Early strain-related changes in enzyme activity in osteocytes following bone loading in vivo.

The skeleton's architecture is matched to the changing loads to which it is subjected because mechanical loading directly or indirectly infl … proportional to the strain magnitude in the bone tissue in the immediate vicinity of the cells. In osteocytes, although the G6PD activity in each individual cell was unchanged by loading, the number of cells displaying activity was increased. This increase was also directly proportional to the applied strain in that area of the cortex (52% compared with 26% …

Intracellular Staphylococcus aureus in osteoblasts and osteocytes and its impact on bone homeostasis during osteomyelitis.

Osteomyelitis is a severe infection of bone tissue that can lead to bone loss and even osteonecrosis. This condition is mostly caused by Gram-positive bacteria, with Staphylococcus aureus being the most common etiological agent. Among the pathophysiological mechanisms involved in osteomyelitis, the ability of S. aureus to be internalized by osteoblasts or osteocytes and to survive within these cells, is particularly noteworthy. Infected osteoblasts and osteocytes not only serve as reservoirs in chronic cases of osteomyelitis but also play an active role in the osteoimmunology process, notably …

The impact of ciliary length on the mechanical response of osteocytes to fluid shear stress.

BACKGROUND: Osteocytes are crucial for detecting mechanical stimuli and translating them into biochemical responses within the bone. The primary cilium, a cellular 'antenna,' plays a vital role in this process. However, there is a lack of direct correlation between cilium length changes and osteocyte mechanosensitivity changes. This study aims to reveal the relationship between ciliary length and nitric oxide (NO) release in osteocytes to show how primary cilia may be involved in reducing osteocyte mechanosensitivity caused by microgravity. MATERIALS AND METHODS: We used the MLO-Y4 cell line …

Single-Cell Transcriptomic Analysis Identifies Senescent Osteocytes That Trigger Bone Destruction in Breast Cancer Metastasis.

Breast cancer bone metastases increase fracture risk and are a major cause of morbidity and mortality among women. Upon colonization by tumor cells, the bone microenvironment undergoes profound reprogramming to support cancer progression, which disrupts the balance between osteoclasts and osteoblasts and leads to bone lesions. A deeper understanding of the processes mediating this reprogramming could help develop interventions for treating patients with bone metastases. Here, we demonstrated that osteocytes (Ot) in established breast cancer bone metastasis develop premature senescence and a di…

Mature primary human osteocytes in mini organotypic cultures secrete FGF23 and PTH1-34-regulated sclerostin.

INTRODUCTION: For decades, functional primary human osteocyte cultures have been crucially needed for understanding their role in bone anabolic processes and in endocrine phosphate regulation via the bone-kidney axis. Mature osteocyte proteins (sclerostin, DMP1, Phex and FGF23) play a key role in various systemic diseases and are targeted by successful bone anabolic drugs (anti-sclerostin antibody and teriparatide (PTH1-34)). However, cell lines available to study osteocytes produce very little sclerostin and low levels of mature osteocyte markers. We have developed a primary human 3D organoty…

Mitochondrial Transcription Factor A added to Osteocytes in a Stressed Environment has a Cytoprotective Effect.

The main precipitant of glucocorticoid-associated femoral head osteonecrosis is widely accepted to be an ischemic-hypoxic event, with oxidat … he addition of glucocorticoid to a hypoxic environment and osteocytic cell necrosis. Using cultured osteocytes MLO-Y4 in a 1% hypoxic environment (hypoxia) to which 1µM dexamethasone (Dex) was added (Dex(+)/hypoxia(+)), an immunocytochemical study was conducted using 8-hydroxy-2'-deoxyguanosine (8-OHdG), an index of oxidative stress, and hypoxia inducible fa …

The Bromodomain Inhibitor N-Methyl pyrrolidone Prevents Osteoporosis and BMP-Triggered Sclerostin Expression in Osteocytes.

