alpha4beta3delta GABA(A) receptors characterized by fluorescence resonance energy transfer-derived measurements of membrane potential.

Selective modulators of gamma-aminobutyric acid, type A (GABA(A)) receptors containing alpha(4) subunits may provide new treatments for epilepsy and premenstrual syndrome. Using mouse L(-tk) cells, we stably expressed the native GABA(A) receptor subunit combinations alpha(3)beta(3)gamma(2,) alpha(4)beta(3)gamma(2), and, for the first time, alpha(4)beta(3)delta and characterized their properties using a novel fluorescence resonance energy transfer assay of GABA-evoked depolarizations. GABA evoked concentration-dependent decreases in fluorescence resonance energy transfer that were blocked by GA…

Comparison of the Medical Uses and Cellular Effects of High and Low Linear Energy Transfer Radiation.

Exposure to ionizing radiation can occur during medical treatments, from naturally occurring sources in the environment, or as the result of a nuclear accident or thermonuclear war. The severity of cellular damage from ionizing radiation exposure is dependent upon a number of factors including the absorbed radiation dose of the exposure (energy absorbed per unit mass of the exposure), dose rate, area and volume of tissue exposed, type of radiation (e.g., X-rays, high-energy gamma rays, protons, or neutrons) and linear energy transfer. While the dose, the dose rate, and dose distribution in tis…

Time-resolved spectroscopy of radiative energy transfer between a conjugated oligomer and polymer in solution.

We report a short investigation of the energy transfer process between the conjugated oligomer 1,4-bis(9-ethyl-3-carbazo-vinylene)-9,9-dihexyl-fluorene (BECV-DHF) and the conjugated polymer poly[2-methoxy-5-(3,7-dimethyloctyloxy)-1,4-phenylene-vinylene] - end capped with DMP (MDMO-PPV). The radiative energy transfer (RET) process shows a time delay, and the formation of the excimer causes a further delay. All these processes were studied using time-resolved spectroscopy (TRS), which has three-dimensional (3D) features with wavelength, intensity and time (picosecond) as the X, Y and Z-axis, res…

Disruption of Energy Transfer and Redox Status by Sulfite in Hippocampus, Striatum, and Cerebellum of Developing Rats.

Patients with sulfite oxidase (SO) deficiency present severe brain abnormalities, whose pathophysiology is not yet elucidated. We evaluated the effects of sulfite and thiosulfate, metabolites accumulated in SO deficiency, on creatine kinase (CK) activity, mitochondrial respiration and redox status in hippocampus, striatum and cerebellum of developing rats. Our in vitro results showed that sulfite and thiosulfate decreased CK activity, whereas sulfite also increased malondialdehyde (MDA) levels in all brain structures evaluated. Sulfite further diminished mitochondrial respiration and increased

Modulation of defect-mediated energy transfer from ZnO nanoparticles for the photocatalytic degradation of bilirubin.

In recent years, nanotechnology has gained significant interest for applications in the medical field. In this regard, a utilization of the ZnO nanoparticles for the efficient degradation of bilirubin (BR) through photocatalysis was explored. BR is a water insoluble byproduct of the heme catabolism that can cause jaundice when its excretion is impaired. The photocatalytic degradation of BR activated by ZnO nanoparticles through a non-radiative energy transfer pathway can be influenced by the surface defect-states (mainly the oxygen vacancies) of the catalyst nanoparticles. These were modulated…

Enhanced Energy Transfer from a Metal-Organic Framework to a Highly Confined Organic Phosphorescent Dye.

Three-Dimensional Metal-Organic Frameworks (MOFs) are crystalline, porous hybrid materials with well-defined pore structures, offering isotr … rmed using (cryo)fluorescence spectroscopy on powders and thin films. We consider Förster Resonance Energy Transfer (FRET) as a process of energy transfer between the pore walls of MIL-68 and 11-DTCz-BP. The aggregation control leads to a quantum yield enhancement, while FRET enables long lifetimes of phosphorescence at room-temperature.

