Pulmonary tuberculosis in Asian elephants (Elephas maximus): histologic lesions with correlation to local immune responses.

Although Mycobacterium tuberculosis infection is an important health concern for Asian elephants (Elephas maximus), no studies have evaluated the associated local immune responses or histologic lesions. In primates including humans, latent tuberculosis is distinguished by well-organized granulomas with TH1 cytokine expression, whereas active disease is characterized by poorly organized inflammation and local imbalance in TH1/TH2 cytokines. This study examined archival, formalin-fixed, paraffin-embedded lung samples from 5 tuberculosis-negative and 9 tuberculosis-positive Asian elephants. Lesio…

Liquid semen storage in elephants (Elephas maximus and Loxodonta africana): species differences and storage optimization.

Artificial insemination plays a key role in the genetic management of elephants in zoos. Because freshly extended semen is typically used for artificial insemination in elephants, it has become imperative to optimize conditions for liquid storage and semen transport. The objectives of this study were to examine the interactions between different extenders and storage temperatures on sperm total motility, progressive motility, and acrosomal integrity in Asian (Elephas maximus) and African (Loxodonta africana) elephants. Ejaculates were collected by rectal massage, diluted using a split-sample t…

Assessment of diurnal urinary cortisol excretion in Asian and African elephants using different endocrine methods.

Longitudinal urine samples were collected from Asian and African elephants to assess sample processing and immunoassay techniques for monitoring adrenal activity. Temporal profiles of urinary cortisol measured by RIA and EIA, with and without dichloromethane extraction, were similar; all correlation coefficients were >0.90. However, based on regression analyses, cortisol immunoactivity in extracted samples was only 72-81% of that of unextracted values. Within assay technique, RIA values were only 74-81% of EIA values. Collection of 24-hr urine samples demonstrated a clear diurnal pattern of gl…

Comparative endocrinology of cycling and non-cycling Asian (Elephas maximus) and African (Loxodonta africana) elephants.

Up to 14% of Asian and 29% of African elephants in captivity are not cycling normally or exhibit irregular cycles based on progestin profiles. To determine if ovarian acyclicity is related to other disruptions in endocrine activity, serum pituitary, thyroid, adrenal, and ovarian hormones in weekly samples collected for 6-25 months were compared between normal cycling (n=22 each species) and non-cycling (n=6 Asian; n=30 African) elephants. A subset of cycling females (n=4 Asian, 7 African) also were blood sampled daily during the follicular phase to characterize the peri-ovulatory period. In no…

Clinical and pathological findings of a newly recognized disease of elephants caused by endotheliotropic herpesviruses.

The unique clinical and pathological findings in nine Asian (Elephas maximus) and two African (Loxodonta africana) elephants from North American Zoos with a highly fatal disease caused by novel endotheliotropic herpesviruses are described. Identification of the viruses by molecular techniques and some epidemiological aspects of the disease were previously reported. Consensus primer polymerase chain reaction (PCR) combined with sequencing yielded molecular evidence that confirmed the presence of two novel but related herpesviruses associated with the disease, one in Asian elephants and the seco…

MEASURES OF DISEASE FREQUENCY FOR ELEPHANT ENDOTHELIOTROPIC HERPESVIRUS INFECTION AND HEMORRHAGIC DISEASE IN THE UNITED STATES ELEPHANT POPULATION FROM 2014 TO 2024.

Elephant endotheliotropic herpesvirus hemorrhagic disease (EEHV-HD) is an important cause of morbidity and mortality in juvenile Asian (Elephas maximus) and African (Loxodonta africana) elephants in human care. Measures of disease frequency have not been rigorously established for EEHV in any elephant population. The objective of this retrospective descriptive epidemiologic study was to determine period prevalence, cumulative incidence, and incidence rate (IR) for EEHV infection and EEHV-HD in the US elephant population between 2014 and 2024. A case of EEHV infection was defined as an elephant…

Colonic sand impaction with cecal rupture and peritonitis in an adult African savanna elephant, and review of noninfectious causes of gastrointestinal disease in elephants.

