybrids Sh. flexneri multiplication was investigated in minimal medium in the presence of some tricarboxylic acid cycle intermediates (fumarate, succinate, malate) and pyruvate. It was established that in spite of the loss of virulence in the rate of the multiplication of the hybrid strains, in comparison with that of the virulent recipient strains of Sh. flexneri, considerable differences were not observed.

Heightened innate immunity may trigger chronic inflammation, fatigue and post-exertional malaise in ME/CFS.

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is characterized by unexplained fatigue, post-exertional malaise (PEM), and cognitive dysfunction. ME/CFS patients often report a prodrome consistent with infection. We present a multi-omics analysis based on plasma metabolomic and proteomic profiling, and immune responses to microbial stimulation, before and after exercise. We report evidence of an exaggerated innate immune response after exposure to microbial antigens; impaired energy production involving the citric acid cycle, beta-oxidation of fatty acids, and urea cycle energy pr…

Escherichia coli O157:H7 sublethally injured by freezing stress: Physiological characteristics and mechanisms underlying its formation and resuscitation.

Freezing stress can induce sublethal injury in foodborne pathogens, allowing injured cells to evade detection and subsequently resuscitate, thereby posing a hidden food safety risk. This study investigated the formation, physiological characteristics, and resuscitation of freezing-induced sublethally injured Escherichia coli O157:H7 at -20 °C and explored the underlying mechanisms using untargeted metabolomics. After 21 h of freezing, the proportion of sublethally injured cells exceeded 80%, accompanied by cell surface collapse, impaired membrane integrity, and depleted intracellular protein a