Uranyl-ion Directed Hydrogelation of Cholate for Enhanced Emission Resolution and Energy Transfer-driven Sensing.

In this work, we report the first example of an actinide (UO2 2+) induced hydrogel formation. We discovered that the gel phase led to a remarkable improvement in the resolution of the emission from the uranyl cation. This phenomenon was previously seen only in sophisticated uranyl complexes in organic solvents or in the solid-state. The organized and confined gel microenvironment facilitated efficient energy transfer from UO2 2+→ Eu3+ in the hydrogel matrix. Additionally, this "turn-on" energy transfer strategy provided a simple method for uranyl sensing using commercially available ingredient…

Robust Optimization for Spot-Scanning Proton Therapy based on Dose-Linear-Energy-Transfer Volume Constraints.

PURPOSE: Historically, spot-scanning proton therapy (SSPT) treatment planning uses dose-volume constraints and linear-energy-transfer (LET) volume constraints separately to balance tumor control and organs-at-risk (OARs) protection. We propose a novel dose-LET-volume constraint (DLVC)-based robust optimization (DLVCRO) method for SSPT in treating prostate cancer to obtain a desirable joint dose and LET distribution to minimize adverse events. METHODS AND MATERIALS: DLVCRO treats DLVC as soft constraints that control the shapes of the dose-LET volume histogram (DLVH) curves. It minimizes the o

Revealing the Ultrafast Energy Transfer Pathways in Energetic Materials: Time-Dependent and Quantum State-Resolved.

Intramolecular vibrational energy transfer is gaining tremendous attention as a regulator of condensed-phase behavior and reactions. In polyatomic molecules, this transfer is an ultrafast process involving multiple modes with numerous quantum states. The inherent complexity and rapid evolution of these processes pose significant challenges to experimental observation, and the high computational costs make full quantum mechanical calculations impractical with current technology. In the intramolecular energy transfer process, whether the doorway modes are intermediaries for transferring energy f…

Comparison of linear energy transfer measurement for therapeutic carbon beam using CR-39 and TLD.

The measurement of linear energy transfer (LET) is crucial for the evaluation of the radiation effect in heavy ion therapy. As two detectors which are convenient to implant into the phantom, the performance of CR-39 and thermoluminescence detector (TLD) for LET measurement was compared by experiment and simulation in this study. The results confirmed the applicability of both detectors for LET measurements, but also revealed that the CR-39 detector would lead to potential overestimation of dose-averaged LET compared with the simulation by PHITS, while the TLD would have a large uncertainty mea…

Reversible Ratiometric Electrochemiluminescence Biosensor Based on DNAzyme Regulated Resonance Energy Transfer for Myocardial miRNA Detection.

Myocardial miRNAs in peripheral blood are closely related to the pathogenic process of myocardial infarction. Rapid identification and accurate quantification of myocardial miRNAs are of great significance to clinical interventions for treating cardiovascular lesions. Therefore, a ratiometric electrochemiluminescence (ECL) biosensor integrating DNAzyme with a resonance energy transfer (RET) system was designed to detect myocardial miRNA. The dual-signal system was composed of rA marked substrate strand functionalized CdTe quantum dots (QDs) as reductive-oxidative (R-O) emitters and Cy5-labeled…

Detection of rabies virus via exciton energy transfer between CdTe quantum dots and Au nanoparticles.

Rabies is a fatal encephalitis caused by the rabies virus. The diagnosis of the disease depends in large part on the exposure history of the victim and clinical manifestations of the disease. Rapid rabies diagnosis is an important step in its prevention and control. Therefore, for accurate and timely diagnosis and prevention of rabies, we developed nanomaterials for a novel photoelectrochemical biosensing approach (PBA) for the rapid and reliable diagnosis of rabies virus. This approach uses high-efficiency exciton energy transfer between cadmium telluride quantum dots and Au nanoparticles and…

Immunoassay for serodiagnosis of Zika virus infection based on time-resolved Förster resonance energy transfer.