Gastrointestinal disorders are among the most common disease processes in captive elephants. Colic is a frequent clinical presentation and may have several infectious and noninfectious causes. Ingestion of sand has been reported in elephants living in enclosures with loose sandy soils. Similar to the situation in horses, sand ingestion can cause intestinal impaction and colic in elephants. Here we describe a case of colonic sand impaction with cecal rupture and peritonitis in an African savanna elephant from a zoologic collection that died after several days of colic. On autopsy, abundant, gri…

The occurrence of elephant endotheliotropic herpesvirus infection in wild and captive Asian elephants in Thailand: Investigation based on viral DNA and host antibody.

BACKGROUND AND AIM: Elephant endotheliotropic herpesvirus (EEHV) is a serious disease, threatening the life of young elephants. Many elephants have been infected with no clinical signs and may serve as carriers spreading this disease. It is important to monitor the disease through clinical signs and molecular diagnosis. In this study we investigated the occurrence of EEHV and the efficiency of different techniques used to monitor EEHV infection in various samples and populations of Asian elephants. MATERIALS AND METHODS: Blood and trunk swabs were collected from live elephants, while visceral…

Low gH/gL (Sub)Species-Specific Antibody Levels Indicate Elephants at Risk of Fatal Elephant Endotheliotropic Herpesvirus Hemorrhagic Disease.

Elephant endotheliotropic herpesviruses (EEHVs), of which eleven (sub)species are currently distinguished, infect either Asian (Elephas maximus) or African elephants (Loxodonta species). While all adult elephants are latently infected with at least one EEHV (sub)species, young elephants, specifically those with low to non-detectable EEHV-specific antibody levels, may develop fatal hemorrhagic disease (EEHV-HD) upon infection. However, animals with high antibody levels against EEHV(1A) gB, an immunodominant antigen recognized by antibodies elicited against multiple (sub)species, may also occasi…

Supernumerary Tusks in African Savanna Elephants (Loxodonta africana): A Case Series and Consideration of Developmental Mechanisms.

Supernumerary teeth are well recognised in humans and domestic animals but are rarely reported in elephants. This case series describes four African savanna elephants (Loxodonta africana) with multiple tusk structures occupying a single alveolus. A 7-year-old bull presented with a deformed left tusk and behavioural changes. Clinical examination revealed two coronal openings, followed by eruption of three accessory tusks. Conservative intracavitary treatment and selective surgical removal of the accessory tusks preserved the primary tusk, which maintained normal eruption and function at 36-mont…

SINGLE-DOSE, MULTIPLE-DOSE, AND THERAPEUTIC DRUG MONITORING PHARMACOKINETICS OF FIROCOXIB IN ASIAN ELEPHANTS (ELEPHAS MAXIMUS).

Firocoxib is a COX-2-selective nonsteroidal anti-inflammatory drug (NSAID) with limited effects on COX-1, which means it likely has fewer side effects than typically associated with other NSAIDs. This study determined possible doses of firocoxib based on single- and multidose pharmacokinetic trials conducted in 10 Asian elephants (Elephas maximus). Initially, two single oral dose trials (0.01 and 0.1 mg/kg) of a commercially available tablet (n = 6) and paste (n = 4) formulation were used to determine a preferred dose. The 0.1 mg/kg dose was further evaluated via IV single dose (n = 3) and ora…

Of Elephants and Other Mammals: A Comparative Review of Reproductive Tumors and Potential Impact on Conservation.

Reproductive tumors can impact conception, pregnancy, and birth in mammals. These impacts are well documented in humans, while data in other mammals are limited. An urgent need exists to understand the reproductive impact of these lesions in endangered species, because some endangered species have a documented high prevalence of reproductive tumors. This article documents that the prevalence of both benign and malignant neoplasia differs between African and Asian elephants, with Asian elephants more frequently diagnosed and negatively affected by both. The prevalence of these tumors across mam…

Comparison of methods for faecal hormone preservation and analysis in African savanna elephants under field conditions.

