On the control of arginine metabolism in chicken kidney and liver.

Arginases have been found to be located on the external side of the inner mitochondrial membrane of chicken kidney and liver. Transamidinase has been detected within the liver mitochondrial matrix space. Arginases and transamidinase act upon two different intracellular arginine pools. Penetration of arginine into the matrix space occurs only in respring mitochondria and in the presence of anions such as acetate and phosphate; D-arginine, L-ornithine, D-'ornithine and L-lysine penetrate with the same modalities. L-Histidine penetrates only kidney mitochondria. Because of transamidinase compartm

Exofacial phospholipids at the plasma membrane: ill-defined targets for early infection processes.

The eukaryotic plasma membrane (PM) consists largely of phospholipids and proteins, and separates the intracellular compartments from the extracellular space. It also serves as a signaling platform for cell-to-cell communication and an interaction platform for the molecular crosstalk between pathogens and their target cells. Much research has been done to elucidate the interactions between pathogens and host membrane proteins. However, little is known about the interactions between pathogens and membrane phospholipids, although reports have described a contribution of phospholipids to cell rec

Host-parasite relationship of Trypanosoma corvi in Ornithomyia avicularia.

The first description of an electron microscopic study of Trypanosoma corvi in the vector Ornithomyia avicularia is reported. There is a close association between vector and parasite in the midgut, ileum and rectum of the vector. The midgut distribution of parasites is determined by the peritrophic membrane, which confines the early infection to the endoperitrophic space. Parasites escape from the ruptured region of the peritrophic membrane at the pylorus to gain access to the ectoperitrophic space, where intense multiplication occurs. The resulting, smaller epimastigotes attach to the cuticle

Comparative ribosome profiling uncovers a dominant role for translational control in Toxoplasma gondii.

BACKGROUND: The lytic cycle of the protozoan parasite Toxoplasma gondii, which involves a brief sojourn in the extracellular space, is characterized by defined transcriptional profiles. For an obligate intracellular parasite that is shielded from the cytosolic host immune factors by a parasitophorous vacuole, the brief entry into the extracellular space is likely to exert enormous stress. Due to its role in cellular stress response, we hypothesize that translational control plays an important role in regulating gene expression in Toxoplasma during the lytic cycle. Unlike transcriptional profil

Sodium-dependent carnitine transport in human placental choriocarcinoma cells.

The JAR human placental choriocarcinoma cells were found to transport carnitine into the intracellular space by a Na(+)-dependent process. The transport showed no requirement for anions. The Na+-dependent process was saturable and the apparent Michaelis-Menten constant for carnitine was 12.3 +/- 0.5 microM. Na+ activated the transport by increasing the affinity of the transport system for carnitine. The transport system specifically interacted with L-carnitine, D-carnitine, acetyl-DL-carnitine and betaine. 6-N-Trimethyllysine and choline had little or no effect on carnitine transport. Of the t…

[Ultramicroscopic studies on the human corpus luteum menstruationis and graviditatis].

In high active steroid producing cells of the human corpus luteum we can find in the cytoplasm a well developed smooth endoplasmic reticulum in form of smooth tubules and homogenously spread vesicles resulting from an active metabolic process. We also find specific concertinalike folded membrane complexes which too belong to the endoplasmic reticulum. Numerous mainly tubular mitochondria contain dense lipoid inclusions. Their close conection with the endoplasmic vesicles is expression of a direct interaction of both cell organelles in progesterone production. The transport of the intracellular

Intracellular Toxic AGEs (TAGE) Triggers Numerous Types of Cell Damage.

The habitual intake of large amounts of sugar, which has been implicated in the onset/progression of lifestyle-related diseases (LSRD), induces the excessive production of glyceraldehyde (GA), an intermediate of sugar metabolism, in neuronal cells, hepatocytes, and cardiomyocytes. Reactions between GA and intracellular proteins produce toxic advanced glycation end-products (toxic AGEs, TAGE), the accumulation of which contributes to various diseases, such as Alzheimer's disease, non-alcoholic steatohepatitis, and cardiovascular disease. The cellular leakage of TAGE affects the surrounding cell

A contribution to the pathogenesis of otosclerosis.

In a histochemical and electron microscopical study the pathogenesis of otosclerosis was investigated. The morphological changes in the extra- and intracellular space indicated a complex metabolic disturbance of all tissue components. The observed chondrocytic chondrolysis stressed the role of the cartilage remnants in the otic capsule as an etiological factor. There was morphological evidence of a superimposed enzymatic defect.

[Banded filamentous associates in the intra- and extracellular space in connection with collagen degradation (author's transl)].

Banded fibrous associates are described in the extracellular space of connective tissue from human endometrium, Ehlers-Danlos syndrome and of tendon rupture. In the cases of morbus Dupuytren these associates are also found as intracellular inclusions. The banded structures are interpreted as states of an enzymatically induced degradation of collagen in correlation with Type-III collagen.

Identification of proteins interacting with human SP110 during the process of viral infections.

