Effects of environmental factors on host-parasite interaction patterns in backyard-tethered goats of Kerala, India.

The environment is the most important stratum in the epidemiological triad of a parasitic disease and any variations in the environmental factors may decide the dynamic occurrence and existence of different lifecycle stages of these parasites. The present study investigated the correlations between key biometeorological and demographical parameters with the incidence of different gastrointestinal parasites and hemoparasites among goats. Four hundred and thirty-two goats were screened for parasitic infection in a yearlong survey conducted from July 2022 to June 2023 in the Teaching Veterinary C

Non-Vesicular Lipid Transport Machinery in Leishmania donovani: Functional Implications in Host-Parasite Interaction.

Eukaryotic cells have distinct membrane-enclosed organelles, each with a unique biochemical signature and specialized function. The unique identity of each organelle is greatly governed by the asymmetric distribution and regulated intracellular movement of two important biomolecules, lipids, and proteins. Non-vesicular lipid transport mediated by lipid-transfer proteins (LTPs) plays essential roles in intra-cellular lipid trafficking and cellular lipid homeostasis, while vesicular transport regulates protein trafficking. A comparative analysis of non-vesicular lipid transport machinery in prot

Recent Advances in Chemotherapeutics for Leishmaniasis: Importance of the Cellular Biochemistry of the Parasite and Its Molecular Interaction with the Host.

Leishmaniasis, a category 1 neglected protozoan disease caused by a kinetoplastid pathogen called Leishmania, is transmitted through dipteran insect vectors (phlebotomine, sand flies) in three main clinical forms: fatal visceral leishmaniasis, self-healing cutaneous leishmaniasis, and mucocutaneous leishmaniasis. Generic pentavalent antimonials have long been the drug of choice against leishmaniasis; however, their success is plagued with limitations such as drug resistance and severe side effects, which makes them redundant as frontline therapy for endemic visceral leishmaniasis. Alternative

Innate immune cell response to host-parasite interaction in a human intestinal tissue microphysiological system.

Protozoan parasites that infect humans are widespread and lead to varied clinical manifestations, including life-threatening illnesses in immunocompromised individuals. Animal models have provided insight into innate immunity against parasitic infections; however, species-specific differences and complexity of innate immune responses make translation to humans challenging. Thus, there is a need for in vitro systems that can elucidate mechanisms of immune control and parasite dissemination. We have developed a human microphysiological system of intestinal tissue to evaluate parasite-immune-spec

Non-coding RNAs in Host-Pathogen Interactions: Subversion of Mammalian Cell Functions by Protozoan Parasites.

Pathogens have evolved mechanisms to modulate host cell functions and avoid recognition and destruction by the host damage response. For many years, researchers have focused on proteins as the main effectors used by pathogens to hijack host cell pathways, but only recently with the development of deep RNA sequencing these molecules were brought to light as key players in infectious diseases. Protozoan parasites such as those from the genera Plasmodium, Toxoplasma, Leishmania, and Trypanosoma cause life-threatening diseases and are responsible for 1000s of deaths worldwide every year. Some of t

A synthetic workflow for coordinated direct observation and genetic tagging applied to a complex host-parasite interaction.

An important aspect influencing host specificity is a parasite's compatibility, or ability, to infect a potential host. Here, we examine the compatibility between different trematode genotypes of the same species and several host species. To execute this study, we developed a synthetic workflow which combines the use of a fluorescent dye and standard molecular techniques to study host-parasite interactions and host specificity. The utility of the fluorescent dye, BIODIPY FL C₁₂, was evaluated to label and track larval trematodes during experimental infections using the Cerithidea californica-t…

Covariance in species diversity and facilitation among non-interactive parasite taxa: all against the host.

