Tonic inhibition in principal cells of the amygdala: a central role for α3 subunit-containing GABAA receptors.

GABAergic inhibition in the amygdala is essential in regulating fear and anxiety. Although fast "phasic" inhibition arising through the activation of postsynaptic GABA(A) receptors (GABA(A)Rs) has been well described in the amygdala, much less is known about extrasynaptic GABA(A)Rs mediating persistent or tonic inhibition and regulating neuronal excitability. Here, we recorded tonic currents in the basolateral (BLA) nucleus and the lateral (LA) nucleus of the amygdala. While all BLA principal cells expressed a robust GABAergic tonic current, only 70% of LA principal cells showed a tonic curren

Expression levels of the alpha4 subunit of the GABA(A) receptor in differentiated neuroblastoma cells are correlated with GABA-gated current.

The alpha4 subunit of the GABA(A) receptor (GABAR) is capable of rapid plasticity, increased by chronic exposure to positive GABA modulators, such as the neurosteroid 3alpha-OH-5alpha[beta]-pregnan-20-one (THP). Here, we show that 48 h exposure of differentiated neuroblastoma cells (IMR-32) to 100 nM THP increases alpha4 expression, without changing the current density or the concentration-response curve. Increased expression of alpha4-containing GABAR was verified by a relative insensitivity of GABA (EC(20))-gated current to modulation by the benzodiazepine (BZ) lorazepam (0.01-100 microM), a

The action of perchlorate on malignant-hyperthermia-susceptible muscle.

To better understand the altered skeletal muscle excitation-contraction (E-C) coupling that occurs in malignant hyperthermia, we have examined the potentiating actions of perchlorate in intact muscle fiber bundles, isolated sarcoplasmic reticulum (SR) vesicles, and the purified ryanodine receptor/Ca2+ release channel (RyR) isolated from malignant-hyperthermia-susceptible (MHS) and normal porcine muscle. The concentration of perchlorate that half-maximally potentiated twitch tension (2.5-3.5 mM) was not significantly different for MHS and normal muscles. The effect of perchlorate on fractional

Postoperative left ventricular contractility in the cardiac surgical patient: an evaluation of the force-interval relationship.

The force-interval relationship--the dependence of cardiac contractility on the rate and pattern of stimulation--was evaluated for its potential use in monitoring patients in the period immediately following heart surgery. Six patients were studied for three days after coronary artery bypass grafting. The monitoring instrumentation used during operation included a catheter-tip micromanometer introduced into the left ventricle, a pair of ultrasonic transducers placed on the left ventricular (LV) epicardium to monitor minor-axis dimension, pacing electrodes placed on the right atrium, and system

Combined multi-metric assessment of diaphragm contractile function in healthy humans: Feasibility, validity and reliability.

The combined use of subcostal ultrasonography and respiratory manometry represents a novel, integrative method for quantifying diaphragm contractile function (force, velocity and power). We evaluated the technical feasibility, construct validity and within-day test-retest reliability of this method during non-volitional, volitional and reflexive respiratory perturbations in healthy adults. Two independent cohorts were studied. In Experiment 1 (n = 10), diaphragm excursion (subcostal ultrasonography) and transdiaphragmatic pressure (Pdi, manometry) were measured during unilateral magnetic phren

Central excitability contributes to supramaximal volitional contractions in human incomplete spinal cord injury.

Despite greater muscle fatigue in individuals with spinal cord injury (SCI) when compared to neurologically intact subjects using neuromuscular electrical stimulation (NMES)protocols, few studies have investigated the extent of volitional fatigue in motor incomplete SCI. Using an established protocol of 20 repeated, intermittent, maximal volitional effort (MVE) contractions, we previously demonstrated that subjects with incomplete SCI unexpectedly demonstrated a 15% increase in peak knee extensor torques within the first five MVEs with minimal evidence of fatigue after 20 contraction. In the p

Reduced neuromuscular quantal content with normal synaptic release time course and depression in canine motor neuron disease.

Hereditary canine spinal muscular atrophy is an autosomal dominant version of motor neuron disease in which motor units exhibit extensive dysfunction before motor terminal or axonal degeneration appear. We showed in a previous paper that motor endplate currents (EPCs) are reduced and that failures of nerve-evoked EPCs appear in the homozygote medial gastrocnemius (MG) muscle in which failing motor units are also found, suggesting a presynaptic deficit of ACh release. To examine this further, we performed a detailed analysis of synaptic release properties in the MG muscle of homozygotes and com

The effects of caffeine on the contraction of the frog heart.

1. The force of contraction and the membrane potentials have been measured from preparations of frog heart, with methods that also allow the rapid exchange of the extracellular fluid.2. In regularly beating preparations, caffeine induces only weak contractures at temperatures above 15 degrees C, but it does cause a marked potentiation of the twitch responses; longer exposure results in a depression of contraction. The build up and decline of the twitch strength, on addition and on removal of caffeine approximates to a single exponential, time constant 26-45 sec, and this time constant is not a

Investigating cannabidiol rescue of stress-induced cognitive impairment and metabolism-driven safety in Human-on-a-Chip® models.