Zika virus (ZIKV) is a mosquito-borne pathogen causing a febrile illness with arthralgia, conjunctivitis and rash. The complications include Guillain-Barré syndrome, congenital brain and other abnormalities and miscarriage. The serodiagnosis of ZIKV infection is hampered by cross-reactivity with other members of the Flavivirus family, notably dengue (DENV). This report describes a novel serological platform for the diagnosis of ZIKV infection. The approach utilizes time-resolved Förster resonance energy transfer (TR-FRET) elicited by two chromophore-labeled proteins (a ZIKV antigen and a super…

Boronate Affinity Fluorescent Nanoparticles for Förster Resonance Energy Transfer Inhibition Assay of cis-Diol Biomolecules.

Förster resonance energy transfer (FRET) has been essential for many applications, in which an appropriate donor-acceptor pair is the key. Traditional dye-to-dye combinations remain the working horses but are rather nonspecifically susceptive to environmental factors (such as ionic strength, pH, oxygen, etc.). Besides, to obtain desired selectivity, functionalization of the donor or acceptor is essential but usually tedious. Herein, we present fluorescent poly(m-aminophenylboronic acid) nanoparticles (poly(mAPBA) NPs) synthesized via a simple procedure and demonstrate a FRET scheme with suppre…

Nanostructured biosensor for detecting glucose in tear by applying fluorescence resonance energy transfer quenching mechanism.

In this paper, a nanostructured biosensor is developed to detect glucose in tear by using fluorescence resonance energy transfer (FRET) quenching mechanism. The designed FRET pair, including the donor, CdSe/ZnS quantum dots (QDs), and the acceptor, dextran-binding malachite green (MG-dextran), was conjugated to concanavalin A (Con A), an enzyme with specific affinity to glucose. In the presence of glucose, the quenched emission of QDs through the FRET mechanism is restored by displacing the dextran from Con A. To have a dual-modulation sensor for convenient and accurate detection, the nanostru…

Generalized master equation with non-Markovian multichromophoric Förster resonance energy transfer for modular exciton densities.

A generalized master equation (GME) governing quantum evolution of modular exciton density (MED) is derived for large scale light harvesting systems composed of weakly interacting modules of multiple chromophores. The GME-MED offers a practical framework to incorporate real time coherent quantum dynamics calculations of small length scales into dynamics over large length scales, and also provides a non-Markovian generalization and rigorous derivation of the Pauli master equation employing multichromophoric Förster resonance energy transfer rates. A test of the GME-MED for four sites of the Fen…

Catabolic cancer-associated fibroblasts transfer energy and biomass to anabolic cancer cells, fueling tumor growth.

Fibroblasts are the most abundant "non-cancerous" cells in tumors. However, it remains largely unknown how these cancer-associated fibroblasts (CAFs) promote tumor growth and metastasis, driving chemotherapy resistance and poor clinical outcome. This review summarizes new findings on CAF signaling pathways and their emerging metabolic phenotypes that promote tumor growth. Although it is well established that altered cancer metabolism enhances tumor growth, little is known about the role of fibroblast metabolism in tumor growth. New studies reveal that metabolic coupling occurs between cataboli

Efficiency of energy transfer from organic dye molecules to CdSe-ZnS nanocrystals: nanorods versus nanodots.

We report on comparative experimental study of FRET efficiency in two different systems: organic dye molecules (donors) and CdSe-ZnS core-shell nanodots or nanorods (acceptors). Fluorescein isothiocyanate was bound chemically to the surface of nanocrystals using cysteine as a linker and the conjugates were embedded into the polymeric films. Contrary to intuitive presumptions based on the order of magnitude larger molar absorption coefficient for nanorods, the experiment demonstrated almost equal efficiency in the energy transfer from FITC to nanorods and nanodots. This effect is attributed to …
