Electron transfer kinetics during the reduction and turnover of the cytochrome caa3 complex from Bacillus subtilis.

The cytochrome caa3 complex from Bacillus subtilis is a member of the cytochrome oxidase superfamily of respiratory enzyme complexes. The key difference in the cytochrome caa3 complex lies in the addition of a domain, homologous with mitochondrial cytochrome c, that is fused to the C-terminal end of its subunit II. Measurements of steady-state and transient reduction kinetics have been carried out on the cytochrome caa3 complex. Reduction of the cyanide-bound enzyme with ascorbate and N,N,N',N'-tetramethyl-p-phenylenediamine (TMPD) supports a sequence of electron transfer in which cytochromec

Cortical cytochrome oxidase activity is reduced in Alzheimer's disease.

A defect in energy metabolism may play a role in the pathogenesis of neurodegenerative diseases, such as Alzheimer's disease. In the present study, we examined the activities of the enzymes that catalyze oxidative phosphorylation in frontal, temporal, parietal, and occipital cortex from Alzheimer's disease patients and age-matched controls. Complex I and complex II-III activities showed a small decrease in occipital cortex, but were unaffected in the other cortical areas. The most consistent change was a significant decrease of cytochrome oxidase (complex IV) activity of 25-30% in the four cor

Mitochondrial complex I mutations in Caenorhabditis elegans produce cytochrome c oxidase deficiency, oxidative stress and vitamin-responsive lactic acidosis.

Mitochondrial dysfunction, with an estimated incidence of 1 in 10 000 live births, is among the most common genetically determined conditions. Missense mutations in the human NDUFV1 gene, which encodes the 51 kDa active site subunit of the NADH-ubiquinone oxidoreductase or complex I, can lead to severe neurological disorders. Owing to the rare and often sporadic nature of mitochondrial disorders, the mechanisms of pathogenesis of most mutations remain poorly understood. We have generated transgenic strains of Caenorhabditis elegans that express disease-causing mutations in the nuo-1 gene, the

Cox7a2l Variation Influences Complex III and Mitochondrial Supercomplex Differences in 3xTG-AD Mixed-Background Mice.

Mitochondrial dysfunction, including impaired respiration and increased reactive oxygen species (ROS), is an early feature of Alzheimer's disease (AD) models. Electron transport chain supercomplexes (mSCs) regulate respiratory efficiency and ROS generation, yet genetic determinants of mSC organization in AD models remain underappreciated. Cox7a2l is known to promote CIII2/CIV association and CIV incorporation into mSCs. We examined the commonly used mixed-background B6;129S 3xTg-AD mice and B6129SF2/J controls aged 12-15 months, as well as specific rat AD models for changes in mSC organization

Euro-Asian hybrids of Echinococcus multilocularis from red foxes in northern and northeastern Poland result from secondary contact between long-isolated populations.

We investigated the mitochondrial genetic diversity of Echinococcus multilocularis in red foxes from northern and northeastern Poland, and tested the hypothesis that the presence of Asian haplotypes in Poland stems from their introduction from Northeast Asia to Eastern Europe. We targeted the cytochrome c oxidase subunit 1 gene (cox1) of 263 tapeworms from 59 red foxes hunted in selected districts of Pomorskie, Warmińsko-Mazurskie, and Podlaskie Voivodships. We calculated several measures of genetic variation, and constructed median-joining haplotype networks and a RelTime timetree. In additio

Methionine coordination to the heme iron and its displacement by nitric oxide in the CcoP subunit of Pseudomonas aeruginosa cytochrome cbb3 oxidase.

Ligand substitution is common in signaling mechanisms of heme proteins. Methionine at the heme iron can get displaced by both endogenous and exogeneous ligands, including NO. The latter is particularly relevant in low-O2 environments and affects properties of metalloproteins in electron-transfer (ET) chains. Inspection of the recent cryo-EM structures of cytochrome (cyt) cbb3 oxidases from Pseudomonas aeruginosa (Pa) suggested that FeS(Met) distances in the CcoP subunit are too long for Met ligation to the heme iron, yet this ligation is critical for high reduction potentials to support downhi

Genetic diversity and population structure of Oncomelania hupensis in Sichuan Province, China: implications for schistosomiasis control.

