G119S ace-1 mutation conferring insecticide resistance detected in the Culex pipiens complex in Morocco.

BACKGROUND: Arboviruses are controlled through insecticide control of their mosquito vector. However, inconsiderate use of insecticides often results in the selection of resistance in treated populations, so that monitoring is required to optimize their usage. Here, Culex pipiens (West Nile and Rift Valley Fever virus vector) specimens were collected from four Moroccan cities. Levels of susceptibility to the organophosphate (OP) insecticide malathion were assessed using World Health Organization (WHO)-recommended bioassays. Individual mosquitoes were tested for the presence of the G119S mutati

malERA: An updated research agenda for insecticide and drug resistance in malaria elimination and eradication.

Resistance to first-line treatments for Plasmodium falciparum malaria and the insecticides used for Anopheles vector control are threatening malaria elimination efforts. Suboptimal responses to drugs and insecticides are both spreading geographically and emerging independently and are being seen at increasing intensities. Whilst resistance is unavoidable, its effects can be mitigated through resistance management practices, such as exposing the parasite or vector to more than one selective agent. Resistance contributed to the failure of the 20th century Global Malaria Eradication Programme, an

Insecticide resistance in the field populations of the Asian tiger mosquito Aedes albopictus in Beijing: resistance status and associated detoxification genes.

BACKGROUND: Aedes (Stegomyia) albopictus (Skuse) is an invasive and widespread mosquito species that can transmit dengue, chikungunya, yellow fever, and Zika viruses. Its control heavily relies on the use of insecticides. However, the efficacy of the insecticide-based intervention is threatened by the increasing development of resistance to available insecticides. Understanding the current status and potential mechanisms of insecticide resistance is an important prerequisite for devising strategies to maintain the sustainability of vector control programs. In this study, we investigated the cu…

Resistance to commonly used insecticides and underlying mechanisms of resistance in Aedes aegypti (L.) from Sri Lanka.

BACKGROUND: Drastic increases of dengue fever (DF) over the past few years have prompted studies on the development of resistance to insecticides in the mosquito vector, Aedes aegypti (Linnaeus). In Sri Lanka control of the vector population is essentially achieved using larvicides (temephos) and adulticides (principally pyrethroids). The present study investigates resistance to commonly used insecticides and underlying mechanisms of Ae. aegypti in selected sites in Sri Lanka. METHODS: In this study, susceptibility to three commonly used adulticides (malathion, permethrin and deltamethrin) an

Implications of insecticide resistance for malaria vector control with long-lasting insecticidal nets: evidence from health facility data from Benin.

BACKGROUND: Insecticide-based interventions have averted more than 500 million malaria cases since 2000, but insecticide resistance in mosquitoes could bring about a rebound in disease and mortality. This study investigated whether insecticide resistance was associated with increased incidence of clinical malaria. METHODS: In an area of southern Benin with insecticide resistance and high use of insecticide-treated nets (ITNs), malaria morbidity and insecticide resistance were measured simultaneously in 30 clusters (villages or collections of villages) multiple times over the course of 2 years…

Molecular monitoring reveals the presence of multiple insecticide resistance markers in mosquito disease vectors in Türkiye.

Insecticide resistance in mosquito vectors often involves multiple molecular mechanisms. Here, we analysed resistance-associated mechanisms in Culex pipiens and Aedes albopictus populations from 14 locations across Türkiye using a multi-marker molecular approach. We combined species identification, biotype resolution within the Cx. pipiens complex, and molecular screening of key target-site resistance markers (kdr, CHS1, and ace-1), together with genes associated with metabolic detoxification. Within Cx. pipiens, resistance-associated marker profiles varied among populations with heterogeneous…

Malaria transmission dynamics, vector diversity, and insecticide resistance profiles in two health districts of southern Benin.

BACKGROUND: Malaria transmission in Benin is heterogeneous, influenced by ecological settings and environmental changes affecting Plasmodium spp. prevalence, vector species composition, and insecticide resistance. Comprehensive local data on transmission dynamics remain limited, particularly in southern Benin. This study assessed malaria transmission, vector diversity, and insecticide resistance in two health districts to support the National Malaria Control Programme in designing targeted and effective vector control strategies. METHODS: A repeated cross-sectional study was conducted in two …

Evaluation of Insecticide Resistance and Associated Enzyme Activity in Field Populations of Diamondback Moth (Plutella xylostella).

The diamondback moth, Plutella xylostella (L.), is a major pest of cruciferous crops with a remarkable capacity to develop insecticide resistance. The present study evaluated the susceptibility, resistance ratios, and detoxification mechanisms of field populations collected from Andhra Pradesh, India, against tolfenpyrad 15 EC and emamectin benzoate 5 SG. Laboratory selection studies revealed declining LC50 values over successive generations reared without insecticide pressure, indicating partial recovery of susceptibility. Field bioassays showed resistance ratios ranging from 9.8 to 13.1 for …

Challenges in insecticide resistance management: Implications of alternating selection in the yellow fever mosquito, Aedes aegypti.

