Loss of Niemann Pick type C proteins 1 and 2 greatly enhances HIV infectivity and is associated with accumulation of HIV Gag and cholesterol in late endosomes/lysosomes.

BACKGROUND: Cholesterol pathways play an important role at multiple stages during the HIV-1 infection cycle. Here, we investigated the role of cholesterol trafficking in HIV-1 replication utilizing Niemann-Pick Type C disease (NPCD) cells as a model system. RESULTS: We used a unique NPC2-deficient cell line (NPCD55) that exhibited Gag accumulation as well as decreased NPC1 expression after HIV infection. Virus release efficiency from NPCD55 cells was similar to that from control cells. However, we observed a 3 to 4-fold enhancement in the infectivity of virus released from these cells. Fluore

HIV-1 RNA rectal shedding is reduced in men with low plasma HIV-1 RNA viral loads and is not enhanced by sexually transmitted bacterial infections of the rectum.

BACKGROUND: Among human immunodeficiency virus (HIV)-infected men who have sex with men (MSM) taking combination antiretroviral therapy (cART), the impact of rectal sexually transmitted infections (STIs) on rectal HIV-1 shedding is unknown. METHODS: Human immunodeficiency virus type 1 (HIV-1) RNA was quantified from rectal swabs collected for Neisseria gonorrhoeae (GC) and Chlamydia trachomatis (CT) screening of HIV-1-infected MSM. Correlations of STIs with rectal viral load were explored using multinomial regression modeling. HIV-1 coreceptor tropism was predicted from sequencing in a subset

ZNF598 enhances FAT10-mediated non-covalent stabilization of Tat to promote HIV-1 transcription and replication.

Human immunodeficiency virus type 1 (HIV-1) Tat is essential for efficient viral transcription and replication, and its stability is tightly controlled by host factors. In our previous study, we showed that ZNF598 stabilizes Tat and promotes HIV-1 replication through an E3 ligase activity-independent mechanism. Here, we identify FAT10 as a critical mediator of this effect. FAT10 knockout in HEK293T cells or FAT10 knockdown in Jurkat T-cell infection models markedly impaired the ability of ZNF598 to suppress Tat K48-linked ubiquitination, increase Tat abundance, and enhance HIV-1 replication. Enhanced screening and bacterial sexually-transmitted infection diagnoses after HIV pre-exposure prophylaxis initiation.

BACKGROUND: Recipients of HIV pre-exposure prophylaxis (PrEP) experience higher rates of gonorrhea and chlamydia diagnoses than non-recipients. However, it is unclear if these observations reflect a causal relationship between PrEP initiation and acquisition of sexually transmitted infections (i.e., behavioral "risk compensation") or a diagnostic bias related to PrEP recipients screening more frequently than non-recipients. METHODS: We conducted a self-controlled case series study comparing rates of gonorrhea and chlamydia diagnoses after versus before PrEP initiation among commercially-insur

Nanopore sequencing enhances the detection of low-frequency variants and mutational linkage in HIV-1 drug resistance.

OBJECTIVES: Low-frequency HIV-1 drug resistance mutations (DRMs) and mutation linkage within viral genomes may contribute to treatment failure and multidrug resistance but are difficult to resolve using conventional sequencing. We evaluated the performance of nanopore long-read sequencing for HIV-1 DRM detection and haplotype analysis. METHODS: In this retrospective study, 230 plasma samples from people living with HIV undergoing resistance testing at Beijing Ditan Hospital between Aug 2024 and July 2025 were analyzed using the nanopore-based G-seq500 platform. DRM profiles and subtype classi

Enhancing Antiretroviral Therapy Initiation for Hospitalized and Recently Discharged People Living With HIV in Johannesburg, South Africa.