Different parasite taxa exploit different host resources and are often unlikely to interact directly. It is unclear, however, whether the diversity of any given parasite taxon is indirectly influenced by that of other parasite taxa on the same host. Some components of host immune defences may operate simultaneously against all kinds of parasites, whereas investment by the host in specific defences against one type of parasite may come at the expense of defence against other parasites. We investigated the relationships between the species diversity of 4 higher taxa of ectoparasites (fleas, suck

Freeze fracture studies on the interaction between the malaria parasite and the host erythrocyte in Plasmodium knowlesi infections.

The freeze fracture technique has been used to study the internal cyto-architecture of the surface membranes of the parasite and erythrocyte in Plasmodium knowlesi infections. Six fracture faces, derived from the plasma membrane and 2 pellicular membranes, have been identified at the surface of the free merozoite. The apposed leaflets of the 2 pellicular membranes show the characteristic features of E fracture faces, a result compatible with the view that the pellicular membranes line a potential cisterna. There is evidence to suggest that there may be changes in the distribution and density o

Imaging host cell-Leishmania interaction dynamics implicates parasite motility, lysosome recruitment, and host cell wounding in the infection process.

Leishmania donovani causes human visceral leishmaniasis. The parasite infectious cycle comprises extracellular flagellated promastigotes that proliferate inside the insect vector, and intracellular nonmotile amastigotes that multiply within infected host cells. Using primary macrophages infected with virulent metacyclic promastigotes and high spatiotemporal resolution microscopy, we dissect the dynamics of the early infection process. We find that motile promastigotes enter macrophages in a polarized manner through their flagellar tip and are engulfed into host lysosomal compartments. Persiste

Metacercariae of Posthodiplostomum Dubois, 1936 (Diplostomidae) infecting freshwater fish worldwide: infection patterns, host-parasite interactions, distribution range, and disease control.

This study aimed to investigate the infection patterns, host-parasite interactions, distribution ranges, and control diseases by Posthodiplostomum metacercariae in freshwater fish on a global scale. We analyzed parasitological data from 203 freshwater fish species across 17 orders and 44 families, predominantly representing the Leuciscidae and Cichlidae families. The analysis identified a total of 10 Posthodiplostomum metacercariae species, although several studies did not identify the parasites to the species level. In host fish, prevalence, intensity and abundance levels of Posthodiplostomum…

Protocol for efficient germination of Castilleja (Orobanchaceae) for chemical haustorium induction and parasite-host interaction studies.

We present a protocol for germinating Castilleja seeds and growing seedlings for hormone treatments and plant-plant interaction assays. We describe steps for seed sterilization, stratification, and transplantation, ensuring germination and seedling development. We detail procedures for haustorium formation, including strigolactone treatment and co-inoculation to induce root interactions with a host plant. This protocol includes optimized nutrient conditions and microscopy techniques to ensure experimental reproducibility. This approach enables reliable investigation of haustorium formation in

Interaction between Intestinal Parasites and the Gut Microbiota: Implications for the Intestinal Immune Response and Host Defence.

Intestinal parasites, including helminths and protozoa, account for a significant portion of the global health burden. The gastrointestinal (GI) tract not only serves as the stage for these parasitic infections but also as the residence for millions of microbes. As the intricacies of the GI microbial milieu continue to unfold, it is becoming increasingly apparent that the interactions between host, parasite, and resident microbes help dictate parasite survival and, ultimately, disease outcomes. Across both clinical and experimental models, intestinal parasites have been shown to impact microbi

Dual single-cell and bulk RNA sequencing reveal transcriptional profiles underlying heterogenous host-parasite interactions in human peripheral blood mononuclear cells.

Toxoplasma gondii, a zoonotic apicomplexan that infects over a billion people worldwide, can cause early death in immunocompromised individuals and defects in foetal brain development. Toxoplasma is also a major cause of abortion in small ruminants. When Toxoplasma encounters host cells, several outcomes are possible. For example, the parasite can enter the host cell or can inject its effector proteins into the cell without entering. These heterogenous outcomes occur simultaneously in the same host and likely determine disease pathogenesis. Yet, current knowledge of host-Toxoplasma interaction

Integrative RNA-Seq analysis of host-parasite interactions and microbiota shifts in the gills of two fish species infected with monogeneans.

