Evaluation of Medial Olivocochlear Neural Efferent Pathway in Tinnitus Perception in Normal-hearing Individuals.

OBJECTIVE: The objective of this study is to investigate a possible role of the Medial Olivocochlear (MOC) efferent neural pathway and neural connections responsible for tinnitus generation in silence/sensory deprivation. DESIGN: By placing normal hearing participants in a sound booth for 10 minutes, silence/sensory deprivation was created. This offered assessment of MOC neural pathway in normal hearing participants in silence. Hyperactivity of MOC neural pathway was assessed by its more suppressive effect on Transient Otoacoustic Emissions (TEOAEs) in silence. The required auditory measureme

Rac1 Guides Porf-2 to Wnt Pathway to Mediate Neural Stem Cell Proliferation.

The molecular and cellular mechanisms underlying the anti-proliferative effects of preoptic regulator factor 2 (Porf-2) on neural stem cells (NSCs) remain largely unknown. Here, we found that Porf-2 inhibits the activity of ras-related C3 botulinum toxin substrate 1 (Rac1) protein in hippocampus-derived rat NSCs. Reduced Rac1 activity impaired the nuclear translocation of β-catenin, ultimately causing a repression of NSCs proliferation. Porf-2 knockdown enhanced NSCs proliferation but not in the presence of small molecule inhibitors of Rac1 or Wnt. At the same time, the repression of NSCs prol

Associations between Proprioceptive Neural Pathway Structural Connectivity and Balance in People with Multiple Sclerosis.

Mobility and balance impairments are a hallmark of multiple sclerosis (MS), affecting nearly half of patients at presentation and resulting in decreased activity and participation, falls, injuries, and reduced quality of life. A growing body of work suggests that balance impairments in people with mild MS are primarily the result of deficits in proprioception, the ability to determine body position in space in the absence of vision. A better understanding of the pathophysiology of balance disturbances in MS is needed to develop evidence-based rehabilitation approaches. The purpose of the curre

Constructing a new nigrostriatal pathway in the Parkinsonian model with bridged neural transplantation in substantia nigra.

The physical repair and restoration of a completely damaged pathway in the brain has not been achieved previously. In a previous study, using excitatory amino acid bridging and fetal neural transplantation, we demonstrated that a bridged mesencephalic transplant in the substantia nigra generated an artificial nerve pathway that reinnervated the striatum of 6-hydroxydopamine (6-OHDA)-lesioned rats. In the current study, we report that a bridged mesencephalic transplant can anatomically, neurochemically, and functionally reinstate the 6-OHDA-eradicated nigro-striatal pathway. An excitatory amino

Unravelıng non-canonıcal Wnt sıgnalıng ın neural development and dısorders: a revıew of Wnt/PCP and Wnt/ca²⁺ pathways.

Non-canonical Wnt signaling pathways, particularly the Planar Cell Polarity (Wnt/PCP) and Wnt/Ca²⁺ branches, play critical roles in regulating neural development and maintaining cellular homeostasis within the nervous system. This review provides a comprehensive evaluation of the molecular and cellular functions of these pathways, with a focus on their involvement in cytoskeletal dynamics, cellular polarity, and calcium signaling. Non-canonical Wnt signaling is essential for key neurodevelopmental processes, including neural tube closure, neuronal migration, axon guidance, and synaptic plastic

Dynamics of hidden brain states in post-stroke cognitive impairment: Linking neural activity to neurotransmitter systems and genetic pathways.

BACKGROUND: Post-stroke cognitive impairment (PSCI) is associated with abnormal dynamic functional connectivity, yet the temporal dynamic of brain activity and their underlying molecular mechanisms remain unclear. METHODS: Participants were classified into two groups based on neuropsychological assessments: PSCI group (N = 67) and post-stroke with no cognitive impairment (NPSCI) group (N = 65), alongside 47 healthy controls (HCs). Dynamic brain states were analyzed using a Hidden Markov Model (HMM) with the Brainnetome Atlas, yielding metrics like fractional occupancy (FO), mean dwell time (M

Modulations in neural pathways excitability post transcutaneous spinal cord stimulation among individuals with spinal cord injury: a systematic review.

INTRODUCTION: Transcutaneous spinal cord stimulation (TSCS), a non-invasive form of spinal cord stimulation, has been shown to improve motor function in individuals living with spinal cord injury (SCI). However, the effects of different types of TSCS currents including direct current (DC-TSCS), alternating current (AC-TSCS), and spinal paired stimulation on the excitability of neural pathways have not been systematically investigated. The objective of this systematic review was to determine the effects of TSCS on the excitability of neural pathways in adults with non-progressive SCI at any lev…

Pathways to adolescent social anxiety: Testing interactions between neural social reward function and perceived social threat in daily life.

Recent theories suggest that for youth highly sensitive to incentives, perceiving more social threat may contribute to social anxiety (SA) symptoms. In 129 girls (ages 11-13) oversampled for shy/fearful temperament, we thus examined how interactions between neural responses to social reward (vs. neutral) cues (measured during anticipation of peer feedback) and perceived social threat in daily peer interactions (measured using ecological momentary assessment) predict SA symptoms two years later. No significant interactions emerged when neural reward function was modeled as a latent factor. Seco

Modulation of intracellular calcium activity in interstitial cells of Cajal by inhibitory neural pathways within the internal anal sphincter.

