RNA sequencing of olfactory bulb in Parkinson's disease reveals gene alterations associated with olfactory dysfunction.

The olfactory bulb is involved early in the pathophysiology of Parkinson's disease (PD), which is consistent with the early onset of olfactory dysfunction. Identifying the molecular mechanisms through which PD affects the olfactory bulb could lead to a better understanding of the pathophysiology and etiology of olfactory dysfunction in PD. We specifically aimed to assess gene expression changes, affected pathways and co-expression network by whole transcriptomic profiling of the olfactory bulb in subjects with clinicopathologically defined PD. Bulk RNA sequencing was performed on frozen human

Olfactory system involvement in natural scrapie disease.

The olfactory system (OS) is involved in many infectious and neurodegenerative diseases, both human and animal, and it has recently been investigated in regard to transmissible spongiform encephalopathies. Previous assessments of nasal mucosa infection by prions following intracerebral challenge suggested a potential centrifugal spread along the olfactory nerve fibers of the pathological prion protein (PrP(Sc)). Whether the nasal cavity may be a route for centripetal prion infection to the brain has also been experimentally studied. With the present study, we wanted to determine whether prion

Formation of glomerular maps in the olfactory system.

Sensory perception relies on the decoding of external stimuli into an internal neuronal representation, which requires precise connections between the periphery and the brain. In the olfactory system the axons of chemosensory neurons with the same odorant receptor coalesce into common glomeruli in the olfactory bulb, forming a receptor-topic map. The creation of this map begins prenatally when axons navigate towards the bulb, resort in a receptor-specific manner and terminate in a broad area interdigitated with other axon populations; distinct glomeruli form postnatally. While the initial proc

Chromogranin A-immunoreactive cells in the olfactory system of anuran amphibians.

Chromogranin A (CgA) is a member of the granin family of acidic proteins that are present in the secretory granules of many endocrine and neuroendocrine cells. The specific function(s) of these proteins is not known, but they seem to be the precursors of biologically active peptides, and they may act as helper proteins in the sorting and packaging of peptide hormones and neuropeptides. Using indirect immunohistochemistry, we have found CgA immunoreactivity in the primary olfactory epithelia, the vomeronasal epithelia, the olfactory nerves, and the olfactory bulbs of tadpoles of the American to

Quantitative observations on the nasal epithelia and olfactory innervation in bats. Suggested design mechanisms for the olfactory bulb.

The nasal epithelia of two species of bats were quantified with respect to relative surface areas and olfactory epithelial volumes. In the macrosmatic Aribeus jamaicensis 55.9% of the nasal cavity surface was covered by olfactory epithelium (232.4 mm2), in contrast to only 28.9% in the microsmatic Myotis lucifugus (36.4 mm2). The roles of the various nasal epithelia have been discussed as they may relate to olfaction, respiration and echolocation. In the olfactory bulbs of both species, the estimated concentration of mitral cells approximated at 2,500/mm2 compared to an olfactory nerve concent

Histological and ultrastructural characteristics of the primordial vomeronasal organ in lungfish.

Many vertebrates have two anatomically distinct olfactory organs--the olfactory epithelium and the vomeronasal organ--to detect chemicals such as general odorants and pheromones in their environment. The vomeronasal organ is not present in fish but is present in vertebrates of a higher order than amphibians. Among all extant fishes, the lungfish is considered to be genetically and phylogenetically closest to tetrapods. In this study, we examined the olfactory organs of African lungfish, Protopterus annectens, by lectin histochemistry, immunohistochemistry, and transmission electron microscopy.

Distribution of vasoactive intestinal polypeptide-like immunoreactivity in the olfactory bulb of the rainbow trout (Salmo gairdneri).

The distribution of vasoactive intestinal polypeptide-like structures in the olfactory bulb of the rainbow trout was studied using an indirect-immunoperoxidase technique. Olfactory fibres were very strongly labelled, whereas the fibres or cell bodies in the remaining strata of the olfactory bulb showed no immunoreactivity. In addition, the olfactory nerve fibres were not immunoreactive for methionine- and leucine-enkephalins, motilin, neuropeptide Y, substance P, cholecystokinin-8 and tyrosine-hydroxylase.

Microvasculature of the olfactory organ in the Japanese monkey (Macaca fuscata fuscata).

Olfaction is an important and primitive sense. As its importance has changed with evolution, anatomic adjustments have occurred in its structure and vasculature. Primates are a family of vertebrates that have had to develop their visual system to adapt to the arboreal environment and have evolved from a macrosmatic to a microsmatic species as the optic system has enlarged. This has resulted in anatomic changes of a small but critical area at the base of the brain. This paper describes the three-dimensional vascular anatomy of the olfactory organ of the Japanese monkey (Macaca fuscata fuscata).

Large mechanical changes in the bullfrog olfactory bulb evoked by afferent fiber stimulation.

The isolated olfactory bulb of the bullfrog responds to olfactory nerve stimulation with large mechanical changes in the bulb. The sequence of these changes represents: (1) swelling of the bulb associated with action potential production in the secondary neurons; (2) shrinkage during powerful inhibition of the secondary neurons; and (3) swelling accompanied by repetitive firing of action potentials in the bulb. The effects of bicuculline and Cl-free Ringer on these mechanical changes were examined. The shrinkage of the bulb is closely related with the inhibitory postsynaptic potentials (IPSPs)

Qualitative and quantitative odour discrimation by mitral cells as compared to anterior olfactory nucleus cells.

The responses to odour stimulation of 61 single mitral cells and of 64 single anterior olfactory nucleus (AON) units were simultaneously recorded from rabbits. The test battery consisted of 6 chemical stimuli delivered at two intensity levels corresponding to a hundred-fold difference in concentration. The odour discrimination reaches its best for the mitral cells stimulated weakly; it decreased somewhere when the same cells are stimulated strongly. At AON level the odour discrimination already limited in the case of weak stimuli, almost disappears for strong stimuli. Multidimensional analysis

Structure of the principal olfactory tract.

Although the purpose and importance of the sense of smell in human beings has not been totally clarified, it is one of the principal information channels in macrosmatic animals. It was the first long-distance information system to have appeared in phylogenetic evolution. The objective of this article is to deepen the knowledge of the pathways that join the olfactory epithelium with the cortical olfaction areas, to better understand olfactory dysfunction in human beings. Differential staining and marking techniques were applied to histologic sections obtained from 155 animals of different speci

Odorant receptor-mediated signaling in the mouse.

In the mouse olfactory system, there are approximately 1000 types of odorant receptors (ORs), which perform multiple functions in olfactory sensory neurons (OSNs). In addition to detecting odors, the functional OR protein ensures the singular gene choice of the OR by negative-feedback regulation. ORs also direct the axonal projection of OSNs both globally and locally by modulating the transcriptional levels of axon-guidance and axon-sorting molecules. In these latter processes, the second messenger, cAMP, plays differential roles in the fasciculation and targeting of axons. In this review, we
