A de novo microdeletion in a patient with inner ear abnormalities suggests that the 10q26.13 region contains the responsible gene.

Microdeletions in the 10q26.1 region are related to intellectual disability, growth delay, microcephaly, distinctive craniofacial features, cardiac defects, genital abnormalities and inner ear abnormalities. The genes responsible for inner ear abnormalities have been narrowed to fibroblast growth factor receptor 2 gene (FGFR2), H6 family homeobox 2 gene (HMX2) and H6 family homeobox 3 gene (HMX3). An additional patient with distinctive craniofacial features, congenital deafness and balance dysfunctions showed a de novo microdeletion of 10q26.11q26.13, indicating the existence of a gene respons

Sleep Disturbances in Pediatric Craniofacial Syndromes.

Sleep is essential for healthy growth and neurocognitive development in children, however, sleep disturbances in the pediatric population are still largely undiagnosed. Children with craniofacial abnormalities are at particular risk for developing sleep disturbances, which can have long-lasting neurodevelopmental, cardiovascular, and respiratory consequences. This article seeks to fill knowledge gaps in screening, evaluation, and management of common sleep disorders in children with craniofacial abnormalities. With early identification and treatment, clinicians can significantly improve the ov…

Research progress of proteomics in congenital craniofacial anomalies.

Congenital craniofacial anomalies (CFAs) are among the most common birth defects, significantly affecting the appearance, oral function and mental health of patients. These anomalies are etiologically complex, involving genetics, environmental factors and gene-environment interactions. While genetic studies have identified numerous potential causal genes/risk loci for CFAs, the pathogenic mechanisms still largely remain elusive. Proteomics, the large-scale analysis of proteins, offers a comprehensive view of disease pathogenesis and their systemic effects. During the past two decades, the appl

Tessier Number 3 and 4 Clefts: Clinical Presentation and Associated Clefts in a South African Population.

INTRODUCTION: The defects found in Tessier clefts number 3 and number 4 come in various forms in different patients. These variations have to a great extent affected not only documentation of these craniofacial defects but invariably their treatment and communication amongst craniofacial researchers. This study has not only documented the clinical presentation of these clefts in a South African population but has also incorporated the clinical presentation of Tessier clefts number 3 and 4 from different regions of the world. METHODS: The records of 8 patients, who had been treated for either

Psychological adjustment to craniofacial conditions (excluding oral clefts): A review of the literature.

OBJECTIVE: A congenital craniofacial anomaly (CFA) is expected to impact upon several domains of psychological, emotional and social functioning, yet no recent reviews have comprehensively summarised the available literature. Further, existing reviews tend to draw upon literature in the field of cleft lip and palate, and do not give substantive attention to other types of CFAs. DESIGN: A review of 41 papers published between January 2000 and March 2016 pertaining to psychological adjustment to CFAs. MAIN OUTCOME MEASURES: Findings are presented according to key psychological domains: General

Craniofacial syndromes.

LEARNING OBJECTIVES: After studying this article, the participant should be able to: 1. Recognize the clinical presentations of commonly seen craniofacial syndromes. 2. Understand the most serious complications associated with each syndrome. 3. Formulate the best age-appropriate surgical plans. SUMMARY: Craniofacial syndromes fall into two major categories-those associated with craniosynostosis, and those associated with clefts. Each has a different set of potential complications requiring a unique approach for surgical management. Craniosynostosis is a congenital disorder in which one or mor

Orthodontic and Craniofacial Skeletal Alterations in Individuals With Thalassemia: A Scoping Review.

BACKGROUND: Thalassemia is a well-recognized hereditary blood disorder with systemic complications. However, its craniofacial and orthodontic alterations remain incompletely characterized. Current evidence is fragmented, and no comprehensive synthesis has been published hitherto. AIMS: To map and synthesize craniofacial and orthodontic alterations in individuals with thalassemia by identifying morphological patterns and associated clinical factors. METHODS: This scoping review included cross-sectional studies, cohort studies, and clinical trials retrieved from four databases and gray literat

The society for craniofacial genetics and developmental biology 46th annual meeting.

The Society for Craniofacial Genetics and Developmental Biology (SCGDB) held its 46th Annual Meeting at Cincinnati Children's Hospital Medical Center in Cincinnati, Ohio on October 10th-12th, 2023. On the first day of the meeting, Drs. Sally Moody and Justin Cotney were each honored with the SCGDB Distinguished Scientist Awards for their exceptional contributions to the field of craniofacial biology. The following two days of the meeting featured five sessions that highlighted new discoveries in signaling and genomic mechanisms regulating craniofacial development, human genetics, translational

[Rational use of distraction osteogenesis in craniofacial surgery].

Distraction osteogenesis, initially developed by Ilizarov for limb, is the tissular extension caused by the progressive space of the osseous pieces following an osteotomy. Distraction is osteogenesic and histogenic. Twenty-five years ago, at the instigation of McCarthy, this technique was used to handle the craniofacial malformations in the various floors of the face : mandibular, mediofacial and cranial. The most wide-spread protocols respect a latency period from 0 to 7 days, a rhythm of distraction from 1 to 2mm a day in 2 at 4 times and a period of consolidation from 4 to 8 weeks. Distract

Macrocephaly syndromes.

Anatomic megalencephaly resulting in macrocephaly occurs in more than 100 multiple congenital anomaly syndromes. A number of macrocephaly syndromes show accompanying somatic overgrowth, but some show normal somatic growth. This discussion provides a review of several macrocephaly syndromes that might be encountered by the pediatric neurologist. Growth patterns, craniofacial features, congenital anomalies, central nervous system imaging findings, performance, and mode of inheritance for each condition are reviewed as well as recent developments in the molecular genetic testing for these conditi