The internal anal sphincter (IAS) functions to maintain continence. Previous studies utilizing mice with cell-specific expression of GCaMP6f revealed two distinct subtypes of intramuscular interstitial cells of Cajal (ICC-IM) with differing Ca2+ activities in the IAS. The present study further examined Ca2+ activity in ICC-IM and its modulation by inhibitory neurotransmission. The spatiotemporal properties of Ca2+ transients in Type II ICC-IM mimicked those of smooth muscle cells (SMCs), indicating their joint participation in the "SIP" syncytium. Electrical field stimulation (EFS; atropine pr

Neural Injury of the Dopaminergic Pathways in Patients with Middle Cerebral Artery Territory Infarct: A Diffusion Tensor Imaging Study.

The mesocortical tract (MCT) and mesolimbic tract (MLT), dopaminergic pathways originating from the ventral tegmental area in the midbrain to the ventral striatum (nucleus accumbens) and prefrontal cortex, play a crucial role in regulating incentive salience. This study aimed to investigate the potential changes in the MCT and MLT pathways following ischemic stroke, such as middle cerebral artery (MCA) infarction. We enrolled thirty-six patients with MCA infarction and forty healthy individuals with no history of psychiatric or neurological disorders. Using diffusion tensor tractography, we ex

Ablation of Death-Associated Protein Kinase 1 Changes the Transcriptomic Profile and Alters Neural-Related Pathways in the Brain.

Death-associated protein kinase 1 (DAPK1), a Ca2+/calmodulin-dependent serine/threonine kinase, mediates various neuronal functions, including cell death. Abnormal upregulation of DAPK1 is observed in human patients with neurological diseases, such as Alzheimer's disease (AD) and epilepsy. Ablation of DAPK1 expression and suppression of DAPK1 activity attenuates neuropathology and behavior impairments. However, whether DAPK1 regulates gene expression in the brain, and whether its gene profile is implicated in neuronal disorders, remains elusive. To reveal the function and pathogenic role of DA

Effects of acute and chronic midthoracic spinal cord injury on neural circuits for male sexual function. II. Descending pathways.

In normal animals, microstimulation of the medullary reticular formation (MRF) has two effects on efferent neurons in the motor branch of the pudendal nerve (PudM). MRF microstimulation depresses motoneuron reflex discharges (RD) elicited by dorsal nerve of the penis (DNP) stimulation and produces long latency sympathetic fiber responses (SFR). The midthoracic spinal location of these descending MRF-PudM projections was studied electrophysiologically using a variety of acute and chronic lesions. Chronic lesions, in 27 mature male rats, included dorsal (DHx) or lateral (LHx) hemisections or mod

An Update about Mechanisms of Acupuncture Treatment for Mental Disorders: We Focus on Signaling Pathways, Neurotransmitters, and Neural Circuits.

This article serves to summarize the mechanism of acupuncture in the treatment of mental disorders. Mental disorders currently have a wide impact, and the number of patients is increasing every year, imposing a heavy burden on the entire society. Among the current first-line treatment methods, psychotherapy is questioned due to its limited efficacy, while drug treatment often comes with adverse reactions and symptom recurrence. As a historical complementary therapy, many clinical studies have confirmed the efficacy of acupuncture in treating mental disorders. However, the treatment rules and p

Vascular and Neural Transcriptomics Reveal Stage-Dependent Pathways to Inflammation and Cognitive Dysfunction in a Rat Model of Hypertension.

BACKGROUND: Chronic arterial hypertension causes cerebral microvascular dysfunction and increases dementia risk in aging. However, cognitive health preservation by therapeutic blood pressure lowering alone is limited and depends on disease duration, the degree of irreversible tissue damage, and whether microvascular function can be restored. This study aimed to understand molecular and cellular temporospatial mechanisms of disease in the course of hypertension. METHODS: We investigated the effects of initial, early chronic and late chronic hypertension in the frontal brain of spontaneously hy

Neural basis underlying the relation between internet addiction tendency and sleep quality: The intrinsic default-mode network connectivity pathways.

Internet addiction (IA) tendency is considered an addictive behavior that results from excessive Internet use, and severely affecting an individual's physical health, emotion, and sleep. Although previous studies indicated that IA tendency was negatively correlated with sleep quality, the underlying neural basis of this relationship remained unclear. To address this issue, we utilized resting-state functional connectivity (RSFC) analysis to identify the neural pathways of the relationship between IA tendency and sleep quality. The behavioral results indicated a positive correlation between the…

In skeletal muscle and neural crest cells, SMCHD1 regulates biological pathways relevant for Bosma syndrome and facioscapulohumeral dystrophy phenotype.

Many genetic syndromes are linked to mutations in genes encoding factors that guide chromatin organization. Among them, several distinct rare genetic diseases are linked to mutations in SMCHD1 that encodes the structural maintenance of chromosomes flexible hinge domain containing 1 chromatin-associated factor. In humans, its function as well as the impact of its mutations remains poorly defined. To fill this gap, we determined the episignature associated with heterozygous SMCHD1 variants in primary cells and cell lineages derived from induced pluripotent stem cells for Bosma arhinia and microp

HBO Promotes the Differentiation of Neural Stem Cells via Interactions Between the Wnt3/β-Catenin and BMP2 Signaling Pathways.

Hyperbaric oxygen (HBO) therapy may promote neurological recovery from hypoxic-ischemic encephalopathy (HIE). However, the therapeutic effects of HBO and its associated mechanisms remain unknown. The canonical Wnt/β-catenin signaling pathways and bone morphogenetic protein (BMP) play important roles in mammalian nervous system development. The present study examined whether HBO stimulates the differentiation of neural stem cells (NSCs) and its effect on Wnt3/β-catenin and BMP2 signaling pathways. We showed HBO treatment (2 ATA, 60 min) promoted differentiation of NSCs into neurons and oligoden