Human SP110 plays an important role in resisting intracellular pathogens, and hence has become an important drug target for preventing intracellular pathogen diseases, such as tuberculosis, hepatic veno-occlusive disease, and intracellular cancers. Unfortunately, so far little is known about the interactions of SP110 with the other proteins in a cell, which is considered to be the key for revealing its action mechanism and mediated pathway. Using both the genetic and structural analyses as well as the segment-docking approach, we have identified two proteins: the human remodeling and spacing f

[Neuro-hormonal regulation of the function of the enterocyte].

The intestinal epithelial cell, or enterocyte, performs a dual function: absorption and secretion. Absorption takes place in the villi and secretion in the crypts (and perhaps also in the villi). Both functions are controlled by complex extra- and intracellular systems. The extracellular system is mediated by neuromediators released in the subcellular space. Some mediators, like the vasoactive intestinal peptide (VIP) or the prostaglandins, stimulate secretion while others, like enkephalins, stimulate absorption. All these mediators act on intracellular metabolic processes by activating "secon

Alcohol causes alveolar epithelial oxidative stress by inhibiting the nuclear factor (erythroid-derived 2)-like 2-antioxidant response element signaling pathway.

Excessive alcohol use increases the risk of acute lung injury and pneumonia. Chronic alcohol ingestion causes oxidative stress within the alveolar space, including near depletion of glutathione (GSH), which impairs alveolar epithelial and macrophage function, in experimental animals and human subjects. However, the fundamental mechanism(s) by which alcohol induces such profound lung oxidative stress is unknown. Nuclear factor (erythroid-derived 2)-like 2 (Nrf2) is a redox-sensitive master transcription factor that regulates activation of the antioxidant response element (ARE). As the alveolar

Millisecond-Resolved Electronic pH Microdomains Synchronize Calcium Amplification in Cell Clusters.

A central goal for bioelectronic interfaces is to convert electrical inputs into local chemical commands that regulate cell signaling with defined space and time. However, imposing spatially confined extracellular pH (pHo) cues across cell populations with externally programmed millisecond timing remains difficult. Here, we show that an electronically generated pHo microdomain can act as a timed biochemical command that synchronizes intracellular Ca2+ amplification in HL-1 cardiomyocyte clusters. Using a microfabricated electrochemical interface, electronic stimulation produced a spatially con

[Intracellular calcium: physiology and physiopathology].

Many important aspects of our life are regulated by the free cytosolic Ca2+ concentration. The intracellular Ca2+ signal is regulated both in space, frequency and amplitude. Each cell chooses a unique set of Ca2+ signals to control its function. Ca2+ signal transduction is based on rises in free cytosolic Ca2+ concentration. Ca2+ can come from the extracellular space or be released from intracellular stores. Extracellular Ca2+ enters the cell through various types of plasma-membrane Ca2+ channels and leaves the cell using Ca2+ pumps and Na+/Ca(2+)-exchangers. Ca2+ is accumulated in intracellul

[Pulmonary infection induced by Mycobacterium intracellulare in 2 sisters: a biomolecular study of the isolates].

Patient 1 was an 82-year-old woman without symptoms who had been admitted to the hospital because of abnormal shadows on a chest X ray film. The chest X ray showed a round opacity in the right middle lung field and air-space consolidation in the right lower lung field. A computed tomographic (CT) scan showed a nodular opacity with satellite lesions in the right middle lobe and bronchiectasis in the right middle lobe and left lingular segment. Bronchial lavage yielded Mycobacterium intracellulare. Patient 2 was the 77-year-old sister of Patient 1. She had productive cough and fatigue, and chest

On the physiological response of the cerebral cortex to acute stress (reversible asphyxia).

1. Rhesus monkey (Macaca mulatta) foetuses were delivered by Caesarean section 3-10 days before term. Aortic blood and cerebrospinal fluid (c.s.f.) samples were taken, the latter from the cortical subarachnoid space and the cisterna magna. The umbilical cord was clamped and foetal breathing prevented for 14-17 min. Blood and c.s.f. were sampled further during this total asphyxiation and for up to 24 hr thereafter.2. The [K(+)] in the cortical subarachnoid fluid started to rise within 2-3 min after the onset of asphyxia and increased up to 7 times the normal level. The [K(+)] of blood plasma an

Lysosomal dysfunction in neurodegenerative disease.

Examination of genetic risk factors associated with neurodegenerative diseases has provided important mechanistic insights into the pathophysiology of these disorders, and it has implicated defects in lysosomal function as a key component of the neurodegenerative process. The lysosome has a primary role in mediating degradation of both intracellular contents and endocytosed material from the extracellular space. Aggregation of misfolded proteins is a pathological mechanism that is observed across neurodegenerative disorders, and aberrant lysosomal degradation of such proteins plays a pivotal p

Deranged hepatocyte intracellular Ca2+ homeostasis and the progression of non-alcoholic fatty liver disease to hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) is the second leading cause of cancer-related deaths in men, and the sixth in women. Non-alcoholic fatty liver disease (NAFLD) is now one of the major risk factors for HCC. NAFLD, which involves the accumulation of excess lipid in cytoplasmic lipid droplets in hepatocytes, can progress to non-alcoholic steatosis, fibrosis, and HCC. Changes in intracellular Ca2+ constitute important signaling pathways for the regulation of lipid and carbohydrate metabolism in normal hepatocytes. Recent studies of steatotic hepatocytes have identified lipid-induced changes in intra