Schistosomiasis, caused by the infection with Schistosoma japonicum, remains a significant public health concern in China. As the sole intermediate host of S. japonicum, the breeding and spread of Oncomelania hupensis contribute significantly to the potential risk of disease occurrence and transmission. Exploring the population genetics of the snail vector is conducive to better understanding its distribution and dispersal patterns, and provides more data for future snail surveillance and control from a molecular perspective. The genetic diversity and population structure of O. hupensis in Sic

Morphological and molecular characterization of microfilariae in chickens (Gallus gallus domesticus) in Northeastern Thailand.

BACKGROUND: Filariasis is a parasitic disease caused by filarial nematodes. The life cycle involves vertebrates host and hematophagous arthropods. Filarial nematodes are viviparous, and their larvae (microfilaria) are produced into the blood circulation. Microfilariae are widely studied in humans and small animals, but are rarely found in domestic chickens. AIM: We aimed to study the prevalence, morphology, and molecular characteristics of microfilariae in village chickens raised in Thailand. MATERIALS AND METHODS: Sixty thin blood smears were prepared for parasite detection and morphotype o

First record of Argulus japonicus infestation on Cyprinus carpio in Hungary, and the first description of Argulus japonicus europaeus subsp. nov. Keve, 2025.

BACKGROUND: Species belonging to the genus Argulus are globally distributed fish parasites. Their veterinary significance primarily lies in their disruptive presence and their role as mechanical vectors. Although Argulus japonicus Thiele, 1900 is a widely distributed representative of this genus that feeds on freshwater fish, only Argulus foliaceus (Linnaeus, 1758) had previously been reported in Hungary. The aim of this study was to investigate the fish louse fauna in a local common carp (Cyprinus carpio Linnaeus, 1758) population. To the best of our knowledge, this is the first study to repo

New records of Trichinella patagoniensis from Argentina.

Trichinella spiralis was long considered the sole Trichinella species in Argentina. However, since 2004, various Trichinella species, including the encapsulated Trichinella patagoniensis and Trichinella britovi, as well as the unencapsulated Trichinella pseudospiralis, have been detected in the country. The present study aimed to identify Trichinella ML at the species level from cougars naturally infected from Argentina. To this end, muscle tissue samples from one cougar each from Córdoba, Neuquén, and Santa Cruz Provinces were individually analysed using the artificial digestion technique. DN

Molecular characterization of Armigeres subalbatus from Hyderabad region of Telangana state, India.

BACKGROUND OBJECTIVES: The mosquito Armigeres subalbatus (Coquillett, 1898) is a significant vector for Japanese encephalitis infection and breeds in high organic polluted water. Understanding mosquito diversity and their abundance in relation to mosquito-borne diseases is an important component for public health managers. Though the conventional methods for systematic position of mosquito species by using morphological characteristics is a classical method, it requires expertise and well-preserved specimens. Conversely, molecular analysis of mitochondrial cytochrome C oxidase subunit I (COI)

Heme-copper and Heme O2-derived synthetic (bioinorganic) chemistry toward an understanding of cytochrome c oxidase dioxygen chemistry.

Cytochrome c oxidase (CcO), also widely known as mitochondrial electron-transport-chain complex IV, is a multi-subunit transmembrane protein responsible for catalyzing the last step of the electron transport chain, dioxygen reduction to water, which is essential to the establishment and maintenance of the membrane proton gradient that drives ATP synthesis. Although many intermediates in the CcO catalytic cycle have been spectroscopically and/or computationally authenticated, the specifics regarding the IP intermediate, hypothesized to be a heme-Cu (hydro)peroxo species whose O-O bond homolysis

A novel homozygous variant in COX5A causes an attenuated phenotype with failure to thrive, lactic acidosis, hypoglycemia, and short stature.

Genetic defect in the nuclear encoded subunits of cytochrome c oxidase are very rare. To date, most deleterious variants affect the mitochondrially encoded subunits of complex IV and the nuclear genes encoded for assembly factors. A biallelic pathogenic variant in the mitochondrial complex IV subunit COX5A was previously reported in a couple of sibs with failure to thrive, lactic acidosis and pulmonary hypertension and a lethal phenotype. Here, we describe a second family with a 11-year-old girl presenting with failure to thrive, lactic acidosis, hypoglycemia and short stature. Clinical exome
