The Parkinson's disease drug entacapone disrupts gut microbiome homoeostasis via iron sequestration.

Many human-targeted drugs alter the gut microbiome, leading to implications for host health. However, the mechanisms underlying these effects are not well known. Here we combined quantitative microbiome profiling, long-read metagenomics, stable isotope probing and single-cell chemical imaging to investigate the impact of two widely prescribed drugs on the gut microbiome. Physiologically relevant concentrations of entacapone, a treatment for Parkinson's disease, or loxapine succinate, used to treat schizophrenia, were incubated ex vivo with human faecal samples. Both drugs significantly impact

Long-term effects of soluble and insoluble ferric irons on anaerobic oxidation of methane in paddy soil.

Iron-dependent anaerobic oxidation of methane (Fe-AOM) is an important process to reduce methane emissions into the atmosphere. It is well known that iron bioavailability largely influences microbial iron reduction, but the long-term effects of different ferric irons on soil Fe-AOM remain unknown. In this work, paddy soil in the ferruginous zone was collected and inoculated with insoluble ferrihydrite and soluble EDTA-Fe(III) for 420 days. Stable isotope experiments, activity inhibition tests, and molecular biological techniques were performed to reveal the activity, microbial community, and p

Minerals and trace elements in infant nutrition.

Little information is available on absorption and metabolism of minerals and trace elements during infancy. The lack of data is related to the methodological problems involved in these studies. By using stable isotopes as labels, studies can be conducted in infants without introducing exposure to radiation, or any other risk, and studies on bioavailability of minerals and trace elements during early life can therefore be performed. This paper discusses results from studies of trace element/mineral absorption and metabolism in infants, based on stable isotope techniques.

Effects of dietary factors on the pharmacokinetics of 58Fe-labeled hemin after oral administration in normal rats and the iron-deficient rats.

Hemin, iron (III) protoporphyrin chloride (IX), as a stable form of heme iron, has been used in iron absorption studies. The aim of the present study was to elucidate the influences of body iron status and three dietary factors (green tea extract, ascorbic acid, and calcium) on the pharmacokinetics of hemin using stable isotope labeling methods followed by ICP-MS measurement. In this study, a rapid, sensitive, and specific ICP-MS method for the determination of (58)Fe originating from hemin in rat plasma was developed and a rat model of iron deficiency anemia was established. It was found that

A Natural Low Phytic Acid Finger Millet Accession Significantly Improves Iron Bioavailability in Indian Women.

Iron deficiency and anemia are common in low- and middle-income countries. This is due to a poor dietary iron density and low iron absorption resulting from the high inhibitory phytic acid content in cereal and millet-based diets. Here, we report that a naturally occurring low phytic acid finger millet accession (571 mg 100 g-1), stable across three growing seasons with normal iron content (3.6 mg 100 g-1), increases iron absorption by 3-folds in normal Indian women. The accessions differing in grain phytic acid content, GE 2358 (low), and GE1004 (high) were selected from a core collection of

Stereochemistry of the Fe(II)- and Fe(III)-cyanide complexes of the homodimeric Scapharca inaequivalvis hemoglobin. A resonance Raman and FTIR study.

Resonance Raman measurements carried out in parallel on ferrous and ferric Scapharca inaequivalvis dimeric hemoglobin cyanide derivatives allowed the identification of the electron density marker bands and heme core size marker bands in both derivatives in comparison with those obtained for the carbonmonoxy and deoxy adducts. The iron cyanide stretching mode, measured for the first time in a ferrous hemoprotein, has been detected at 455 cm(-1), only 6 cm(-1) lower than in the corresponding ferric derivative. This finding demonstrates that the large free energy difference for complex formation

Janus Reactivity of a Mononuclear Non-Heme Iron(III)-Peroxo Complex Toward Indole Oxidation.

Indole oxidation reaction is a pivotal process in organic chemistry, playing a crucial role in the synthesis of various natural products and pharmaceuticals. To-date, indole oxidation is limited to heme enzymes and synthetic iron porphyrin compounds. Herein, we demonstrate its efficient execution in a nonheme iron system. We report the oxidation of a broad range of indole substrates by a mononuclear nonheme iron(III)-peroxo complex, [FeIII(O2)(12-TMC)]+ (1), to their corresponding dioxygenated products. A significant kinetic isotope effect of 5.2 is determined in the reactions of 1 with N─H an

Analysis of the Preserved Amino Acid Bias in Peptide Profiles of Iron Age Teeth from a Tropical Environment Enable Sexing of Individuals Using Amelogenin MRM.