Monogenean infections represent a significant threat to aquaculture by compromising fish health. These parasites can cause severe histological damage and increase mortality rates. The white snook (Centropomus viridis) and the gilthead seabream (Sparus aurata) are two commercially important fish species that become susceptible to diseases when infected with monogeneans. Understanding the molecular mechanisms underlying host responses to infection is essential for developing effective disease management strategies. In this study, we performed an integrative RNA-Seq analysis using transcriptomic

Live Imaging of Host-Parasite Interactions in a Zebrafish Infection Model Reveals Cryptococcal Determinants of Virulence and Central Nervous System Invasion.

UNLABELLED: The human fungal pathogen Cryptococcus neoformans is capable of infecting a broad range of hosts, from invertebrates like amoebas and nematodes to standard vertebrate models such as mice and rabbits. Here we have taken advantage of a zebrafish model to investigate host-pathogen interactions of Cryptococcus with the zebrafish innate immune system, which shares a highly conserved framework with that of mammals. Through live-imaging observations and genetic knockdown, we establish that macrophages are the primary immune cells responsible for responding to and containing acute cryptoco

Seasonal modulation of immune, hemato-biochemical, and antioxidant responses in Nile tilapia during ectoparasitic infection: insights into host-parasite-environment interactions.

Limited information is available on how seasonal changes affect host immunity, hemato-biochemical status, antioxidant responses, and parasite infection in cultured fish. Therefore, this study evaluated the associations among seasonal changes, parasitic infections (Cichlidogyrus tilapiae and Centrocestus formosanus), and hemato-biochemical, antioxidant, and immune responses in Nile tilapia (Oreochromis niloticus). Fish (n = 593) and water samples were collected seasonally from Abbasa Fish Farm, El-Sharkia Governorate, Egypt. Parasite prevalence was highest in summer and lowest in winter. Infect

First report of Hexamita nelsoni in blue mussel (Mytilus edulis): morphology, phylogeny, and host-parasite interaction.

Bivalve diseases caused by protozoan parasitic infection are monitored by coordinated surveillance programs across European member countries. However, these routine monitoring of bivalve health only survey a few relatively well-known parasites, which may leave undetected a range of less known opportunistic protozoan agents. Under increased stress, notably due to shifting environmental conditions affecting water quality and nutrient access, these opportunistic parasites may turn into pathogens impacting reproduction or growth and therefore decreasing the success and future development of aquacu

Trichomonas vaginalis adhesion protein 65 (TvAP65) modulates parasite pathogenicity by interacting with host cell proteins.

Trichomonas vaginalis (T. vaginalis) is a widespread and important sexually transmitted pathogen. Adherence to the surface of the host cell is the precondition forthis parasite's parasitism and pathogenicity. Adhesion protein 65 (TvAP65) plays a key role in the process of adhesion. However, how TvAP65 mediates the adhesion and pathogenicity of T. vaginalis to host cellsis unclear. In this study, we knocked down the expression of TvAP65 in trophozoites by small RNA interference. The number of T. vaginalis trophozoites adhering to VK2/E6E7 cells was decreased significantly, and the inhibition of

An overview on Leishmania (Mundinia) enriettii: biology, immunopathology, LRV and extracellular vesicles during the host-parasite interaction.

One of the Leishmania species known to be non-infective to humans is Leishmania (Mundinia) enriettii whose vertebrate host is the guinea pig Cavia porcellus. It is a good model for cutaneous leishmaniasis, chemotherapeutic and molecular studies. In the last years, an increased interest has emerged concerning the L. (Mundinia) subgenus after the finding of Leishmania (M.) macropodum in Australia and with the description of other new/putative species such as L. (M.) martiniquensis and 'L. (M.) siamensis'. This review focused on histopathology, glycoconjugates and innate immunity. The presence of