BACKGROUND: Despite increased antiretroviral therapy (ART) access in South Africa, HIV testing and ART initiation are suboptimal in hospital settings. Key gaps include in-hospital case finding, ART initiation support, and primary health care (PHC) facility linkage after discharge. INTERVENTION DEVELOPMENT AND DESCRIPTION: We identified weaknesses in hospital processes by comparing them with PHC HIV services and developed a quality improvement model for implementation in 5 Johannesburg hospitals. We introduced dedicated teams of HIV testing counselors for structured case finding and ART-traine

Development of enhanced HIV-1 non-nucleoside reverse transcriptase inhibitors with improved resistance and pharmacokinetic profiles.

HIV-1 infection is a manageable chronic condition, with non-nucleoside HIV-1 reverse transcriptase inhibitors (NNRTIs) remaining a cornerstone of antiretroviral therapy. Nevertheless, drug resistance to existing therapeutics is a serious and immediate concern. Using structure-based and scaffold-hopping approaches, we designed evolved diarylpyrimidine analogs targeting reverse transcriptase (RT), exploiting chemical space surrounding the NNRTI-binding pocket. We identified compounds 5i3 and 5e2, with robust antiviral efficacy against wild-type HIV-1 and rilpivirine-resistant strains. Encouragin

The minority student open: Utilizing the informal mentoring approach to enhance racial representation in HIV care and research.

INTRODUCTION: The imbalance between those impacted by HIV and the HIV healthcare workforce may be one of the drivers of racial health disparities in the epidemic. To address this, the Academy Council for Racial Equity (ACRE), a committee of the American Academy of HIV Medicine, developed a program of engagement and informal mentorship for underrepresented racial, ethnic and gender minority (UREGM) students and trainees of the health professions. This pilot program was designed to increase interest in HIV related careers among these trainees. METHODS: The program was developed as a career path

Clathrin-nanoparticles deliver BDNF to hippocampus and enhance neurogenesis, synaptogenesis and cognition in HIV/neuroAIDS mouse model.

Brain derived neurotrophic factor (BDNF) promotes the growth, differentiation, maintenance and survival of neurons. These attributes make BDNF a potentially powerful therapeutic agent. However, its charge, instability in blood, and poor blood brain barrier (BBB) penetrability have impeded its development. Here, we show that engineered clathrin triskelia (CT) conjugated to BDNF (BDNF-CT) and delivered intranasally increased hippocampal BDNF concentrations 400-fold above that achieved previously with intranasal BDNF alone. We also show that BDNF-CT targeted Tropomyosin receptor kinase B (TrkB) a

Depot medroxyprogesterone acetate (DMPA) enhances susceptibility and increases the window of vulnerability to HIV-1 in humanized mice.

The progestin-based hormonal contraceptive Depot Medroxyprogesterone Acetate (DMPA) is widely used in sub-Saharan Africa, where HIV-1 is endemic. Meta-analyses have shown that women using DMPA are 40% more likely than women not using hormonal contraceptives to acquire Human Immunodeficiency Virus (HIV-1). Therefore understanding how DMPA increases susceptibility to HIV-1 is an important public health issue. Using C57BL/6 mice and our previously optimized humanized mouse model (NOD-Rag1tm1Mom Il2rgtm1Wjl transplanted with hCD34-enriched hematopoietic stem cells; Hu-mice) where peripheral blood

HIV-1 Matrix Trimerization-Impaired Mutants Are Rescued by Matrix Substitutions That Enhance Envelope Glycoprotein Incorporation.

The matrix (MA) domain of HIV-1 Gag plays key roles in virus assembly by targeting the Gag precursor to the plasma membrane and directing the incorporation of the viral envelope (Env) glycoprotein into virions. The latter function appears to be in part dependent on trimerization of the MA domain of Gag during assembly, as disruption of the MA trimer interface impairs Env incorporation. Conversely, many MA mutations that impair Env incorporation can be rescued by compensatory mutations in the trimer interface. In this study, we sought to investigate further the biological significance of MA tri

A novel multivalent DNA helix-based inhibitor showed enhanced anti-HIV-1 fusion activity.

