Targeted mesenchymal stem cell and vascular endothelial growth factor strategies for repair of nerve defects with nerve tissue implanted autogenous vein graft conduits.

Peripheral nerve gaps exceeding 1 cm require a bridging repair strategy. Clinical feasibility of autogenous nerve grafting is limited by donor site comorbidity. In this study we investigated neuroregenerative efficacy of autogenous vein grafts implanted with tissue fragments from distal nerve in combination with vascular endothelial growth factor (VEGF) or mesenchymal stem cells (MSCs) in repair of rat peripheral nerve defects. Six-groups of Sprague-Dawley rats (n = 8 each) were evaluated in the autogenous setting using a 1.6 cm long peroneal nerve defect: Empty vein graft (group 1), Nerve gra

Acutely-dissociated Schwann cells used in tendon autografts for bridging nerve defects in rats: a new principle for tissue engineering in nerve reconstruction.

A new method of acute dissociation of Schwann cells was used to study the effect of addition of such cells to a tendon autograft--a recently-described graft material--on peripheral nerve regeneration in rats. Autologous Schwann cells were obtained from enzymatic dissociation of predegenerated nerves. The tendon autografts were supplied with Schwann cells through brief in vitro coincubation. Schwann cell-free tendon autografts were used as controls. Axonal outgrowth was measured immunohistochemically after four, seven, and 10 days. At seven days, outgrowth was significantly longer in the pretre

Label-Free Deep-Tissue Peripheral Nerve Detection with a Handheld Multimodal OCT Probe and NerveDetNet.

Peripheral nerves buried beneath intact tissue are difficult to visualize during surgery and remain inaccessible to white light wide-field imaging and other surface optical imaging methods. Existing OCT nerve studies have largely relied on exposed nerves or polarization contrast with limited depth penetration, restricting their value for subsurface intraoperative guidance. Here, we introduce, to our knowledge, the first label-free framework for detecting peripheral nerves beneath unopened tissue and resolving their depth using intensity-based OCT structural signatures alone. The framework comb

Coordinate-Independent Ultrasound Assessment for Complex Tissue Kinematics: A Preliminary Study on Median Nerve Mobility.

OBJECTIVE: This paper presents an ultrasound strain-divergence imaging technique, which quantifies complex tissue kinematics without being constrained by imaging coordinates. An in vivo application of the technique was demonstrated on median nerve (MN) mobility. Transverse mobility of MN, a potential biomarker for evaluating MN entrapment in carpal tunnel syndrome (CTS), is featured with large variations across different active gestures, individuals, and operators, so it remains inconclusive in literature. METHODS: Ultrasound data of the MN in the transverse view during active thumb and finge

Impact of Encapsulation Tissue Growth on Selective Recording in Nerve Cuff Electrodes: A Simulation Study.

Peripheral nerve interfaces (PNIs) allow us to extract motor, sensory and autonomic information from the nervous system and use it as control signals in neuroprosthetic and neuromodulation systems. Recent efforts have aimed to improve the recording selectivity of PNIs, including by using spatiotemporal patterns from multi-contact nerve cuff electrodes as input to a convolutional neural network (CNN). Before such a methodology can be translated to humans, its performance in chronic implantation scenarios must be evaluated. We investigated the performance of a CNN-based selective recording appro

Comparison of renal artery, soft tissue, and nerve damage after irrigated versus nonirrigated radiofrequency ablation.

BACKGROUND: The long-term efficacy of radiofrequency ablation of renal autonomic nerves has been proven in nonrandomized studies. However, long-term safety of the renal artery (RA) is of concern. The aim of our study was to determine if cooling during radiofrequency ablation preserved the RA while allowing equivalent nerve damage. METHODS AND RESULTS: A total of 9 swine (18 RAs) were included, and allocated to irrigated radiofrequency (n=6 RAs, temperature setting: 50°C), conventional radiofrequency (n=6 RAs, nonirrigated, temperature setting: 65°C), and high-temperature radiofrequency (n=6 R

Neuronal differentiation of human adipose tissue-derived stem cells for peripheral nerve regeneration in vivo.

