The present study aimed to develop Dot-ELISA, complement fixation test (CFT) and capillary tube agglutination test (CAT) for serodiagnosis of Babesia equi infection and to compare their sensitivity with each other. For this study, sequential serum samples were collected from four donkeys experimentally infected with B. equi up to 90 days post infection (P.I.). B. equi antigen was prepared from the blood of a donkey showing more than 80% parasitaemia. Dot-ELISA, CF and CA tests were standardized as per the standard method. While performing CFT, it was observed that CFT standardized for the donk

In this study, 241 serum samples from individuals exposed to brucellosis were subjected to the Rose Bengal plate test (RBPT); the titre was estimated by standard tube agglutination test (STAT), with positive > or = 80 IU/ml. Randomly selected sera (n = 81) were analysed by complement fixation test (CFT): titre > or = 1:4 was considered positive. Of 241 sera subjected to RBPT and STAT, 177 were negative in both tests; 5 samples tested negative by RBPT but positive by STAT. None was positive by RBPT and negative by STAT. Of 81 sera subjected to CFT, 23 (28.4%) were positive. Both RBPT and CFT fo

The results of complement fixation (CF) test for equine piroplasmosis on sera from horses destined for international movement from Great Britain and Ireland are presented and analysed. No horses born and continuously resident in the British Isles were found carrying CF antibodies to either Babesia equi or B caballi. Positive animals were found to have association with the following countries where known tick vectors occur: Spain, Portugal, Belgium, France, Poland, USSR and Arabian Gulf countries. Data on the persistence of CF antibodies in animals subjected to repeated testing showed that some

Three techniques for the serological diagnosis of Rocky Mountain spotted fever were compared by testing 417 sera from 178 patients who very probably did not have rickettsial infections and 88 sera from 41 patients who very probably had Rocky Mountain spotted fever (SF). The techniques were complement fixation (CF), indirect fluorescent antibody (IFA), and microagglutination (MA). To avoid possible degradation during unnecessary purification, the antigens were prepared by methods that were as simple as possible. In the CF tests of 417 sera from patients with nonrickettsial diseases there was on

A total of 372 serum samples were collected from Colombian cattle before and during the course of natural Babesia spp. infection on the North Coast of Colombia. The serum samples were used to compare indirect fluorescent antibody (IFA) with complement fixation (CF) tests for diagnosis of babesiosis. The IFA technique detected Babesia argentina antibodies an average of 4.0 weeks earlier than the CF test and Babesia bigemina an average of 2.5 weeks earlier. Both IFA and CF were capable of differentiating B. argentina and B. bigemina infections, however in some cases cross reactions were observed

We evaluated the indirect fluorescent-antibody (IFA) test and complement-fixation (CF) test for diagnosis of equine piroplasmosis in the absence of a gold standard. Using Evan's blue, we estimated the specificity of the IFA test on a parasite-free, field horse population to be 98% (95% confidence interval=97, 99). We observed an excellent test agreement (kappa=0.83) between two collaborating laboratories when the IFA test was performed on identical samples from an endemic area. Using Bayesian analysis with informative prior probability distributions, we estimated the sensitivity of the IFA tes

An enzyme-linked immunosorbent assay (ELISA) was developed to detect immunoglobulin G to Coxiella burnetii phase II. Serum samples from 213 patients who had had Q fever 1 year previously and from 301 blood donors from six localities in Switzerland were tested by ELISA and by indirect fluorescent-antibody (IFA) and complement fixation (CF) tests. The ELISA and the IFA and CF tests detected antibody to C. burnetii in 202 (94.8%), 193 (90.6%), and 166 (77.8%) of the 213 Q fever patients, respectively. With the serum samples from blood donors, the ELISA yielded a higher percentage of positive sera

Anthrax is a zoonotic disease caused by Bacillus anthracis spore-forming bacterium. Since it is primarily a disease of animals, the control in animals, and humans depend on the prevention in livestock, principally cattle, sheep, and goats. Most veterinary vaccines utilize the toxigenic, uncapsulated (pXO1+/pXO2-) B. anthracis strain 34F2 which affords protection through the production of neutralizing antibodies directed to the toxin components Protective Antigen (PA), Lethal Factor (LF), and Edema Factor (EF). The titration of specific antibodies in sera of vaccinated animals is crucial to eva

The live attenuated Brucella abortus strain RB51 is a rifampin-resistant, lipopolysaccharide (LPS) O-chain-deficient mutant of virulent B. abortus 2308. The reduced O-chain content in RB51 prevents this bacterium from inducing antibodies detectable by the conventional serologic tests for bovine brucellosis diagnosis that mainly identify antibodies to LPS. The absence of available serologic tests for RB51 also complicates the diagnosis of possible RB51 infections in humans exposed to this strain. The purpose of this study was to evaluate the suitability of a complement fixation (CF) test perfor

A survey to detect individuals with antibodies to Lassa virus was undertaken among hospital personnel in the eastern and southern provinces of Sierra Leone late in 1974. Sera were evaluated by the complement fixation test. The data obtained showed that some contacts of Lassa fever patients in the 1972 epidemic had developed antibodies to the virus; individuals who had never reported being sick also showed evidence of infection, with significant CF antibody titres in their sera. Surviving Lassa fever patients from the 1972 epidemic still had easily measurable levels of persisting CF antibodies.