The first dental proteomic profile of Iron Age individuals (ca. 2000-1000 years B.P.), collected from the site of Long Long Rak rock shelter in northwest Thailand is described. A bias toward the preservation of the positively charged aromatic, and polar amino acids is observed. It is evident that the 212 proteins identified (2 peptide, FDR <1%) comprise a palimpsest of alterations that occurred both ante-mortem and post-mortem. Conservation of amino acids within the taphonomically resistant crystalline matrix enabled the identification of both X and Y chromosome linked amelogenin peptides. A n

Anaerobic degradation of benzene and other aromatic hydrocarbons in a tar-derived plume: Nitrate versus iron reducing conditions.

The anaerobic degradation of aromatic hydrocarbons in a plume originating from a Pintsch gas tar-DNAPL zone was investigated using molecular, isotopic- and microbial analyses. Benzene concentrations diminished at the relatively small meter scale dimensions of the nitrate reducing plume fringe. The ratio of benzene to toluene, ethylbenzene, xylenes and naphthalene (BTEXN) in the fringe zone compared to the plume zone, indicated relatively more loss of benzene in the fringe zone than TEXN. This was substantiated by changes in relative concentrations of BTEXN, and multi-element compound specific

Carbon isotope fractionation during abiotic reductive dehalogenation of trichloroethene (TCE).

Dehalogenation of trichloroethene (TCE) in the aqueous phase, either on palladium catalysts with hydrogen as the reductant or on metallic iron, was associated with strong changes in delta13C. In general, the delta13C of product phases were more negative than those of the parent compound and were enriched with time and fraction of TCE remaining. For dehalogenation with iron, the delta13C of TCE and products varied from -42/1000 to +5/1000. For the palladium experiments, the final product, ethane, reached the initial delta13C of TCE at completion of the dehalogenation reaction. During dehalogena

Sulfur-carbon loop enhanced efficient nitrogen removal mechanism from iron sulfide-mediated mixotrophic partial denitrification/anammox systems.

This study successfully established Iron Sulfide-Mediated mixotrophic Partial Denitrification/Anammox system, achieving nitrogen and phosphorus removal efficiency of 97.26% and 78.12%, respectively, with COD/NO3--N of 1.00. Isotopic experiments and X-ray Photoelectron Spectroscopy analysis confirmed that iron sulfide enhanced autotrophic Partial Denitrification performance. Meanwhile, various sulfur valence states functioned as electron buffers, reinforcing nitrogen and sulfur cycles. Microbial community analysis indicated reduced heterotrophic denitrifiers (OLB8, OLB13) under lower COD/NO3--N

Threshold ferritin and hepcidin concentrations indicating early iron deficiency in young women based on upregulation of iron absorption.

BACKGROUND: Plasma ferritin is a widely used indicator to detect iron deficiency, but the threshold ferritin that defines iron deficiency remains uncertain. Our aim was to define the ferritin concentration at which the body begins to upregulate iron absorption from the diet; this could provide a functionally-defined threshold of incipient iron deficiency. We hypothesized this threshold ferritin concentration would correspond to the threshold hepcidin concentration at which iron absorption begins to increase. METHODS: We performed a pooled analysis of our stable iron isotope studies (n = 1058)

Matrix Infrared Spectra of Manganese and Iron Isocyanide Complexes.

Mono and diisocyanide complexes of manganese and iron were prepared via the reactions of laser-ablated manganese and iron atoms with (CN)2 in an argon matrix. Product identifications were performed based on the characteristic infrared absorptions from isotopically labeled (CN)2 experiments as compared with computed values for both cyanides and isocyanides. Manganese atoms reacted with (CN)2 to produce Mn(NC)2 upon λ > 220 nm irradiation, during which MnNC was formed mainly as a result of the photoinduced decomposition of Mn(NC)2. Similar reaction products FeNC and Fe(NC)2 were formed during th