(1) Background: In an adult skeleton, bone is constantly renewed in a cycle of bone resorption, followed by bone formation. This coupling process, called bone remodeling, adjusts the quality and quantity of bone to the local needs. It is generally accepted that osteoporosis develops when bone resorption surpasses bone formation. Osteoclasts and osteoblasts, bone resorbing and bone forming cells respectively, are the major target in osteoporosis treatment. Inside bone and forming a complex network, the third and most abundant cells, the osteocytes, have long remained a mystery. Osteocytes are r…

Estrogen deficiency attenuates fluid flow-induced [Ca2+]i oscillations and mechanoresponsiveness of MLO-Y4 osteocytes.

It has been proposed that estrogen regulates the mechanosensitivity of osteocytes; however, the effects of estrogen deficiency that arises during postmenopausal osteoporosis on mechanical stimulation-induced calcium signaling in osteocytes remain elusive. Here, we pretreated MLO-Y4 osteocytes with 10 nM E2 for 2, 3 and 5 d, then simulated postmenopausal conditions either by estrogen withdrawal (EW) from culture medium, or by inhibiting the estrogen receptor by using fulvestrant and estrogen (FE; ICI 182,780) in vitro We investigated [Ca2+]i oscillations and mechanobiologic responses of osteocy…

The induction of a catabolic phenotype in human primary osteoblasts and osteocytes by polyethylene particles.

Polyethylene (PE) wear particles are associated with the osteolysis seen in aseptic loosening that leads to orthopaedic implant failure. Whi … sults suggest that PE particles directly induce a change in the phenotype of mature osteoblasts and osteocytes, consistent with the net loss of bone near orthopaedic implants.

Retrovirus-induced osteopetrosis in mice. Ultrastructural evidence of early virus production in osteoblasts and osteocytes.

Newborn female strain NMRI mice were given injections of a mouse retrovirus (OA MuLV) known to induce osteopetrosis, osteoma, and lymphoma. Femur metaphyses and lumbar vertebrae were investigated ultrastructurally 3 d, 7 d and 28 d after infection. Budding, immature and mature virus was observed associated with osteoblasts and osteocytes, but not with osteoclasts or chondrocytes, 28 d after infection with the virus. No production of virus particles was observed in bone-tissue in mock-treated controls. Thus, the primary target cell for OA virus in bone appears to belong to the osteoblastic/oste…

Osteocyte-Derived Extracellular Vesicles: Bridging the Gap in Skeletal Physiology and Pathophysiology.

PURPOSE OF REVIEW: Osteocytes are the most abundant cells in bone and serve as central skeletal regulators. Recent findings from our and other groups demonstrate that beyond soluble mediators, osteocytes communicate by releasing extracellular vesicles (EVs). This review provides a systematic overview of osteocyte-derived EVs and highlights their importance in diagnostic and therapeutic innovation. RECENT FINDINGS: Accumulating evidence has linked osteocyte mechanotransduction to EV biogenesis and secretion. Mechanically induced calcium oscillations and signaling pathways impact load-induced E…

Noise differentially affects the osteocyte and osteoblast response to fluid shear stress.

Stochastic resonance occurs in non-linear systems when the response to a weak periodic input is strengthened by the presence of noise. Whether a noisy mechanical environment can similarly increase the mechanosensitivity of bone cells, thereby amplifying their response to loading, remains unclear. Both osteocytes and osteoblasts react to fluid flow generated by mechanical loading, although osteocytes are considered the primary mechanosensors in bone. Therefore, the aim of this study was to investigate whether different noise intensities can enhance the mechanosensitivity of MLO-Y4 osteocytes co…

Osteocyte and vascular network in hip osteoarthritis: A multimodal imaging and histological analysis.

OBJECTIVE: Hip osteoarthritis (OA) is characterised by increased remodelling of the subchondral bone, for which the underlying cellular and microvascular mechanisms are unclear. This study investigated alterations in the osteocyte lacunocanalicular network and vascular canal architecture in the femoral head in human hip OA, using a multimodal imaging and histological approach. DESIGN: Femoral head samples from OA patients (n = 8; 4 female, 4 male; age 69 ± 14) undergoing hip replacement, and healthy controls (n = 8; 5 female, 3 male; age 64 ± 12), were imaged by MRI to assess cartilage volume

P62 acts as an intermediator in cadmium-induced osteocyte apoptosis and osteoporosis in mice.

