Teicoplanin, a recently introduced glycopeptide antibiotic, has been used, in combination with other antibiotics, to treat 31 episodes of septicaemia caused by Gram-positive organisms. Teicoplanin has double the activity of vancomycin against many Gram-positive bacteria, but allergic reactions and toxicity appear to be infrequent. A single daily dose is sufficient to maintain therapeutic levels, which is an advantage in conditions requiring long-term treatment. Of the 31 episodes treated, 16 were associated with infective endocarditis, 11 with Hickman catheter infection, two with bone and join…

In the present work, acidic (direct) and basic precipitation (indirect) methods were used to demonstrate the influence of the mode of precipitation on the structural properties of ZnO nanoparticles (NPs). Four samples of ZnO nanoparticles were prepared, two samples via each mode of precipitation. DZOa and IZOa were the aged samples prepared via acidic and basic precipitation methods, and DZOwa and IZOwa were processed without aging. Both precipitation processes were carried out without using any surfactant reagents. Zinc hydroxide precipitate, which was formed during the basic precipitation me

In the last years, the decreasing effectiveness of conventional antimicrobial-drugs has caused serious problems due to the rapid emergence of multidrug-resistant pathogens. This situation has brought attention to other antimicrobial agents like antimicrobial peptides (AMPs), for being considered an alternative to conventional drugs. These compounds target bacterial membranes for their activity, which gives them a broad spectrum of action and less probable resistance development. That is why the peptide-membrane interaction is a crucial aspect to consider in the study of AMPs. The aim of this w

Quorum-sensing inhibitors (QSIs) are being used increasingly in diverse fields, and are likely to end up in the environment, where they may encounter the antibiotics and consequently cause joint effects on biological systems. However, the potential joint effects of QSIs and antibiotics have received little attention. In this study, the joint effects of antibiotics, represented by sulfonamides (SAs) and penicillin, as well as three potential QSIs, were investigated using both Gram-negative (Escherichia coli, E. coli) and Gram-positive bacteria (Bacillus subtilis, B. subtilis). It was found that…

Antimicrobial resistance poses a significant threat to global health. Host defense peptides (HDPs) are being investigated as a strategy to mitigate this threat. Porcine β-defensins (pBDs) are a class of HDPs present in the swine immune system. Our group previously identified pBD‑5 as a component of the innate immune system of pigs and found that a synthetically produced cysteine-protected analogue modulated gene expression in swine gut cells in response to Brachyspira hyodysenteriae infection but lacked direct antimicrobial activity. In this study, we synthesized pBD‑5 with three disulfide bon

The global challenge of antibiotic resistance necessitates the introduction of more effective antibiotics. Here we report a potentially general design strategy, exemplified with vancomycin, that improves and expands antibiotic performance. Vancomycin is one of the most important antibiotics in use today for the treatment of Gram-positive infections. However, it fails to eradicate difficult-to-treat biofilm populations. Vancomycin is also ineffective in killing Gram-negative bacteria due to its inability to breach the outer membrane. Inspired by our seminal studies on cell penetrating guanidini

BACKGROUND: Gram-negative bacteria (GNB) are becoming increasingly resistant to a wide variety of antibiotics. There are currently limited treatments for GNB, and the combination of antibiotics with complementary mechanisms has been reported to be a feasible strategy for treating GNB infection. The inability to cross the GNB outer membrane (OM) is an important reason that a broad spectrum of Gram-positive only class of antibiotics (GPOAs) is lacking. Polymyxins may help GPOAs to permeate by disrupting OM of GNB.
OBJECTIVE: To identify what kind of GPOAs can be aided to broaden their anti-GNB

A series of copper-functionalized SBA-15 (Santa Barbara Amorphous) materials containing the ligands triethoxysilylpropylmaleamic acid (maleamic) or triethoxy-3-(2-imidazolin-1-yl)propylsilane (imidazoline) have been prepared. The nanostructured silica-based systems SBA-maleamic, SBA-imidazoline, SBA-maleamic-Cu and SBA-imidazoline-Cu were characterized by several methods observing that the functionalization took place mainly inside the pores of the mesoporous system. The antimicrobial behaviour of the synthesized materials against Staphylococcus aureus and Escherichia coli was tested observing

A 980nm laser-driven antimicrobial photothermal therapy using poly(vinylpyrrolidone) -coated Prussian Blue nanoparticles (PVP/PB NPs) is demonstrated. This approach allows an efficient eradication of a virulent strain of Gram-negative Escherichia coli (E. coli) associated with urinary tract infection as well as for the ablation of antibiotic resistant pathogens such as methicillin resistant Staphylococcus aureus (MRSA) and extended spectrum β-lactamase (ESBL) E. coli. Interestingly the 980nm irradiation exhibits minimal effect on mammalian cells up to a PVP/PB NPs concentration of 50μgmL(-1),

Two recombinant plasmids pPBT9 and pPBT74 carrying HindIII promoter-active DNA fragments of Bacillus thuringiensis onto the vector pGA24 were studied. The cloned fragment of pPBT9 plasmid has many homologous regions in Bac. thuringiensis genome. This fragment contains three Escherichia coli RNA polymerase binding sites, one of which is responsible for promoting tetracycline resistance of the promoter-deficient enterobacterial tet gene in E. coli cells. The bacillar fragment of pPBT74 plasmid has a single RNA polymerase binding site. Plasmids pPBT9 and pPBT74 were joined to the bacillar vectors

The aim of this study was to identify the bacteria causing neonatal septicemia in a neonatal intensive care unit (NICU) in León, Nicaragua and its relation with bacteria isolated from the environment at the NICU. Our data showed that 74% (34/46) of the bacteria related to newborns with septicemia were Gram-negative and highly resistant to beta-lactams (>85%) and aminoglycosides (80%), leading to treatment failure in 10 neonates with fatal outcome. Although, the prevalence of Gram-positive bacteria (26%) was lower than Gram-negative bacteria, Staphylococcus epidermidis was related to the death …

In the present study, the incidence and antimicrobial resistance patterns of the microorganisms that caused bloodstream infections (BSIs) in a medical-surgical intensive care unit during the years 2005-2007 were determined. The mean BSI incidence density was 6.56 per 1000 patient-days. The incidence density increased linearly during the study period (from 3.57 to 9.60 per 1000 patient-days). Staphylococcus aureus was most frequently isolated (47.3%), followed by Enterococcus spp. (10.8%) and Candida spp. (10.1%). There was a high rate of resistance to several of the prescribed antimicrobials a

Chronic infections are caused by microorganisms that display effective immune evasion mechanisms. Dendritic cell (DC)-dependent T cell-mediated adaptive immunity is one of the mechanisms that have evolved to prevent the occurrence of chronic bacterial infections. In turn, bacterial pathogens have developed strategies to evade immune recognition. In this study, we show that gram-negative and gram-positive bacteria differ in their ability to activate DCs and that gram-negative bacteria are far more effective inducers of DC maturation. Moreover, we observed that only gram-negative bacteria can in…