OBJECTIVE: To evaluate the ability of a tissue-engineered nerve construct composed of a nerve guidance channel and neurally differentiated human adipose tissue-derived stem cells (hASCs) to enhance peripheral nerve regeneration in a rat sciatic nerve model. DESIGN: A 13-mm sciatic nerve gap was bridged with silastic conduits in 64 athymic nude rats, and differentiated hASCs were implanted into the nerve gap. The effect of repetitive renewal of differentiation medium on days 14 and 28 was further tested. Adequate negative controls and isograft controls were used. SETTING: Academic research.

Nerve cell bodies and small ganglia in the connective tissue stroma of human submandibular glands.

The objective of the study was to investigate the presence and distribution of nerve cell bodies and small ganglia in the stroma of human submandibular gland. A retrospective immunohistochemical study in 13 human submandibular glands, fixed in neutral buffered formalin and embedded in paraffin wax, was undertaken. Six glands were excised in the course of radical neck dissection for oral squamous cell carcinoma and were disease-free, six showed sialadenitis, and one was involved by tuberculosis. Primary antibodies applied were neuron specific enolase, synaptophysin, and glial fibrilliary acidic

Ischemic preconditioning prevents skeletal muscle tissue injury, but not nerve lesion upon tourniquet-induced ischemia.

BACKGROUND: Prolonged ischemia followed by reperfusion (I/R) of skeletal muscle results in significant tissue injury. Ischemic preconditioning (IPC), achieved by brief periods of ischemia before sustained ischemia, has been shown to ameliorate I/R injury in a variety of tissues. We demonstrate that tourniquet hind limb ischemia-induced injury of the muscle benefits from IPC, whereas the peripheral nerve suffers from prolonged ischemia time and mechanical deterioration on IPC. METHODS: In anesthetized rats, hind limb ischemia was induced by tourniquet for 3 hours followed by 24 hours of reperf

Tissue plasminogen activator-mediated fibrinolysis protects against axonal degeneration and demyelination after sciatic nerve injury.

Tissue plasminogen activator (tPA) is a serine protease that converts plasminogen to plasmin and can trigger the degradation of extracellular matrix proteins. In the nervous system, under noninflammatory conditions, tPA contributes to excitotoxic neuronal death, probably through degradation of laminin. To evaluate the contribution of extracellular proteolysis in inflammatory neuronal degeneration, we performed sciatic nerve injury in mice. Proteolytic activity was increased in the nerve after injury, and this activity was primarily because of Schwann cell-produced tPA. To identify whether tPA

Optic nerve compression by extramedullary hematopoietic tissue in a patient suffering from beta-thalassemia intermedia.

We carried out an encephalic magnetic resonance (MR) study in a patient with thalassemia intermedia who complained about change of the visual acuity and of the visual field. Narrowing of the optical canal was present and it was related to the compression of hemopoietic tissue masses. We found a clear relationship between the elderly age, the irregular transfusions, and the optic neuropathy. RMN screening of patients w ith thalassemia intermedia should be mandatory if changes in visual function occur.

[Electron microscopic research on the manifestations of transport-metabolic interactions in intraocular nerve tissue grafts].

Electron microscopic study of intraocular grafts of the septal and hippocampal nervous tissue revealed intensive transport and metabolic interactions between various types of the cells. High level of transport exists between the recipient's blood and intraocular fluid, on the one hand, and the graft, on the other one; the abundance of pinocytotic vesicles in the endothelium, pericytes and the glial end-feet of the capillaries, and the presence of microvilli and cilia on the graft limiting glia. Signs of active communications between interstitial glial cells, as well as between gliocytes and ne

Effects of 2-deoxy-D-glucose on sympathetic nerve activity to interscapular brown adipose tissue.

