An approach to α,α-disubstituted α-amino acids is reported. The key step is allyl cyanate-to-isocyanate rearrangement. BACKGROUND: Isocyanates are used in polyurethane production. Dermal exposure to isocyanates can induce contact allergy. The most common isocyanate is diphenylmethane diisocyanate used for industrial purposes. The isomer diphenylmethane-4,4'-diisocyanate (4,4'-MDI) is used in patch testing. Diphenylmethane-4,4'-diamine (4,4'-MDA) is its corresponding amine. This review highlights structural and biosynthetic work on a group of nitrogen-functionalised terpenes that are almost exclusively found in marine invertebrates and the animals that feed on them. The chemical functionality reviewed includes isocyanides, isothiocyanates, formamides, thiocyanates, isocyanates, and dichloroimines. The literature through mid 2003 is reviewed and there are 143 citations.

A mild, metal-free TBAF-catalysed transcarbamoylation of O-phenyl N-alkyl/aryl carbamates enables efficient O- and S-carbamoyl transfer to diverse alcohols and thiols. The protocol delivers carbamates and thiocarbamates in up to 94% yield, tolerating broad functionality and avoiding phosgene, isocyanates and harsh reaction conditions.

We report the clinical findings and the results of inhalation challenge with toluene diisocyanate (TDI) and methacholine in 113 subjects with a history of exposure to TDI and work-related respiratory symptoms. Carbamylation constitutes a posttranslational modification of proteins or amino acids and results from different pathways in vivo.