Cadmium (Cd) is a pervasive environmental pollutant. The skeletal system is particularly susceptible and Cd exposure linked to the development of osteoporosis. Intracellular p62 is closely related to bone remodeling and bone marrow integrity. However, the mechanisms of p62 underlying Cd-induced osteocyte damage and osteoporosis remain inadequately understood. A Cd-induced bone injury model was established in C57BL/6 mice, along with a cellular damage model using murine long bone osteocyte-y4 (MLO-Y4) cells. Our study revealed Cd exposure led to p62 high protein levels, disrupting bone homeosta

Establishment and characterization of a human pre-osteocyte cell line: hOsteo4-E9.

Osteocytes, as the most abundant and long-lived cell type in bone tissue, have garnered significant attention due to their critical roles in bone remodeling, bone mechanobiology, bone-related diseases, and systemic homeostasis. While several well-established murine osteocyte cell lines are available, the development of a reliable human-derived osteocyte cell line remains an unmet need in the field. Here, we report the successful purification of primary bone cells from the adult human lumbar articular process and their immortalization via SV40 antigen transduction. The resulting human cell line…

Mechanism of IRF8 on osteocyte apoptosis in steroid-induced osteonecrosis of the femoral head.

BACKGROUND: Steroid-induced osteonecrosis of the femoral head (SONFH) is a metabolic disorder that leads to structural changes, collapse of the femoral head, and joint dysfunction. This study investigates the role of interferon regulatory factor 8 (IRF8) in osteocyte apoptosis in SONFH, so as to find new targets for the treatment of SONFH. METHODS: Murine long bone osteocyte-Y4 cells were cultured and treated with dexamethasone to establish SONFH cell models. si-IRF8 was transfected into the cells. The expression levels of IRF8, B cell leukemia/lymphoma 2 (Bcl-2), BCL2 associated X (Bax), zin

Osteocyte RANKL Drives Bone Resorption in Mouse Ligature-Induced Periodontitis.

Mouse ligature-induced periodontitis (LIP) has been used to study bone loss in periodontitis. However, the role of osteocytes in LIP remains unclear. Furthermore, there is no consensus on the choice of alveolar bone parameters and time points to evaluate LIP. Here, we investigated the dynamics of changes in osteoclastogenesis and bone volume (BV) loss in LIP over 14 days. Time-course analysis revealed that osteoclast induction peaked on days 3 and 5, followed by the peak of BV loss on day 7. Notably, BV was restored by day 14. The bone formation phase after the bone resorption phase was sugges…

Importance of osteocyte-mediated regulation of bone remodelling in inflammatory bone disease.

Although the impact of osteoblast-osteoclast crosstalk in bone remodelling has been intensively studied, the importance of osteocytes, descendants of osteoblasts, in this process has for a long time been neglected. During their embedding phase, osteocytes undergo considerable phenotypic transformation, from a cuboidal, highly metabolically active osteoblast secreting extracellular matrix to a small, stellate, quiescent osteocyte with numerous long dendrites. Osteocytes are encysted in cavities (lacunae) and their dendritic extensions are located in tunnels (canaliculi) forming a remarkable, hi…

Osteocyte lacunar strain determination using multiscale finite element analysis.

Osteocytes are thought to be the primary mechanosensory cells within bone, regulating both osteoclasts and osteoblasts to control load induced changes in bone resorption and formation. Osteocytes initiate intracellular responses including activating the Wnt/β-catenin signaling pathway after experiencing mechanical forces. In response to changing mechanical loads (strain) the osteocytes signal to cells on the bone surface. However, this process of osteocyte activation appears heterogeneous since it occurs in sub-populations of osteocytes, even within regions predicted to be experiencing similar…