DNA helix-based HIV-1 fusion inhibitors have been discovered as potent drug candidates, but further research is required to enhance their efficiency. The trimeric structure of the HIV-1 envelope glycoprotein provides a structural basis for multivalent drug design. In this work, a "multi-domain" strategy was adopted for design of an oligodeoxynucleotide with assembly, linkage, and activity domains. Built on the self-assembly of higher-order nucleic acid structure, a novel category of multivalent DNA helix-based HIV-1 fusion inhibitor could be easily obtained by a simple annealing course in solu

Blocking CXCL9 Decreases HIV-1 Replication and Enhances the Activity of Prophylactic Antiretrovirals in Human Cervical Tissues.

OBJECTIVES: The interferon-gamma-induced chemokine CXCL9 is expressed in a wide range of inflammatory conditions including those affecting the female genital tract. CXCL9 promotes immune cell recruitment, activation, and proliferation. The role of CXCL9 in modulating HIV-1 infection of cervicovaginal tissues, a main portal of viral entry, however, has not been established. We report a link between CXCL9 and HIV-1 replication in human cervical tissues and propose CXCL9 as a potential target to enhance the anti-HIV-1 activity of prophylactic antiretrovirals. DESIGN: Using ex vivo infection of h

Gag-CA Q110D mutation elicits TRIM5-independent enhancement of HIV-1mt replication in macaque cells.

HIV-1 is strictly adapted to humans, and cause disease-inducing persistent infection only in humans. We have generated a series of macaque-tropic HIV-1 (HIV-1mt) to establish non-human primate models for basic and clinical studies. HIV-1mt clones available to date grow poorly in macaque cells relative to SIVmac239. In this study, viral adaptive mutation in macaque cells, G114E in capsid (CA) helix 6 of HIV-1mt, that enhances viral replication was identified. Computer-assisted structural analysis predicted that another Q110D mutation in CA helix 6 would also increase viral growth potential. A n

Antiretroviral drug resistance mutations sustain or enhance CTL recognition of common HIV-1 Pol epitopes.

Antiretroviral drug resistance and escape from CTL are major obstacles to effective control of HIV replication. To investigate the possibility of combining drug and immune-based selective pressures against HIV, we studied the effects of antiretroviral drug resistance mutations on CTL recognition of five HIV-1 Pol epitopes presented by common HLA molecules. We found that these common drug resistance mutations sustain or even enhance the antigenicity and immunogenicity of HIV-1 Pol CTL epitopes. Variable patterns of cross-reactive and selective recognition of wild-type and corresponding variant

Enhancing access to HIV pre-exposure prophylaxis (PrEP) through a community-based project in New Zealand.

INTRODUCTION: New Zealand has maintained a low HIV prevalence through comprehensive screening, treatment, and prevention programmes. However, challenges remain with access to pre-exposure prophylaxis (PrEP) for at-risk populations, particularly men who have sex with men (MSM). AIM: This study aimed to evaluate the implementation and outcomes of a community-based project that expanded PrEP access via a Standing Order authorising peer HIV testers and pharmacists to initiate short-term PrEP prior to general practitioner follow-up. METHODS: Conducted between May 2020 and July 2021, the project i

JQKD82 Inhibits HIV Infection of Macrophages Through Blocking Viral Entry and Enhancing Antiviral ISG Responses.

JQKD82 is an epigenetic modulator that inhibits lysine-specific demethylase 5 (KDM5), a host factor implicated in HIV latency and cell survival. Although JQKD82 has been studied in latent HIV infection of T cell models, its effects on HIV infection in macrophages remain unclear. Here, we investigated the impact of JQKD82 on HIV infection of primary human monocyte-derived macrophages (MDMs). We observed that treatment of MDMs with non-cytotoxic concentrations of JQKD82 dose-dependently inhibited HIV replication, as evidenced by reduced virus-induced syncytium formation, decreased viral Gag mRNA
