Infralimbic Neurotrophin-3 Infusion Rescues Fear Extinction Impairment in a Mouse Model of Pathological Fear.

The inability to properly extinguish fear memories constitutes the foundation of several anxiety disorders, including panic disorder. Recent findings show that boosting prefrontal cortex synaptic plasticity potentiates fear extinction, suggesting that therapies that augment synaptic plasticity could prove useful in rescue of fear extinction impairments in this group of disorders. Previously, we reported that mice with selective deregulation of neurotrophic tyrosine kinase receptor, type 3 expression (TgNTRK3) exhibit increased fear memories accompanied by impaired extinction, congruent with an…

Controlled cortical impact before or after fear conditioning does not affect fear extinction in mice.

Post-traumatic stress disorder (PTSD) is characterized in part by impaired extinction of conditioned fear. Traumatic brain injury (TBI) is thought to be a risk factor for development of PTSD. We tested the hypothesis that controlled cortical impact (CCI) would impair extinction of fear learned by Pavlovian conditioning, in mice. To mimic the scenarios in which TBI occurs prior to or after exposure to an aversive event, severe CCI was delivered to the left parietal cortex at one of two time points: (1) Prior to fear conditioning, or (2) after conditioning. Delay auditory conditioning was achiev…

On the automatic nature of phobic fear: conditioned electrodermal responses to masked fear-relevant stimuli.

Normal subjects (n = 64) were exposed either to pictures of snakes and spiders or to pictures of flowers and mushrooms in a differential conditioning paradigm in which one of the pictures signaled an electric shock. In a subsequent extinction series, these stimuli were presented backwardly masked by another stimulus for half of the subjects, whereas the other half received non-masked extinction. In support of a hypothesis that suggests that nonconscious information-processing mechanisms are sufficient to activate responses to fear-relevant stimuli, differential skin conductance response to mas…

Adversity-induced relapse of fear: neural mechanisms and implications for relapse prevention from a study on experimentally induced return-of-fear following fear conditioning and extinction.

The efficacy of current treatments for anxiety disorders is limited by high relapse rates. Relapse of anxiety disorders and addiction can be triggered by exposure to life adversity, but the underlying mechanisms remain unexplored. Seventy-six healthy adults were a priori selected for the presence or absence of adverse experiences during childhood (CA) and recent past (RA; that is, past 12 months). Participants underwent fear conditioning (day 1) and fear extinction and experimental return-of-fear (ROF) induction through reinstatement (a model for adversity-induced relapse; day 2). Ratings, aut…

The impact of stress on fear memory retention: A meta-analysis of rodent fear conditioning studies.

Pavlovian fear conditioning is a widely used behavioural task for studying fear memory in rodents. During conditioning, rodents learn to associate a conditioned stimulus (e.g., context or tone; contextual or auditory fear conditioning, CFC or AFC, respectively) with an aversive one (e.g., footshock), resulting in a conditioned fear response. Fear memory retention is assessed thorough freezing behaviour, a species-specific defensive reaction, observed during exposure to the conditioned stimulus alone. Fear memory is influenced by sex and stress, with stress exposure prior to conditioning potent…

Fear in the mind's eye: the neural correlates of differential fear acquisition to imagined conditioned stimuli.

Mental imagery is involved in both the expression and treatment of fear-related disorders such as anxiety and post-traumatic stress disorder. However, the neural correlates associated with the acquisition and generalization of differential fear conditioning to imagined conditioned stimuli are relatively unknown. In this study, healthy human participants (n = 27) acquired differential fear conditioning to imagined conditioned stimuli paired with a physical unconditioned stimulus (i.e. mild shock), as measured via self-reported fear, the skin conductance response and significant right anterior i…

Screening for Perinatal Anxiety Using the Childbirth Fear Questionnaire: A New Measure of Fear of Childbirth.

Fear of childbirth affects as many as 20% of pregnant people, and has been associated with pregnancy termination, prolonged labour, increased risk of emergency and elective caesarean delivery, poor maternal mental health, and poor maternal-infant bonding. Currently available measures of fear of childbirth fail to fully capture pregnant people's childbirth-related fears. The purpose of this research was to develop a new measure of fear of childbirth (the Childbirth Fear Questionnaire; CFQ) that would address the limitations of existing measures. The CFQ's psychometric properties were evaluated …

Specificity of emotional maintenance processes among contamination fears and blood-injection-injury fears.

Research evidence consistently demonstrates a relation between disgust and anxiety-related pathology. Despite ample evidence implicating a functional role of disgust in anxiety, limited research has focused on the process by which disgust influences anxiety. Recent evidence indicates that fear of responding with disgust predicts contamination fears, thus elucidating a process by which disgust contributes to contamination fears. In the current study, we tested whether fear of responding with disgust is specific to contamination fears or generalizes to blood-injection-injury (BII) fears. Undergr…

Fear of pain and fear of falling among younger and older adults with musculoskeletal pain conditions.

