The nuclear pore complex protein Nup88 is overexpressed in tumor cells. Immunohistochemical studies have shown that this overexpression is linked to higher aggressiveness of colorectal carcinoma and to enhanced metastatic potential of melanoma cells. Sera of patients with autoimmune liver disease contained antibodies reactive with nuclear lamins. PURPOSE OF REVIEW: Acute necrotizing encephalopathy (ANE) presents with fulminant encephalopathy and characteristic brain lesions following viral infection. The rarity and unpredictability of the disorder have significantly impaired its study. Growing recognition of ANE and the discovery of causative missense mutations in the nuclear pore gene RANBP2 give promising steps toward unraveling this disease. The serological hallmark of primary biliary cirrhosis (PBC) is the presence of antimitochondrial antibodies. OBJECTIVE: To investigate the significance of autoantibodies in patients with primary biliary cirrhosis (PBC).
METHODS: A anti-mitochondrial antibodies-M2(AMA-M2), anti-BCOADC-E2 PDC-E2 OGDC-E2 antibodies (anti-3E/BPO), anti-SP100 antibodies (anti-SP100), anti-promyelocytic leukemia (anti-PML), anti-gp210 antibodies (anti-gp210),and anti-Ro-52 were detected respectively in 330 suspected PBC cases by Western blotting.

We have recently observed reactivity of primary biliary cirrhosis (PBC) sera with several proteins bearing N-acetylglucosamine residues from … e as antigen, 62, 60 and 54 kDa protein bands corresponding to components of the p62 complex in the nuclear pore complex (Kita et al. Biochem. 113, 377-382) were observed in 7, 5 and 6 samples respectively, of the 31 PBC sera. Our data suggest that PBC sera recognize not only the 210 kDa protein but also the p62 complex proteins.
