Light- and ROS-responsive nanomicelles enable targeted combination photothermal molecular therapy in melanoma cells and spheroids.

Stimuli-responsive nanocarriers that respond to both exogenous and endogenous cellular stimuli are highly desirable to enable high payload delivery. Here, we designed novel micelles that are responsive to multiple stimuli, including light (exogenous) and reactive oxygen species (ROS, endogenous). We showed that this dual-stimuli response of light and ROS-responsive micelles (LRMs) can be leveraged to reverse the resistance of melanoma cells to a small molecule drug, trametinib. LRMs were designed with hyaluronic acid (HA) that specifically targets CD44 receptors in B16F0 melanoma cells and sph

Transcriptomic profiling and targeted validation reveal molecular mechanisms of oxygen therapy in high-altitude cerebral injury.

BACKGROUND: Exposure to high-altitude hypoxia is associated with an increased risk of impaired brain structure and function, with oxidative stress and neuroinflammation widely recognized as key mechanisms involved. In this context, hyperbaric oxygen therapy is considered a potential intervention; however, the mechanism by which it affects cerebral function changes caused by high-altitude exposure remains to be further elucidated. OBJECTIVE: This study aims to explore and compare the therapeutic effects of normobaric oxygen (NBO) and hyperbaric oxygen (HBO) on high-altitude cerebral injury (HA

Novel EBV LMP-2-affibody and affitoxin in molecular imaging and targeted therapy of nasopharyngeal carcinoma.

Epstein-Barr virus (EBV) infection is closely linked to several human malignancies including endemic Burkitt's lymphoma, Hodgkin's lymphoma and nasopharyngeal carcinomas (NPC). Latent membrane protein 2 (LMP-2) of EBV plays a pivotal role in pathogenesis of EBV-related tumors and thus, is a potential target for diagnosis and targeted therapy of EBV LMP-2+ malignant cancers. Affibody molecules are developing as imaging probes and tumor-targeted delivery of small molecules. In this study, four EBV LMP-2-binding affibodies (ZEBV LMP-212, ZEBV LMP-2132, ZEBV LMP-2137, and ZEBV LMP-2142) were ident

[RENAL CELL CARCINOMA METASTASIS TO BLADDER DURING MOLECULAR TARGETED THERAPY WITH PAZOPANIB: REPORT OF TWO CASES].

Renal cell carcinoma (RCC) metastasis to the bladder is rare. We report two cases that occurred metachronously during pazopanib treatment for other metastases. To our knowledge, this is the first report to demonstrate bladder metastasis from RCC during molecular targeted therapy with pazopanib. (Case 1) A woman in her 60s was referred to our department for evaluation of an incidental right renal tumor. Dynamic CT showed a 6 cm renal cell carcinoma. In February 201X she underwent laparoscopic right radical nephrectomy, revealing clear cell carcinoma (grade 1>2), stage pT3aN0M0. In February 201X…

Dissecting molecular mechanisms of resistance to NOTCH1-targeted therapy in T-cell acute lymphoblastic leukemia xenografts.

Despite substantial progress in treatment of T-cell acute lymphoblastic leukemia (T-ALL), mortality remains relatively high, mainly due to primary or acquired resistance to chemotherapy. Further improvements in survival demand better understanding of T-ALL biology and development of new therapeutic strategies. The Notch pathway has been involved in the pathogenesis of this disease and various therapeutic strategies are currently under development, including selective targeting of NOTCH receptors by inhibitory antibodies. We previously demonstrated that the NOTCH1-specific neutralizing antibody

BCR-ABL PCR testing in chronic myelogenous leukemia: molecular diagnosis for targeted cancer therapy and monitoring.

The use of tyrosine kinase inhibitors (TKIs) to treat chronic myeloid leukemia (CML) represents the paradigm for modern targeted cancer therapy. Importantly, molecular monitoring using BCR-ABL real-time quantitative reverse transcription polymerase chain reaction (RQ-PCR) for assessing treatment efficacy and quantitating minimal residual disease is a major determinate of practical therapeutic decision-making in the long-term management of this now chronic disease. Herein, we present an overview of CML and the use of TKIs for targeted CML therapy, with an emphasis on the role, application and f

Complete molecular remission of chronic eosinophilic leukemia complicated by CNS disease after targeted therapy with imatinib.