BACKGROUND: The fear-avoidance model of pain accounts for the debilitating consequences of strong fear as a component of reactions to painful conditions (eg, movements become anxiety-provoking because they cause pain). The model, which is supported in the literature, posits that fear of pain interferes with recovery because it leads to avoidance of beneficial activity. Despite the high prevalence of pain among the elderly, investigations of fear of pain in this population are scarce. A related construct, fear of falling, has been studied among elderly (but not younger) adults as an age-specifi…

Failure to overcome 'innate' fear: a developmental test of the non-associative model of fear acquisition.

The non-associative, Darwinian theory of fear acquisition proposes that some individuals fail to overcome biologically-relevant fears (e.g. height) because they (1) do not have sufficient safe exposure to the relevant stimuli early in life or (2) are poor habituators who have difficulty 'learning not to fear'. These two hypotheses were tested in a longitudinal birth cohort study. Study 1 found evidence for reduced exposure to height stimuli in childhood for individuals with a fear of heights compared to study members without fear. Study 2 found evidence for higher levels of stress reactivity (…

Inactivation of medial or lateral orbitofrontal cortex during fear extinction did not interfere with fear renewal.

Hyperactive orbitofrontal cortical activation is commonly seen in patients of obsessive-compulsive disorder (OCD). Previous studies from our laboratory showed that for rats with aberrant activation of the orbitofrontal cortex (OFC) during the extinction phase, they were unable to use contexts as the reference for proper retrieval of fear memory during renewal test. This result supported the phenomenon that many OCD patients show poor regulation of fear-related behavior. Since there are robust anatomical connections of the OFC with the fear-circuit, we aim to further examine whether the OFC is …

The next generation of fear generalization.

Generalizing from past experience is a fundamental feature of cognition. Although generalization has been of long-standing interest to diverse domains in psychological science, the past decade has seen a rapid acceleration of fear generalization research in particular. The field of human fear generalization is at a pivotal juncture where it is now possible to synthesize extensive behavioral work, competing theoretical models, animal and human neuroscience, and clinical and individual-differences research to address enduring gaps. In this review, we provide an updated account of where the field…

Neuroscience in pictures: Neuroscience of fear.

Fear is an evolutionarily adaptive mechanism that enables organisms to detect and respond to potential threats. Over the past century, theories of fear have evolved from Pavlovian and behavioral frameworks to neuroscientific models that emphasize specific neural circuits, neurotransmission, and plasticity. This pictorial review synthesizes key concepts underlying the neuroscience of fear, starting with a historical narrative of fear as conceptualized by conditioning and behaviorism theories, to the roles of different neurobiology structures, networks, to concepts of memory consolidation, recon…

Neuromodulation of Fear and Anxiety Circuits.

Anxiety is a complex and heterogeneous condition consisting of multiple component processes and neural underpinnings. Recent neural accounts for anxiety have expanded beyond the canonical anxiety and fear circuitry centered on the amygdala to a distributed network. A burgeoning form of clinical intervention with remarkable potential to directly engage the neural substrates of anxiety is non-invasive brain stimulation (NIBS), which is especially notable for the ability to modulate macro- and meso-scopic circuits and networks. NIBS is poised as a powerful research tool with the ability to extend…

[Perioperative anxiety and fear of death].

An important field of anesthesiology but also of psychiatry and psychotherapy, is perioperative anxiety and especially the fear of death. In this review article the most important types of anxiety in the individual phases before, during and after surgery are presented and diagnostic aspects as well as risk factors are discussed. Benzodiazepines can classically be used therapeutically here, but in recent years the preoperative anxiety-reducing effects of e.g., supporting talks, acupuncture, aroma therapy, and relaxation methods have come more into focus, because benzodiazepines promote postoper…

Fear of hypoglycemia-An underestimated problem.

INTRODUCTION: Fear of hypoglycemia (FOH) is a phenomenon that affects people with diabetes experiencing hypoglycemia. On the one hand, FOH is an adaptive mechanism that helps to protect patients from hypoglycemia and its consequences. On the other hand, the non-normative level of FOH causes anxiety and tension, disturbs normal functioning, and makes normoglycemia maintenance difficult. OBJECTIVE: The main objective of this review was to describe factors influencing FOH and methods of measurement of FOH levels. Moreover, we highlighted the impact of the new technologies used in diabetes therap…

Treatment Options for Fear of Blushing.

PURPOSE OF REVIEW: To review mechanisms of blushing and fear of blushing from physiological, neuropharmacological and psychological viewpoints, and to evaluate current forms of treatment for blushing-related fear. RECENT FINDINGS: Blushing appears to be driven primarily by sympathetic adrenomedullary and neural vasodilator discharge, possibly in association with secondary neurovascular inflammation. Psychological risk factors for fear of blushing include social anxiety, coupled with heightened self-focused attention and inflated beliefs about the likelihood and social costs of blushing. In ad…