This manuscript combined two studies that assessed the effects of cannabidiol (CBD) exposure in distinct organ-on-a-chip models. Hesperos, Inc.'s Human-on-a-Chip® (HoaC) technology utilizes custom-designed human-based, serum-free, and pumpless microphysiological systems (MPS). The first study, study 1, assessed the level of off-target toxicity for CBD and its associated metabolites, 7-carboxy cannabidiol and 7-hydroxy cannabidiol, using a multi-organ heart-liver system. In study 1, CBD was delivered to systems with and without the inclusion of a liver organ module to evaluate metabolic contrib

Thyroid hormone effects on the State IV proton motive force in rat liver mitochondria.

In order to further investigate the mechanisms regulating the control of mitochondrial respiration by thyroid hormone, the proton motive force was measured during State IV respiration in liver mitochondria isolated from euthyroid, hyperthyroid, hypothyroid and T3-treated hypothyroid rats. The proton motive force was significantly higher in the hyperthyroid group due to an increased delta pH. The proton motive force of hypothyroid mitochondria was lower than controls due to a decreased membrane potential. The proton motive force for the T3-treated hypothyroid group did not differ from the euthy

CCCP Facilitates Aminoglycoside to Kill Late Stationary-Phase Escherichia coli by Elevating Hydroxyl Radical.

Improving the efficacy of existing antibiotics is significant for combatting antibiotic resistance that poses a major threat to human health. Carbonyl cyanide m-chlorophenylhydrazine (CCCP), a well-known protonophore for dissipating proton motive force (PMF), has been widely used to block the PMF-dependent uptake of aminoglycoside antibiotics and thus suppress aminoglycoside lethality. Here, we report that CCCP and its functional analog FCCP, but not other types of protonophores, unprecedently potentiate aminoglycosides (e.g., tobramycin and gentamicin) by 3-4 orders of magnitude killing of Es

A comparison of the effects of 6 weeks of traditional resistance training, plyometric training, and complex training on measures of strength and anthropometrics.

Complex training (CT; alternating between heavy and lighter load resistance exercises with similar movement patterns within an exercise session) is a form of training that may potentially bring about a state of postactivation potentiation, resulting in increased dynamic power (Pmax) and rate of force development during the lighter load exercise. Such a method may be more effective than either modality, independently for developing strength. The purpose of this research was to compare the effects of resistance training (RT), plyometric training (PT), and CT on lower body strength and anthropome

Reduced endplate currents underlie motor unit dysfunction in canine motor neuron disease.

Hereditary canine spinal muscular atrophy (HCSMA) is an autosomal dominant degenerative disorder of motor neurons. In homozygous animals, motor units produce decreased force output and fail during repetitive activity. Previous studies suggest that decreased efficacy of neuromuscular transmission underlies these abnormalities. To examine this, we recorded muscle fiber endplate currents (EPCs) and found reduced amplitudes and increased failures during nerve stimulation in homozygotes compared with wild-type controls. Comparison of EPC amplitudes with muscle fiber current thresholds indicate that

Anticipatory postural adjustments in stance and grip.

The reactive forces and torques associated with moving a hand-held object between two points are potentially destabilising, both for the object's position in the hand and for body posture. Previous work has demonstrated that there are increases in grip force ahead of arm motion that contribute to object stability in the hand. Other studies have shown that early postural adjustments in the legs and trunk minimise the potential perturbing effects on body posture of rapid voluntary arm movement. This paper documents the concurrent evolution of grip force and postural adjustments in anticipation o

Stretching and potentiation for performance optimization: effects on upper limbs in competitive swimmers.

This study examined the acute effects of combining short-duration static stretching with resistance-based activation, designed to induce post-activation performance enhancement (PAPE), on upper-limb muscle mechanics and neuromuscular performance. Fourteen competitive swimmers (7 male, 7 female) completed three randomized warm-up protocols: (1) general aerobic activity (CON), (2) Aerobic activity + 2 × 30 s of static stretching for latissimus dorsi and pectoralis major (SS), and (3) Aerobic activity + static stretching + 3 × 5 reps of resistance-band pull-overs (SS/PAPE). Role of the α Clamp in the Protein Translocation Mechanism of Anthrax Toxin.

Membrane-embedded molecular machines are utilized to move water-soluble proteins across these barriers. Anthrax toxin forms one such machine through the self-assembly of its three component proteins--protective antigen (PA), lethal factor, and edema factor. Upon endocytosis into host cells, acidification of the endosome induces PA to form a membrane-inserted channel, which unfolds lethal factor and edema factor and translocates them into the host cytosol. Translocation is driven by the proton motive force, composed of the chemical potential, the proton gradient (ΔpH), and the membrane potentia

Diminished contractile response to increased heart rate in intact human left ventricular hypertrophy. Systolic versus diastolic determinants.

BACKGROUND: Experimental studies indicate that in addition to diastolic dysfunction, hypertrophied myocardium can display depressed contractile responses, particularly at rapid heart rates, compounding reserve limitations. This study tests whether such abnormalities exist in intact human subjects at physiological paced rates and, if so, whether they are linked to simultaneous rate-dependent deterioration in diastolic function. METHODS AND RESULTS: Ten subjects with left ventricular hypertrophy (LVH) and 8 normal control subjects were studied. Most LVH patients presented with dyspnea and/or pu