Many cases of hypereosinophilia, formerly classified as hypereosinophilic syndrome, can now be characterized as chronic eosinophilic leukemia (CEL) based on the demonstration of characteristic genetic markers indicating clonality of hematopoiesis. Here we report on a 33-year-old male patient with central nervous system manifestations of CEL and an excellent response to low-dose imatinib (Glivec). Molecular analysis demonstrated a constitutive activation of the platelet-derived growth factor receptor-alpha (PDGFR-A) as the mechanism of responsiveness to imatinib.

Beyond a cancer stem cell marker: molecular state dependent functions of CD133 and state targeted therapy.

CD133 (prominin-1) is among the most widely used cancer stem cell markers, yet two decades of research have yielded persistent contradictions: CD133-negative cells can be tumorigenic, CD133-positive and -negative states can interconvert, and CD133 perturbation produces opposing phenotypes across models, while CD133 directed therapies have encountered toxicities consistent with on-target/off-tumor effects. We propose that these inconsistencies share a common origin: CD133 positivity does not define a homogeneous functional entity. Rather, the functional identity of CD133 is jointly encoded by P

Unraveling Lennox-Gastaut Syndrome: From Molecular Pathogenesis to Precision Diagnosis and Targeted Therapy Evolving Therapeutic Strategies.

Lennox-Gastaut syndrome (LGS) is a rare and severe developmental and epileptic encephalopathy characterized by multiple drug-resistant seizure types, mandatory tonic seizures, cognitive and behavioral impairment, and distinctive electroencephalographic features, including slow spike-wave discharges and generalized paroxysmal fast activity. Despite decades of therapeutic advances, LGS remains associated with profound lifelong disability and the absence of a single disease-defining molecular mechanism. Recent advances in genetics, neurophysiology, and network neuroscience have reframed LGS as a

Multi-layered molecular profiling informs the diagnosis and targeted therapy of desmoplastic small round cell tumor.

Desmoplastic small round cell tumor (DSRCT) is an ultra-rare sarcoma with limited treatment options. Here, we show that comprehensive molecular profiling informs diagnosis and individualized therapy in this disease. We report the results of whole-genome/exome, transcriptome, and DNA methylome analyses performed in 30 refractory DSRCT patients, complemented by (phospho)proteomic profiling in nine, within a nationwide precision oncology program. In eight patients (27%), DSRCT was diagnosed only after molecular profiling. Although DSRCTs have "quiet" genomes, 28 patients (93%) received 107 molecu

Molecular pathways and emerging therapeutic strategies in advanced thyroid cancer: from targeted therapy to precision oncology.

The most prevalent endocrine cancer is thyroid cancer. It may act in a very disparate manner; ranging between slow growing, well differentiated tumours to an aggressive anaplastic subtype. Initial treatment with standard therapy, surgery, radioactive iodine, the thyroid-stimulating hormone inhibition and in some cases, chemotherapy or external-beam radiations are effective with the differentiated cancers at the early stages. Nevertheless, there is no choice with tumours, which become unresponsive to iodine, metastasized, or un- differentiated. Recent developments in molecular oncology have elu

The evolving landscape of the Warburg effect in gastric cancer: From molecular mechanisms to targeted therapy.

BACKGROUND: Gastric cancer (GC) remains a major health burden because of late-stage diagnosis and resistance to systemic therapy. MAIN BODY: In GC, the Warburg effect is driven by interconnected hypoxia and oncogenic signalling, non-coding RNA networks and transcriptional and epigenetic rewiring. These regulators converge on glucose transport and core glycolytic enzymes, including HK2, PFKFB3, PKM2 and LDHA. The resulting increase in aerobic glycolysis, pyruvate-to-lactate conversion and lactate export supports biosynthesis, redox homeostasis and survival under metabolic stress. Beyond its bi

Synergistic Regulation of Tumor Immunity by Integrins and Lectins: From Molecular Mechanisms to Dual-Targeted Therapy.

The high invasiveness and metastatic potential of malignant tumors represent major obstacles to successful clinical treatment and are closely associated with poor patient prognosis. These processes rely heavily on the dynamic remodeling of the tumor microenvironment. Within this milieu, integrins-a family of transmembrane receptors mediating cell-matrix and cell-cell interactions-along with lectins capable of recognizing specific carbohydrate structures, form a complex and coordinated regulatory network that collectively drives tumor progression. This review systematically elucidates the key m
