Coxiella burnetii induces reorganization of the actin cytoskeleton in human monocytes.

Coxiella burnetii, an obligate intracellular bacterium which survives in myeloid cells, causes Q fever in humans. We previously demonstrated that virulent C. burnetii organisms are poorly internalized by monocytes compared to avirulent variants. We hypothesized that a differential mobilization of the actin cytoskeleton may account for this distinct phagocytic behavior. Scanning electron microscopy demonstrated that virulent C. burnetii stimulated profound and polymorphic changes in the morphology of THP-1 monocytes, consisting of membrane protrusions and polarized projections. These changes we…

The role of the cytoskeleton in the polarized growth of the germ tube in Candida albicans.

Cells of the dimorphic yeast Candida albicans are easily induced to germinate in synchrony. Using germinating cells of strain FC18, we examined the effects of several drugs that are known to affect the cytoskeleton on growth and cytoskeletal organization. Cytochalasin A (CA), an inhibitor of actin function, inhibited the germination of the yeast cells and changed the cylindrical expansion of the apex of the germ tube to swelling growth. Effects of CA on the organization of actin were examined with rhodamine-phalloidin (Rh-Ph), which specifically stains F-actin. In CA-untreated cells, Rh-Ph sta…

Influence of Ca2+ depletion on cytoskeleton and nucleolus morphology in Trypanosoma brucei.

Trypanosoma brucei bloodstream forms were incubated in a calcium-free medium containing 10 microM ethyleneglycol-bis-(beta-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA). Under these conditions, addition of 5 microM calcium ionophore A23187 led to striking morphological alterations, as judged by light and electron microscopy. The cytoskeleton of trypanosomes consists of a subpellicular corset of microtubules. Characteristically four of these microtubules are attached invariantly to an extension of the endoplasmic reticulum at the flagellar attachment site. Specifically in this area calciu…

The liquid crystalline nature of the cytoskeleton in epidermal cells of the chaetognath Sagitta.

The chaetognaths have a multilayered epidermis, which is not covered by cuticle, except in the head region. Two kinds of cells are found in the epidermis: the filament-rich cells, adjacent to the basement membrane, and superficial cells, which are filament poor. The filament-rich cells, which are linked by gap junctions and columnar junctions, are highly developed in the collarette region, which joins the head and the trunk. As elsewhere in the epidermis these cells are covered by the filament poor cells which are linked by zonulae adhaerentes, gap junctions and septate junctions. The filament

Hormonally induced changes in the cytoskeleton of human thyroid cells in culture.

Serially cultivated thyroid follicular cells are not active in hormone synthesis but retain a thyrotropin-responsive adenylate cyclase. The exposure of such cells to thyrotropin leads to an increase in the concentration of intracellular cAMP and a drastic change in morphology including a total cytoplasmic arborization. The present communication describes these changes at the cytoskeletal level using a cell line derived from a human functioning thyroid adenoma. Phase contrast microscopy showed that the cytoplasmic arborization was preceded by a total disappearance of stress fibers, visible with

Mechanotherapy enhances nanodrug uptake by overcoming the actin cytoskeleton damping effect.

Efficient nanodrug therapy in solid tumors is limited not only by extracellular barriers but also by the mechanical state of the plasma membrane (PM). Although mechanotherapy remodels the extracellular matrix (ECM) and improves tissue-level delivery, its effect on transmembrane transport remains unclear. Here, we show that matrix stiffness is transmitted to the PM through the actomyosin cytoskeleton and thereby regulates nanoparticle (NP) uptake. Within a hepatocellular carcinoma-relevant stiffness range, matrix stiffening promoted excessive F-actin polymerization and stress-fiber formation, p…

Immunomodulatory cytoskeleton of bionic periosteum affects mitochondrial dynamics to promote osteogenesis.

PURPOSE: Periosteum is a key regulatory hub in the immune microenvironment of bone repair and the mitochondrial dynamics of bone marrow mesenchymal stem cells (BMSCs) directly determine the osteogenic efficiency; therefore, how to promote bone healing through the immunomodulation of mitochondrial dynamics is a key challenge. METHODS: Inspired by the natural periosteum, we designed a bionic periosteum that integrates topology and controlled Semaphorin 3 A (Sema3A) release. Hyaluronic acid-coated Sema3A granular microsols spontaneously formed a nucleus-sheath structure under a high-voltage elec

Polystyrene microplastics induce liver toxicity and cytoskeleton alterations in female rats.

Microplastics (MPs) are emerging environmental pollutants that have received increasing attention in recent years. However, data on their potential risks to mammalian species remain limited. In this study, female rats were exposed via oral gavage to pristine (PPS) and fluorescent polystyrene (FPS) particles (5 μm diameter) over four estrous cycles. FPS-MPs were detected in hepatic tissue (133 ± 35 particles), confirming their translocation. Exposure to PPS-MPs increased superoxide dismutase (SOD) (2.58-fold) and catalase (CAT) activity (1.36-fold), while reducing protein sulfhydryl levels (PSH

CD44-downregulation in multiple myeloma inhibits cytoskeleton rearrangement through actin depolymerization.

It is well known that multiple myeloma (MM) cells are highly dependent on the bone marrow (BM) microenvironment. However, the complex interactions and signaling pathways between MM cells and BM stromal cells remain inadequately defined. In this study, we employed an in vitro coculture model to investigate these interactions. We found that coculturing MM cells with BM-derived HS5 stromal cells stimulated the secretion of hyaluronic acid (HA) and interleukin-6 (IL-6), and significantly increased the expression of CD44 and F-actin stress fibers polymerization in MM cells. Among the three hyaluron

Emerging Arthropod-Borne Viruses Hijack the Host Cell Cytoskeleton During Neuroinvasion.