The effects on firing rate of sympathetic nerves to interscapular brown adipose tissue were measured after induction of intracellular glycopenia by peripheral or central administration of 2-deoxy-D-glucose (2-DG). Injection of 2-DG (250 mg/kg body wt) into the jugular vein rapidly suppressed the sympathetic activity, which declined 84% within 10 min after the injection. This suppression persisted for at least 40 min. Hyperglycemia did not affect the sympathetic activity but partially inhibited the suppressive effect of 2-DG injection. Acute vagotomy failed to block the suppressive effect of 2-

Responses of smooth muscle strips from penile erectile tissue to drugs and transmural nerve stimulation.

1. The mechanical response to drugs and to electrical stimulation of nerves was investigated in isolated strips of intrinsic smooth muscle from the corpora cavernosa penis of the rat. 2. Noradrenaline caused muscle strips to contract in a dose-dependent manner. Contractions could be blocked by pretreatment with the alpha-adrenoreceptor antagonist, phentolamine. 3. Acetylcholine and carbachol had no effect on the baseline tension of muscle strips. Both drugs were relatively ineffective in relaxing noradrenaline-contracted strips. 4. Field stimulation of isolated muscle strips elicited contracti

[Mixed connective tissue disease (Sharp syndrome) with involvement of the trigeminal nerve and facial paralysis].

The authors report a case of mixed connective tissue disease with involvement of the fifth and seventh cranial nerves. While involvement of the trigeminal nerve is well-known, that of the seventh nerve is much less so. However, seventh nerve involvement is beginning to be recognized in other connective tissue diseases such as scleroderma. Neurological manifestations in Sharp syndrome are reviewed.

Silk Fibroin Hydrogel Microneedles Loaded with Recombinant Human Nerve Growth Factor for Corneal Tissue Engineering.

Corneal nerves are essential for maintaining the functional integrity of the ocular surface. Damage to corneal nerves can lead to corneal issues and impaired vision. Current treatments for corneal nerve damage are inadequate, thus highlighting the need for innovative therapeutic approaches. In this study, we present a hydrogel microneedle system designed to facilitate the sustained release of recombinant human nerve growth factor (rhNGF). The microneedle features a tip composed of glycidyl methacrylate modified silk fibroin (SFMA) loaded with rhNGF, photopolymerized for structural integrity, w

In situ tissue profile of rat trigeminal nerve in trigeminal neuralgia using spatial transcriptome sequencing.

BACKGROUND: Trigeminal neuralgia (TN) is the most common neuropathic disorder in the maxillofacial region. The etiology and pathogenesis of TN have not been clearly determined to date, although there are many hypotheses. OBJECTIVE: The goal of this study was to investigate the interactions between different types of cells in TN, particularly the impact and intrinsic mechanism of demyelination on the trigeminal ganglion, and to identify new important target genes and regulatory pathways in TN. METHODS: TN rat models were prepared by trigeminal root compression, and trigeminal nerve tissues we

Metastatic infiltration of nervous tissue and periosteal nerve sprouting in multiple myeloma induced bone pain

Multiple myeloma (MM) is a neoplasia of B plasma cells that often induces bone pain. However, the mechanisms underlying myeloma-induced bone pain (MIBP) are mostly unknown. Using a syngeneic MM mouse model, we show that periosteal nerve sprouting of calcitonin-gene related protein (CGRP + ) and growth associated protein 43 (GAP43 + ) fibres occurs concurrent to the onset of nociception and its blockade provides transient pain relief. MM patient samples also showed increased periosteal innervation. Mechanistically, we investigated MM induced gene expression changes in the dorsal root ganglia (D

Novel electrospun conduit based on polyurethane/collagen enhanced by nanobioglass for peripheral nerve tissue engineering.

Peripheral nerve injury can significantly affect the daily life of individuals with impaired nerve function and permanent nerve deformity. One of the most common treatments is autograft transplantation. Tissue engineering is one of the efficient methods to regenerate injured nerves using scaffolds, cells, and growth factors. Conduits, which are produced by a variety of techniques, could be used as an alternative treatment for patients with damaged nerves. The electrospinning technique is one of the most important and widely used methods for generating nanofiber conduits from biocompatible poly
