Effect of mixtures of dietary fibres on the enzyme activity of the rat caecal microflora.

The enzyme activity of the caecal microflora from weanling rats was determined after feeding 1 of 3 basal diets (purified fibre-free; purified plus cellulose; and stock), with or without additional dietary fibre (pectin, i-carrageenan or carboxymethylcellulose 5% w/w). The wet weight of caecal contents and total bacterial numbers were similar for the purified fibre-free and purified plus cellulose diets, yet were significantly higher in animals fed the stock diet. Pectin supplementation of the basal diets had no effect of caecal bacterial numbers, but significantly increased total nitrate redu

Reexamining the risks of drinking-water nitrates on public health.

BACKGROUND: Nitrates in drinking water are generally considered the sole source of nitrite poisoning with methemoglobinemia in infantile methomoglobinemia (IM). However, IM, which occurs during the first 4 months of life, is actually a constellation of cyanosis and hypoxia associated with methemoglobinemia that can result from several other causes. METHODS: This review reexamines the role of nitrate levels in drinking water as a cause of IM and identifies other sources of nitrates that can affect public health and cause chronic diseases. RESULTS: Causes of IM include nitrites in foods, envir

Prognosis of physiological disorders in physic nut to N, P, and K deficiency during initial growth.

The description of physiological disorders in physic nut plants deficient in nitrogen (N), phosphorus (P) and potassium (K) may help to predict nutritional imbalances before the appearance of visual symptoms and to guide strategies for early nutrient supply. The aim of this study was to evaluate the growth of physic nuts (Jatropha curcas L.) during initial development by analyzing the gas exchange parameters, nutrient uptake and use efficiency, as well as the nitrate reductase and acid phosphatase activities and polyamine content. Plants were grown in a complete nutrient solution and solutions

Denitrification in pathogenic bacteria: for better or worst?

A large variety of physiological and taxonomic groups have the ability to use nitrogen oxides as alternative electron acceptors. Brucella spp. is an alpha-proteobacteriaceae that induces a persistent disease in some mammals. Recent work has revealed that a denitrifying gene cluster is important in the interaction of Brucella neotomoae with its host.

Dissimilatory Nitrate Reduction to Ammonium (DNRA) and Denitrification Pathways Are Leveraged by Cyclic AMP Receptor Protein (CRP) Paralogues Based on Electron Donor/Acceptor Limitation in Shewanella loihica PV-4.

Under anoxic conditions, many bacteria, including Shewanella loihica strain PV-4, could use nitrate as an electron acceptor for dissimilatory nitrate reduction to ammonium (DNRA) and/or denitrification. Previous and current studies have shown that DNRA is favored under higher ambient carbon-to-nitrogen (C/N) ratios, whereas denitrification is upregulated under lower C/N ratios, which is consistent with our bioenergetics calculations. Interestingly, computational analyses indicate that the common cyclic AMP receptor protein (designated CRP1) and its paralogue CRP2 might both be involved in the

Nitric oxide levels in Behçet's disease.

Behçet's disease (BD) is a chronic multisystemic disorder which is characterized by a relapsing systemic inflammatory process. In certain inflammatory conditions such as rheumatoid arthritis, over production of nitric oxide (NO) could damage host cells and tissues, either directly and/or following reaction with other free radicals, such as superoxide anion to form species including peroxynitrite or hydroxyl radicals. Excessive superoxide radical production and impaired antioxidant mechanism in both the neutrophils and plasma of patients with BD have been reported. Our study was designed to inv

NirA is a Cyanide-Tolerant Nitrite Reductase Which Protects Pseudomonas aeruginosa From Self-Poisoning.

Host production of nitric oxide in response to P. aeruginosa results in accumulation of nitrite and nitrate at the infection site, with both utilised for anaerobic respiration to support survival. Nitric oxide and nitrite also act as aerobic respiratory inhibitors. P. aeruginosa must overcome these toxic metabolites alongside self-produced cyanide to persist at the infection site. We previously identified a novel nitrite reductase (NirA) that supports P. aeruginosa virulence in a wide range of infection models. In this work, we demonstrate that mutation of nirA inhibits growth of P. aeruginosa

Nitrogen Assimilation and Protein Synthesis in Wheat Seedlings as Affected by Mineral Nutrition. II. Micronutrients.

Activity of nitrate reductase from Triticum aestivum L. seedlings was decreased by deficiencies of molybdenum, zinc, and chlorine. Nitrate accumulated in molybdenum-deficient seedlings, declined in zinc-deficient seedlings, and was unaffected by the other micronutrient treatments. Glutamic acid dehydrogenase activity was decreased by deficiency of molybdenum, the only nutrient that affected the enzyme. Glutamine synthetase activity was decreased only by copper deficiency, and glutamic-oxaloacetic transaminase was not affected by any micronutrient deficiencies. Incorporation of (14)C-leucine in

The Noncanonical Pathway for In Vivo Nitric Oxide Generation: The Nitrate-Nitrite-Nitric Oxide Pathway.

