Molecular characterization of Corynebacterium pseudotuberculosis isolates using ERIC-PCR.

Caseous lymphadenitis is an infectious sheep and goats disease caused by Corynebacterium pseudotuberculosis and characterized by abscesses in superficial and visceral lymph nodes. C. pseudotuberculosis strains isolated from these hosts have been shown to be very difficult to type by the existing methods. The aim of this study is evaluating the potential of the Enterobacterial Repetitive Intergenic Consensus (ERIC-PCR) as a tool for molecular typing of C. pseudotuberculosis strains isolated in sheep. One hundred and twenty seven isolates of C. pseudotuberculosis were isolated from lesions suspe…

Xylitol production by recombinant Corynebacterium glutamicum under oxygen deprivation.

Wild-type Corynebacterium glutamicum produced 0.6 g l(-1) xylitol from xylose at a productivity of 0.01 g l(-1) h(-1) under oxygen deprivation. To increase this productivity, the pentose transporter gene (araE) from C. glutamicum ATCC31831 was integrated into the C. glutamicum R chromosome. Consequent disruption of its lactate dehydrogenase gene (ldhA), and expression of single-site mutant xylose reductase from Candida tenuis (CtXR (K274R)) resulted in recombinant C. glutamicum strain CtXR4 that produced 26.5 g l(-1) xylitol at 3.1 g l(-1) h(-1). To eliminate possible formation of toxic intrac…

Inflammatory pseudotumor of the liver revealing gynecological Corynebacterium infection.

Inflammatory pseudotumor (IPL) of the liver is a rare benign clinicopathologic entity, which has been associated with infectious conditions, including alimentary, digestive tract and mouth infections. We report a new case, where a patient developed hepatic IPL that revealed endometritis related to Corynebacterium infection; the patient had a favorable outcome of hepatic IPL after appropriate antibiotic therapy initiation. Our findings therefore confirm that antibiotic therapy may be effective in patients with hepatic IPL associated with underlying infections, avoiding unnecessary surgery. Our …

[Acute Corynebacterium diphtheriae aortic endocarditis with negative blood cultures].

UNLABELLED: Acute endocarditis with negative blood culture is a challenge requiring close cooperation between several specialists. CASE REPORT: A 15-year-old boy presented with an acute aortic endocarditis due to Corynebacterium diphtheriae. Blood cultures were negative due to an empiric anti-infective therapy. Valvular replacement with a mechanical prosthesis failed to cure the sepsis, which resolved after a cryopreserved allograft implantation. CONCLUSION: This case underlines the efficiency of the highly specialized bacteriological centers and the advantages of using a cryopreserved allog…

Infection due to Corynebacterium species in marrow transplant patients.

A Corynebacterium species consistently resistant to all antibiotic therapy except vancomycin caused bacteremia in 32 of 284 (11%) marrow transplant patients. Twenty-one patients had colonization or infection before bacteremia. Twenty-six of the 32 patients were males, and males older than 16 years were infected significantly more often than females over 16, or than all patient under 16. A case-control study showed that infected patients had greater exposure to antibiotics; more often had failure of engraftment and persistent granulocytopenia; were in laminar air-flow rooms less often; and had …

Carriage of non-toxigenic Corynebacterium diphtheriae by Australian Aboriginals.

A survey of 200 Aboriginals in Western Australia revealed five carriers of non-toxogenic Corynebacterium diphtheriae; four of these were variety mitis and one was variety gravis.

Identification and characterization of an ene-reductase from Corynebacterium casei.

The asymmetric reduction of α, β-unsaturated compounds conjugated with electron-withdrawing group by ene-reductases (ERs) is a valuable method for the synthesis of enantiopure chiral compounds. This study introduced an ER from Corynebacterium casei (CcER) which was heterologously expressed in Escherichia coli BL21(DE3), and the purified recombinant CcER was characterized for its biocatalytic properties. CcER exhibited the highest specific activity at 40 °C and pH 6.5, and showcased appreciable stability below 40 °C over a pH range of 6.0-7.0. The enzyme displayed high resistance to methanol. C…

Unusual source of recurrent Corynebacterium bacteraemia in an immunocompromised patient.

We describe a unique case of a patient with acute myeloid leukaemia (AML), with recurring infections during chemotherapy from chronic nasal carriage of non-diphtherial Corynebacterium, who was eventually diagnosed as she presented with neutropaenic sepsis. Identifying (often multiple) sources of infection in immunocompromised patients is crucial but deciding whether multiple organisms, which in health are considered as commensals, are actually pathogenic during vulnerable states-can be clinically difficult. Our case highlights the efforts to correctly identify the actual source of this rare or…

Induction of glutamic acid production by copper in Corynebacterium glutamicum.

From the previous transcriptome analysis (Hirasawa et al. Biotechnol J 13:e1700612, 2018), it was found that expression of genes whose expression is regulated by stress-responsive transcriptional regulators was altered during penicillin-induced glutamic acid production in Corynebacterium glutamicum. Therefore, we investigated whether stress treatments, such as copper and iron addition, could induce glutamic acid production in C. glutamicum and found that the addition of copper did induce glutamic acid production in this species. Moreover, we also determined that glutamic acid production levels…

Corynebacterium freneyi as a cause of early prosthetic valve endocarditis.

Infective endocarditis (IE) is associated with severe complications and a high mortality rate. Identification of the causative pathogen is crucial to optimise treatment. We present a case of prosthetic valve endocarditis caused by Corynebacterium freneyi, a very rare cause of human infection and not previously reported as a cause of IE. Despite proper antibiotic therapy, the patient eventually needed surgery after progression of the infection. After surgery, he quickly recovered without evidence of relapse during an 8-month follow-up period. This report highlights critical decision making in a…

Corynebacterium striatum bacteremia associated with cellulitis in a patient with cirrhosis.

Corynebacteria are a bacteria usually associated as a contaminant due to their presence in normal human skin and mucosal membranes. However, they are increasingly becoming recognized as an opportunistic pathogen. Corynebacteria can be pathogens in immunocompromised patients or those with malignancies or prosthetic devices. We present a rare case of bacteremia due to multidrug resistant Corynebacterium striatum in a 52-year-old male with cirrhosis. The patient had lower extremity cellulitis which likely served as a port of entry.

Septic arthritis in a native knee due to Corynebacterium striatum.

We describe a case of septic arthritis in a native knee due to Corynebacterium striatum, gram-positive bacilli that are usually commensal organisms of skin and mucosal membranes, but are seldom implicated in native septic arthritis. An 84-year-old man with Corynebacterium striatum septic arthritis of his native left knee and no response to conventional antibiotic therapy. Thus, the patient was allowed to take dalbavancin for compassionate use, with an excellent clinical outcome. This case emphasizes de role of Corynebacterium striatum in native joint infections and highlights the importance of…

[A case of diphtheria in the Netherlands due to an infection with Corynebacterium ulcerans].

A 58-year-old woman was admitted due to a pseudomembranous pharyngitis. The patient had not been vaccinated against diphtheria. Corynebacterium ulcerans was cultured from a throat swab. The production of diphtheria toxin by these bacteria was demonstrated with PCR and an immunoprecipitation test. The patient was cared for in respiratory isolation and was treated with benzylpenicillin. She quickly recovered and was discharged four days after admission. A contact investigation did not reveal any dissemination of the toxin-producing C. ulcerans and a source was not found. In spite of the large-sc…
