Gut microbiome patterns depending on children's psychosocial stress: Reports versus biomarkers.

AIM: Chronic stress increases disease vulnerability factors including inflammation, a pathological characteristic potentially regulated by the gut microbiota. We checked the association between the gut microbiome and psychosocial stress in children/adolescents and investigated which stress parameter (negative versus positive emotion, self-report versus parental report, events versus emotions, biomarker cortisol versus parasympathetic activity) is the most relevant indicator herein. METHODS: Gut microbiome sequencing was completed in fecal samples from 93 Belgian 8-16y olds. Stress measures in

Real-time analysis of gut flora in Entamoeba histolytica infected patients of Northern India.

BACKGROUND: Amebic dysentery is caused by the protozoan parasite Entamoeba histolytica and the ingestion of quadrinucleate cyst of E. histolytica from fecally contaminated food or water initiates infection. Excystation occurs in the lumen of small intestine, where motile and potentially invasive trophozoites germinate from cysts. The ability of trophozoites to interact and digest gut bacteria is apparently important for multiplication of the parasite and its pathogenicity; however the contribution of resident bacterial flora is not well understood. We quantified the population of Bacteroides,

Phloroglucinol Degradation in the Rumen Promotes the Capture of Excess Hydrogen Generated from Methanogenesis Inhibition.

Strategies to manage metabolic hydrogen ([H]) in the rumen should be considered when reducing ruminant methane (CH4) emissions. However, little is known about the use of dietary treatments to stimulate rumen microorganisms capable of capturing the [H] available when CH4 is inhibited in vivo. The effects of the phenolic compound phloroglucinol on CH4 production, [H] flows and subsequent responses in rumen fermentation and microbial community composition when methanogenesis is inhibited were investigated in cattle. Eight rumen fistulated Brahman steers were randomly allocated in two groups recei

The dynamics of the female microbiome: unveiling abrupt changes of microbial domains across body sites from prepartum to postpartum phases.

UNLABELLED: The microbial ecosystem of women undergoes enormous changes during pregnancy and the perinatal period. Little is known about the extent of changes in the maternal microbiome beyond the vaginal cavity and its recovery after birth. In this study, we followed pregnant women [maternal prepartum (mpre), n = 30] into the postpartum period [1 month postpartum, maternal postpartum (mpost), n = 30]. We profiled their oral, urinary, and vaginal microbiome; archaeome; mycobiome; and urinary metabolome and compared them with those of nonpregnant (np) women (n = 29). Overall, pregnancy status (

Lyophilized rumen fluid from goats fed different roughage-based diets: fermentation characteristics, microbial communities, and post-reconstitution in vitro fermentation performance.

INTRODUCTION: Fresh rumen fluid is a promising donor material for rumen fluid transplantation (RFT), but poor storage stability and batch variability limit its use. METHODS: In this study, lyophilized rumen fluid samples from goats fed alfalfa hay or corn stover diets were designated AHP and CSP, respectively. For the in vitro fermentation assay, portions of lyophilized rumen fluid from each batch within each dietary group were pooled and reconstituted; the substrate-supplemented groups were designated AH and CS, whereas AH-CON and CS-CON served as the corresponding substrate-free controls.

Characterisation of the bacterial and archaeal microbiota in processed colostrum collected from a spring-calving dairy herd.

Colostrum feeding is critical for neonatal calf health, providing immunoglobulins (Ig) and other bioactive compounds that support immune function and early microbiome development. While the microbiota of fresh colostrum has been characterised, colostrum on commercial dairy farms is often refrigerated and reheated prior to feeding - practices that may alter its microbial composition. Therefore, the objective of this study was to characterise the prokaryotic community of refrigerated and reheated (processed) colostrum collected immediately before calf feeding. Twenty-one processed colostrum samp

Astragaloside IV alleviates GDM via regulating gut microbiota and gut microbiota metabolomic.

BACKGROUND: Gestational diabetes mellitus (GDM), a severe pregnancy disorder, is a temporary form of diabetes that occurs during gestation. Astragaloside IV (AS IV), a natural and effective composition of Astragalus membranaceus, shows pharmacological effects against diabetes. On the contrary, the effects of AS IV on GDM development are still not clear. This study aims to investigate the role of AS IV in alleviating GDM in rats and determine whether AS IV exerts its anti-GDM properties through the regulation of gut microbiota and metabolite modulation. METHODS: There were six pregnant SD rats

Route of fecal microbiota transplantation delivery determined the dynamics and predictability of donor microbe colonization.

BACKGROUND: Fecal microbiota transplantation (FMT) and the colonization of delivered donor microbes has been reported to improve the negative effects (decrease in body weight, diarrhea, and gut barrier disruption) associated with weaning in pigs. However, delivery of FMT in pigs is still invasive and predicting the colonization or rejection of donor microbes remains challenging. Therefore, this study developed a non-invasive in-feed delivery of FMT and evaluated the effect of FMT mode of delivery on growth performance, gut physiology, microbiota dynamics, and predictability of colonization or

Impacts of dietary fiber level on growth performance, apparent digestibility, intestinal development, and colonic microbiota and metabolome of pigs.