Arthropod-borne viral infections, ranging from asymptomatic to fatal diseases, are expanding from endemic to nonendemic areas. Climate change, deforestation, and globalization favor their spread. Although arboviral manifestations mainly determine the onset of generalized symptoms, distinct clinical signs have been assessed, depending on the particular arthropod-borne virus (arbovirus) involved in the infectious process. A number of arboviruses cause neuroinvasive diseases in vertebrate hosts, with acute to chronic outcomes. Long-term neurological sequelae can include cognitive dysfunction and

Reorganization of the mouse oocyte' cytoskeleton after cultivation under simulated weightlessness.

Female germ cells provide the structural basis for the development of a new organism, while the main molecular mechanisms of the impact of weightlessness on the cell remain unknown. The aim of this work was to determine the relative content and distribution of the main proteins of microtubules and microfilaments, to assess the relative RNA content of genes in mouse oocytes after short-term exposure to simulated microgravity, and to determine the potential for embryo development up to the 3-cell stage. Before starting the study, BALB/c mice were divided into two groups. One group received water

Cross-reacting material 197 (CRM197) affects actin cytoskeleton of endothelial cells.

CRM197, cross-reacting material 197, is a mutant of diphtheria toxin (DTx). CRM197 is used in pharmacology as a carrier protein. It has been recently shown that CRM197 causes breakdown in actin filaments. In order to show intracellular localization of CRM197 and visualize cell structure via actin cytoskeleton, endothelial cells were cultured and subjected to CRM197 in vitro. To address the interaction between CRM197 and actin both experimental and theoretical studies were carried out. Colocalization of CRM197 with actin filaments was determined by immunofluorescence microscopy. Following 24-ho…

Integrin-linked kinase regulates oligodendrocyte cytoskeleton, growth cone, and adhesion dynamics.

Integrin-linked kinase (ILK), a focal adhesion protein, brokers the link between cytoskeleton, cell membrane, and extracellular environment. Here, we demonstrate a role for ILK in laminin-2-mediated adhesion in primary murine oligodendrocytes (OLs) - with ILK loss leading to severe defects in process branching and outgrowth. These defects were partially recovered when the ILK-depleted OLs were instead grown on the non-integrin-activating substrate poly-l-lysine. Intriguingly, ILK loss on the neutral poly-l-lysine substrate led to swelling at the tips of OL processes, which we identified as enl…

F‑actin cytoskeleton reorganization is associated with hepatic stellate cell activation.

The activation of hepatic stellate cells (HSCs) is involved in the development of hepatic fibrosis. Previous studies have indicated that the acquisition of certain properties by activated HSCs is highly dependent on the reorganization of the actin cytoskeleton. However, direct evidence showing that the reorganization of the actin cytoskeleton is responsible for HSC activation is lacking. The aim of the present study was to investigate the role of cytoskeletal reorganization during HSC activation and to clarify the underlying mechanism. HSC-T6 cells were treated either with the F-actin stabiliz…

Adapter protein SH2B1beta cross-links actin filaments and regulates actin cytoskeleton.

The Src homology 2 (SH2) domain-containing adapter protein SH2B1beta plays a role in severe obesity, leptin and insulin resistance, and infertility. SH2B1beta was initially identified as a Janus tyrosine kinase 2 (JAK2) substrate, and it has been implicated in cell motility and regulation of the actin rearrangement in response to GH and platelet-derived growth factor. SH2B1beta is also required for maximal actin-based motility of Listeria. Here we have used a low-speed pelleting assay and electron microscopy to demonstrate that SH2B1beta has two actin-binding sites and that it cross-links acti

Direct interaction between anthrax toxin receptor 1 and the actin cytoskeleton.

The protective antigen component of anthrax toxin binds the I domain of the anthrax toxin receptors, ANTXR1 and ANTXR2, in a manner akin to how integrins bind their ligands. The I domains of integrins and ANTXR1 both have high- and low-affinity conformations, and the cytosolic tails of these receptors associate with the actin cytoskeleton. The association of ANTXR1 with the cytoskeleton correlates with weakened binding to PA, although a mechanistic explanation for this observation is lacking. Here, we identified a segment in the cytoplasmic tail of ANTXR1 required for its association with the …

Post-acute alterations in the axonal cytoskeleton after traumatic axonal injury.

All previous analyses of axonal responses to traumatic axonal injury (TAI) have described the ultrastructure of changes in the cytoskeleton and axolemma within 6 h of injury. In the present study we tested the hypothesis that there are, in addition, ultrastructural pathological changes up to 1 week after injury. TAI was induced in the adult guinea pig optic nerve of nine animals. Three animals were killed at either 4 h, 24 h, or 7 days (d) after injury. Quantitative analysis of the number or proportion of axons within 0.5-micro m-wide bins showed an increase in the number of axons with a diame…

RUNX1 N6-methyladenosine methylation enhances cytoskeleton remodelling and boosts cardiac fibrosis.

AIMS: The RUNX family of transcription factors is critical for heart development, physiology, and cardiovascular disease. However, current models of transcription factor binding seldom incorporate RNA modifications, and the latest methods that include them remain limited. This gap impedes accurate profiling of transcription factor affinities. In particular, the role of N6-methyladenosine (m6A)-mediated mechanisms in regulating RUNX factors during cardiac fibrosis is still poorly understood. METHODS AND RESULTS: RNA sequencing of human atrial fibrillation tissues identified transcription facto