Insecticide resistance in Aedes aegypti is driven by multiple mechanisms, yet the dynamics of their development under defined selection regimes remain unclear. Here, we established laboratory selection lines from a field-collected parental strain (AeStA) using permethrin, malathion, or alternating exposures to these two insecticides across successive generations. Continuous selection with permethrin rapidly produced extremely high resistance (resistance ratio, RR = 2700) within five generations, whereas malathion selection yielded only low-level tolerance (RR ≈ 3). Alternating permethrin and m…

Genetic markers associated with the widespread insecticide resistance in malaria vector Anopheles funestus populations across Tanzania.

BACKGROUND: Anopheles funestus is a leading vector of malaria in most parts of East and Southern Africa, yet its ecology and responses to vector control remain poorly understood compared with other vectors such as Anopheles gambiae and Anopheles arabiensis. This study presents the first large-scale survey of the genetic and phenotypic expression of insecticide resistance in An. funestus populations in Tanzania. METHODS: We performed insecticide susceptibility bioassays on An. funestus mosquitoes in nine regions with moderate-to-high malaria prevalence in Tanzania, followed by genotyping for r…

Insecticide resistance and characteristics of mutations related to target site insensitivity of diamondback moths in Taiwan.

Diamondback moth (DBM, Plutella xylostella) is the most significant pest of cruciferous vegetables as they rapidly develop high-level resistance to many insecticides. Monitoring DBM susceptibility and target-site mutation frequency is essential for pest control. In this study, 10 insecticides were tested on 11 field populations. Frequencies of target-site mutations (including para, ace1, Rdl1, RyR1, and nAChRα6 genes) were estimated by pyrosequencing. Insecticides registered after 2007 for DBM control in Taiwan, i.e., spinetoram, chlorantraniliprole, chlorfenapyr, metaflumizone, and flubendiam

Experimental evolution supports the potential of neonicotinoid-pyrethroid combination for managing insecticide resistance in malaria vectors.

The introduction of neonicotinoids for managing insecticide resistance in mosquitoes is of high interest as they interact with a biochemical target not previously used in public health. In this concern, Bayer developed a combination of the neonicotinoid clothianidin and the pyrethroid deltamethrin (brand name Fludora Fusion) as a new vector control tool. Although this combination proved to be efficient against pyrethroid-resistant mosquitoes, its ability to prevent the selection of pyrethroid and neonicotinoid resistance alleles was not investigated. In this context, the objective of this work…

Chlorfenapyr: irritant effect compared to other insecticides and its intrinsic toxicity in multiple-insecticide-susceptible and -resistant Anopheles stephensi (Diptera: Culicidae).

BACKGROUND & OBJECTIVES: For effective management of vector resistance there is a need for new insecticide molecules with novel modes of action. For desired toxic effect of an insecticide, apart from other behavioural aspects, toxicity and chemical nature of the molecule are important that may cause irritability in the mosquito to the insecticide affecting the uptake. In this study, a pyrrole class insecticide, chlorfenapyr (a late acting insecticide) was tested for its irritability against multiple-insecticide-susceptible and -resistant strains of Anopheles stephensi Liston 1901 (Diptera: Cul

Resistance to insecticides in insect vectors of disease: est alpha 3, a novel amplified esterase associated with amplified est beta 1 from insecticide resistant strains of the mosquito Culex quinquesfasciatus.

Vector control programmes in many countries face the dual problems of parasite drug resistance and insecticide resistance in the insect vectors of the disease. Here we report for the first time a new esterase-based insecticide resistance mechanism in the filariasis vector Culex quinquefasciatus. The field collected COL strain of C. quinquefasciatus from Columbia was heterogeneous for organophosphorus insecticide resistance. On native polyacrylamide gels it had an elevated beta-naphthyl acetate specific esterase with the same Rf as that for the Est beta 1s involved in insecticide resistance in …

Integrated transcriptome-microbiome analysis reveals a host-microbe interplay associated with insecticide resistance in Aedes albopictus.

INTRODUCTION: Aedes albopictus is the primary vector of major arboviral diseases such as dengue fever, chikungunya fever, and Zika virus disease, and its control is highly dependent on chemical insecticides. However, the long-term use of pyrethroid insecticides has led to the development of insecticide resistance in Ae. albopictus, which severely undermines the efficacy of vector control programs. METHODS: Ae. albopictus populations were collected from five sites in Guangdong and Hainan provinces, China. Beta-cypermethrin resistance levels were determined via bioassays, with resistance ratios…

Enzymatic mechanisms of insecticide resistance in Anopheles sp mosquitoes: Implications for malaria vector control in Indonesia.

Malaria remains a major public health concern worldwide, with Anopheles mosquitoes serving as its primary vectors. The widespread use of synthetic insecticides in malaria control programs has resulted in the development of insecticide resistance in these mosquitoes, endangering control efforts. This study investigates metabolic resistance mechanisms in wild-caught Anopheles spp. from three regencies in Indonesia (Pesawaran, Pangandaran, and Purworejo), with an emphasis on enzymatic activity and ace-1 gene mutations. Anopheles mosquitoes were collected using human landing capture (HLC) and cow-…