In contrast to nitric oxide, which has well established and important roles in the regulation of blood flow and thrombosis, neurotransmission, the normal functioning of the genitourinary system, and the inflammation response and host defense, its oxidized metabolites nitrite and nitrate have, until recently, been considered to be relatively inactive. However, this view has been radically revised over the past decade and more. Much evidence has now accumulated demonstrating that nitrite serves as a storage form of nitric oxide, releasing nitric oxide preferentially under acidic and/or hypoxic c

Molecular mechanisms associated with polyphosphate accumulation in Microcystis aeruginosa under nitrate limitation.

Bloom-forming cyanobacteria frequently exhibit "luxury phosphorus uptake" and polyphosphate (PolyP) accumulation under nitrate-limited conditions, yet the underlying molecular regulatory mechanisms remain unclear. Our previous study quantitatively demonstrated PolyP accumulation in Microcystis aeruginosa under nitrate limitation. Building on this foundation, the present study investigated the molecular responses of M. aeruginosa to nitrate limitation using integrated transcriptomic and physiological approaches. Nitrate limitation markedly suppressed nitrogen assimilation, as evidenced by reduc

Gallium nitrate in the treatment of lymphoma.

Gallium nitrate is effective and well tolerated for the treatment of cancer-related hypercalcemia. At somewhat higher doses, gallium nitrate also has cytotoxic activity against a variety of cancers. The probable mechanism is inhibition of both ribonucleotide reductase and a protein tyrosine phosphatase. Radioactive gallium ((67)Ga) is concentrated at sites of malignant lymphoma, Hodgkin's disease, and other tumors. Gallium nitrate has substantial single-agent activity in the treatment of patients with advanced lymphoma and has also shown activity when used in combination with other agents. Sig

[Genetic basis of the variability of nitrate reduction trait in Yersinia pestis strains].

The genetic basis of the varying ability to reduce nitrate in strains belonging to different biovars and subspecies of plague-causing microbe has been investigated and the inability to reduce nitrate observed in different intraspecies groups of Yersinia pestis has been shown to stem from mutations in different genes involved in the expression of this trait. The absence of denitrifying activity in strains of altaica and hissarica subspecies was not due to a mutation at position 613 of the periplasmic reductase napA observed in the strains of the biovar medievalis of the main subspecies, but rat

Selective pressure of PFOA on microbial community: Enrichment of denitrifiers harboring ARGs and the transfer of ferric-electrons.

Perfluorooctanoic acid (PFOA), a class of permanent organic pollutants, is frequently detected in surface and ground water, with the latter … a the porin-cytochrome c extracellular electrons transfer system (EET), promoting the expression of nitrate reductases, which in turn further enhanced denitrification. In summary, PFOA regulated the microbial community structure and influenced microbial TN removal functions and increased the contribution of ARGs by the denitrifier hosts, but the PFOA-induced product …

Cerium relieves the inhibition of nitrogen metabolism of spinach caused by magnesium deficiency.

Magnesium is one of the essential elements for plant growth and cerium is a beneficial element for plant growth. However, the effects of the fact that cerium improves the nitrogen metabolism of plants under magnesium deficiency is poorly understood. The main aim of the study was to determine the role of cerium in the amelioration of magnesium-deficiency effects in spinach plants. Spinach plants were cultivated in Hoagland's solution. They were subjected to magnesium deficiency and to cerium chloride administered in the magnesium-present media and magnesium-deficient media. Spinach plants grown

Assimilation of exogenous cyanide cross talk in Oryza sativa L. to the key nodes in nitrogen metabolism.

Exogenous cyanide (CN-) effects on nitrogen (N) uptake, transport, and assimilation in rice seedlings were investigated at the biochemical and molecular levels. Seedlings were treated with either a 2-d or 4-d supply of potassium cyanide (KCN) in the nutrient solution containing nitrate (NO3-) or ammonium (NH4+). Although a KCN-induced increase was recorded in the activity of β-cyanoalanine synthase (β-CAS) in rice tissues of both NH4+-fed and NO3--fed seedlings, the former showed a significantly greater assimilation rate for CN- than the latter. The addition of KCN decreased NO3- uptake and as

[Research progress on the effects of atmospheric nitrogen dioxide on plant growth and metabo-lism].

Nitrogen dioxide (NO2) is one type of the atmospheric nitrogen oxides, which is the main component of atmospheric aerosol particles. Reducing the concentration of atmospheric NO2 can decrease the haze in the air. Atmospheric NO2 deposits on plant leaves by both dry and wet deposition. After leaf absorption, atmospheric NO2 was reduced by two metabolism ways: one was the nitrogen metabolism by reductase mainly in cytoplasm and chloroplast, and the other was the pathway of NO2 decomposition reaction in the apoplast and cytoplasm. Plant absorption of NO2 disturbs plant normal growth and physiolog

Co-culturing fungus Penicillium citrinum and strain Citrobacter freundii improved nitrate removal and carbon utilization by promoting glyceride metabolism.

Exploring the interaction between denitrifying microbial species is significant for improving denitrification performance. In this study, the effects of co-culturing fungus Penicillium citrinum and strain Citrobacter freundii on denitrification were investigated. Results showed that the maximum nitrate removal and carbon utilization in co-culture were 68.0 and 14.1 mg·L-1·d-1, respectively. The total adenosine triphosphatase activity was increased to 9.87 U‧mg-1 protein in co-culture, and nicotinamide adenine dinucleotide production was 1.7-2.3 times that of monoculture, attributing to increas