Noninvasive faecal hormone analyses can provide valuable information on the physiological state of wild animals and how they respond to ecological changes or anthropogenic disturbances. However, preservation techniques to prevent hormone alteration can be problematic, and not all are field friendly. We compared five processing methodologies to preserve samples for faecal glucocorticoid, progestagen and thyroid hormone metabolites. Samples were collected from adult zoo Africa savanna elephants (Loxodonta africana) (one male, four females) immediately after defecation. Subsamples were then subje…

Local rainfall is more likely than distant thunderstorms to affect movement behaviour in Northern Kenyan elephants.

Previous research indicates that African savanna elephants change their movements preceding or coincident with local rainfall and it has been suggested that they respond to thunder in remote storms-perhaps reading seismic cues. We therefore aimed to test if elephants in Northern Kenya adhere to distinct daytime movement states between the wet and dry periods, and whether their abrupt movement changes precede local wet periods in response to lightning strikes from a specific compass heading. In our study site, lightning to the North and East often preceded local rainfall and could possibly be u…

Seroprevalence of leptospirosis in domesticated Asian elephants (Elephas maximus) in north and west Thailand in 2004.

Serum samples from Asian elephants (Elephas maximus) in the Kanchanaburi, Chiang Mai and Lampang provinces of Thailand were tested using the microscopic agglutination test against 22 serovars of Leptospira interrogans. A titre of more than 1:100 was used as evidence of infection. In northern Thailand, the seroprevalence was 58 per cent and the prevalent serovars were Leptospira interrogans serovar Sejroe, Leptospira interrogans serovar Tarassovi, Leptospira interrogans serovar Ranarum and Leptospira interrogans serovar Shermani. In western Thailand, the seroprevalence was 57 per cent and the p…

Virological investigation and comparative genomic analysis of elephant endotheliotropic herpesvirus 1B infection in an Australian captive herd of Asian elephants (Elephas maximus).

Elephant endotheliotropic herpesviruses (EEHV) pose a significant threat to the conservation of Asian elephants (Elephas maximus) worldwide, with a high mortality rate in young elephants. However, several components of EEHV virology remain underexplored, particularly for EEHV1B. This study describes a fatal case of EEHV1B infection in a nine-year-old Asian elephant from an ex situ conservation herd, examining herd viral dynamics, tissue viral loads and comparative genomics. This elephant succumbed to haemorrhagic disease within three days of developing clinical signs, despite therapeutic inter…

Florida elephant trunk hybrid aortic repair (FETHAR): a novel frozen elephant trunk technique with complete debranching sparing endograft modification.

Treatment of aortic arch pathology is rapidly evolving. Current techniques utilize a combination of open and endovascular approaches. Existing techniques such as B-SAFER require endograft modification for complete arch debranching which may predispose the graft to failure or endoleak. We developed the FETHAR, a novel method for hybrid aortic repair and demonstrate its use in a patient who presented with an acute dissection with contained rupture. Following repair, we demonstrate excellent seal without endoleak on the post-operative CT scan. Ultimately, this repair allows for complete arch debr

Comparative study of the frozen elephant trunk and classical elephant trunk techniques to supplement total arch replacement for acute type A aortic dissection†.

OBJECTIVES: In patients with acute type A aortic dissection, the use of the frozen elephant trunk (FET) procedure with total arch replacement (TAR) has been indicated for emergency operations to obtain thrombosis of the distal false lumen (FL). However, data comparing the FET and the classical elephant trunk (CET) procedures, including the incidences of mortality, morbidity, spinal cord injury and aortic remodelling, have not yet been reported. The goal of this study was to compare the early outcomes of TAR with the FET and the CET procedures. METHODS: The past 7 years of medical records of 3

The Elephant in the Room: Reckoning With Collegiality and Authorship in Academic Pharmacy.

Collaborative projects help faculty produce scholarship while balancing teaching, research, and service responsibilities. However, faculty's assumptions about contributions and authorship definitions can lead to confusion and disappointment in collaborative scholarship. Lapses in communication can cause delays, uneven workloads, and perceptions of unfairness. Inconsistent application of authorship criteria and a lack of transparency in authorship order may further create frustration and disengagement for future projects, particularly, when their contributions affect promotion and tenure.
