Emerging advances in drug delivery systems (DDSs) for optimizing cancer complications.

The management and treatment of tumor complications pose continuous challenges due to the inherent complexity. However, the advent of drug delivery systems (DDSs) brings promising opportunities to address the tumor complications using innovative technological approaches. This review focuses on common oncological complications, including cancer thrombosis, malignant serous effusion, tumor-associated infections, cancer pain, and treatment-related complications. Emphasis was placed on the application and potential of DDSs in mitigating and treating these tumor complications, and we delved into th…

The Application of PEG-functionalized Amphiphilic Pillararene in Drug Delivery Systems.

As a new generation of supramolecular host macrocyclic molecules, pillararenes display many advantages including simple synthesis routes, rigid electron-rich structures, unique host-guest interactions, and adjustable cavity sizes, which make them widely be used in drug delivery systems for precise diagnosis and treatment of cancer. The molecular functions of pillararenes can be easily expanded through the introduction of different functional groups into the side chain. For example, polyethylene glycol (PEG) which has good biocompatibility and water solubility can be used to construct PEG-funct…

Chitosan Nanoparticle-Based Drug Delivery Systems: Advances, Challenges, and Future Perspectives.

The effectiveness and uses of chitosan nanoparticles (CNPs) in drug delivery systems are examined in this work. Important results include the improved drug encapsulating efficiency: CNPs showed up to 90% encapsulation for different therapeutic agents. Furthermore, the research shows that CNPs provide extended-release patterns, greatly enhancing medication bioavailability especially for hydrophobic compounds. One interesting outcome was the greater drug stability in acidic surroundings, which are common in the stomach, where CNPs turn into a gel and later inflate in the intestine where the drug…

Potential Neuroprotective Strategies using Smart Drug Delivery Systems for Alzheimer's Disease.

BACKGROUND: Alzheimer's disease (AD) is the most common neurological disorder, affecting more than 50 million individuals worldwide and causing gradual but progressive cognitive decline. The rising cost of medical treatment is mostly attributable to AD. There are now mainly a few slightly symptomatic therapeutic options accessible. Although this is not the primary reason, the failure to develop effective treatments for AD is often attributed to the disease's complicated pathophysiology and the wide range of underlying ideas. OBJECTIVE: Studies undertaken over the past decade have aimed to fin

Smart stimuli-responsive drug delivery systems in spotlight of COVID-19.

The world has been dealing with a novel severe acute respiratory syndrome (SARS-CoV-2) since the end of 2019, which threatens the lives of many people worldwide. COVID-19 causes respiratory infection with different symptoms, from sneezing and coughing to pneumonia and sometimes gastric symptoms. Researchers worldwide are actively developing novel drug delivery systems (DDSs), such as stimuli-responsive DDSs. The ability of these carriers to respond to external/internal and even multiple stimuli is essential in creating "smart" DDS that can effectively control dosage, sustained release, individ…

Recent Progress in Bio-Responsive Drug Delivery Systems for Tumor Therapy.

Spatially- and/or temporally-controlled drug release has always been the pursuit of drug delivery systems (DDSs) to achieve the ideal therapeutic effect. The abnormal pathophysiological characteristics of the tumor microenvironment, including acidosis, overexpression of special enzymes, hypoxia, and high levels of ROS, GSH, and ATP, offer the possibility for the design of stimulus-responsive DDSs for controlled drug release to realize more efficient drug delivery and anti-tumor activity. With the help of these stimulus signals, responsive DDSs can realize controlled drug release more precisely…

Emerging technologies to increase gastrointestinal transit times of drug delivery systems.

Apart from already established technologies to increase gastrointestinal transit times, including devices rapidly increasing in size once they have reached the stomach in order to retard the passage through the pylorus, formulations that float on gastric fluids and mucoadhesive drug delivery systems adhering to the gastrointestinal mucosa, there are new technologies emerging that might be game changing. They include mucus permeating nanocarriers that are able to diffuse deeply into the mucus gel layer of the gastric and intestinal mucosa remaining there for a prolonged time period (i), charge-…

Nanomaterial based drug delivery systems for the treatment of neurodegenerative diseases.

With an aging population that has been increasing in recent years, the need for the development of therapeutic approaches for treatment of neurodegenerative disorders (ND) has increased. ND, which are characterized by the progressive loss of the structure or function of neurons, are often associated with neuronal death. In spite of screening numerous drugs, currently there is no specific treatment that can cure these diseases or slow down their progression. Alzheimer's disease (AD), Parkinson's disease (PD), amyotrophic lateral sclerosis (ALS), frontotemporal dementia, Huntington's disease, an

Food-Derived Nanoscopic Drug Delivery Systems for Treatment of Rheumatoid Arthritis.

Rheumatoid arthritis (RA) is a severe systemic inflammatory disease with no cure at present. Recent developments in the understanding of inflammation and nanomaterial science have led to increased applications of nanostructured drug delivery systems in the treatment of RA. The present review summarizes novel fabrications of nanoscale drug carriers using food components as either the delivered drugs or carrier structures, in order to achieve safe, effective and convenient drug administration. Polyphenols and flavonoids are among the most frequently carried anti-RA therapeutics in the nanosystem…