This study aimed to investigate the roughage tolerance of different breeds of pigs. Mashen (MS; n = 80) and Duroc × Landrace × Yorkshire (DLY; n = 80) pigs with an initial body weight of 20 ± 0.5 kg were randomly allotted to four diet treatments (n = 20 of each breed) with different fiber levels. The dietary fiber levels increased by adding 0% to 28% soybean hull to replace corn and soybean meal partially. According to the neutral detergent fiber (NDF) level, all treatments were MS_9N (9% NDF), MS_13.5N (13.5% NDF), MS_18N (18% NDF), MS_22.5N (22.5% NDF), DLY_9N (9% NDF), DLY_13.5N (13.5% NDF)

Effect of Methane Inhibitors on Ruminal Microbiota During Early Life and Its Relationship With Ruminal Metabolism and Growth in Calves.

The present study aimed to determine whether dietary supplementation with methanogen inhibitors during early life may lead to an imprint on the rumen microbial community and change the rumen function and performance of calves to 49-weeks of rearing. Twenty-four 4-day-old Friesian x Jersey cross calves were randomly assigned into a control and a treatment group. Treated calves were fed a combination of chloroform (CF) and 9,10-anthraquinone (AQ) in the solid diets during the first 12 weeks of rearing. Afterward, calves were grouped by treatments until week 14, and then managed as a single group

Compositional and Functional Differences in the Human Gut Microbiome Correlate with Clinical Outcome following Infection with Wild-Type Salmonella enterica Serovar Typhi.

Insights into disease susceptibility as well as the efficacy of vaccines against typhoid and other enteric pathogens may be informed by better understanding the relationship between the effector immune response and the gut microbiota. In the present study, we characterized the composition (16S rRNA gene profiling) and function (RNA sequencing [RNA-seq]) of the gut microbiota following immunization and subsequent exposure to wild-type Salmonella enterica serovar Typhi in a human challenge model to further investigate the central hypothesis that clinical outcomes may be linked to the gut microbi

Structural and functional analysis of the active cow rumen's microbial community provides a catalogue of genes and microbes participating in the deconstruction of cardoon biomass.

BACKGROUND: Ruminal microbial communities enriched on lignocellulosic biomass have shown considerable promise for the discovery of microorganisms and enzymes involved in digesting cell wall compounds, a key bottleneck in the development of second-generation biofuels and bioproducts, enabling a circular bioeconomy. Cardoon (Cynara cardunculus) is a promising inedible energy crop for current and future cellulosic biorefineries and the emerging bioenergy and bioproducts industries. The rumen microbiome can be considered an anaerobic "bioreactor", where the resident microbiota carry out the depoly

Multi-omics analysis of soy isoflavone-induced responses in rumen fermentation, endocrine status and milk production in cows with varying milk yields.

BACKGROUND: Improving milk yield and feed efficiency is pivotal for climate-smart dairy systems, as rumen mediated fermentation governs energy and nitrogen utilization and thereby greenhouse-gas emission intensity. Soybean isoflavones (SIF) may modulate rumen fermentation, yet their effects on rumen function, microbiome features, host endocrine/metabolic responses, and lactation performance-particularly across cows with divergent milk-yield phenotypes-remain unclear. RESULTS: Fifty‑six lactating Holstein cows (28 high‑yield cows, HY; 28 low‑yield cows, LY) were divided into two categories by

Ruminal archaea and bacteria metatranscriptomic responses to supplementation in steers fed low-quality forage.

A basal diet of low-quality forage (LQF) results in decreased forage utilization and animal performance. Protein and starch supplementation can ameliorate these effects by providing additional nitrogen and energy to ruminal microbes. However, the effect of protein and starch supplementation on ruminal microbial gene expression, specifically pathways involved in methanogenesis, is not well-documented. We aimed to evaluate the effect of supplement composition on ruminal microbial gene expression before and after supplementation in steers consuming a basal diet of LQF. Five ruminally-cannulated A

Multi-omics data elucidate parasite-host-microbiota interactions and resistance to Haemonchus contortus in sheep.

BACKGROUND: The integration of molecular data from hosts, parasites, and microbiota can enhance our understanding of the complex biological interactions underlying the resistance of hosts to parasites. Haemonchus contortus, the predominant sheep gastrointestinal parasite species in the tropics, causes significant production and economic losses, which are further compounded by the diminishing efficiency of chemical control owing to anthelmintic resistance. Knowledge of how the host responds to infection and how the parasite, in combination with microbiota, modulates host immunity can guide sele

Effects of medium-chain fatty acid supplementation on performance and rumen fermentation of lactating Holstein dairy cows.

Medium-chain fatty acids (MCFAs) have antimicrobial properties and cause negative or positive effects on animal performance depending on its dosage. We hypothesized that MCFA supplementation at a lower dose (i.e., 0.05-0.2% of dietary DM) would increase rumen pH and milk production without decreasing nutrient digestibility which is typically observed with the higher inclusion rates (i.e., >1% of dietary DM). The objective of this study was to evaluate the effects of MCFA supplementation at a lower dose on productivity, plasma energy metabolite concentrations, apparent total tract nutrient dige
