Pathogens without borders: a review on cross-kingdom transmission strategies and pathogenicity of plant and human pathogens.

Emerging evidence indicates that certain plant pathogens-once thought to be restricted to the plant kingdom-can opportunistically infect humans, particularly those who are immunocompromised. This paradigm-shifting recognition of cross-kingdom pathogenicity underscores complex microbial adaptability and highlights an underexplored public health concern. This review is a compilation of current knowledge on bacterial (Pantoea, Burkholderia, Agrobacterium), fungal (Fusarium, Aspergillus, Curvularia), and viral (e.g., TMV, PMMoV) plant pathogens implicated in human disease. Shared eukaryotic cellul…

Diverse endophytic nitrogen-fixing bacteria isolated from wild rice Oryza rufipogon and description of Phytobacter diazotrophicus gen. nov. sp. nov.

Twenty-three nitrogen-fixing bacteria were isolated from surface-sterilized stems and roots of wild rice Oryza rufipogon. Four clusters were defined among these bacteria by SDS-PAGE protein patterns and further confirmed by IS-PCR finger-printing analysis. Phylogenetic analysis of 16S rRNA gene sequences showed that the representative strains LS 8 and LS 18 of cluster II formed a monophyletic group sharing 94.0-97.3% similarities with defined enterobacterial species within the genera Salmonella, Citrobacter, Pantoea, Klebsiella, and Enterobacter. DNA-DNA hybridization, physiological, biochemic…

Investigations into the in vitro antimicrobial activity and mode of action of the phenazine antibiotic D-alanylgriseoluteic acid.

D-Alanylgriseoluteic acid (AGA) is a potent antimicrobial phenazine compound produced by Pantoea agglomerans (Erwinia herbicola) Eh1087. Susceptibility tests against a range of microbes indicated that AGA had a broad spectrum of antimicrobial activity and was particularly active against Gram-positive pathogens. Comparison of the in vitro efficacy of AGA with eight other antibiotics against 119 clinical isolates of Streptococcus pneumoniae demonstrated that all were inhibited by low concentrations of AGA (minimal inhibitory concentration range <or=0.06-0.75 microg/mL), regardless of their susce…

Engineered Rhizobacteria Exploit Bacterial Vesicles for Cross-Kingdom RNAi and Soilborne Disease Control.

The escalating severity of antifungal resistance necessitates novel crop protection strategies. RNAi is promising but faces challenges including environmental instability of dsRNA and poor cellular delivery efficiency. In this study, we engineered the cotton rhizobacterium Pantoea agglomerans YC4 to deliver dsRNA into Verticillium dahliae. Small RNA-seq revealed that bacterial outer membrane vesicles (OMVs) selectively enrich specific endogenous sRNAs compared with intracellular compartments, with a congruent sRNA enrichment pattern observed in fungal cells, thus confirming OMVs as the primary…

Serendipitous isolation of non-Vibrio bacterial strains carrying the cholera toxin gene from environmental waters in indonesia.

We initially attempted to isolate a Vibrio cholerae O1 El Tor biotype that carries a novel variant of the cholera toxin gene (ctxAB) from environmental waters of Indonesia, where the seventh cholera pandemic by V. cholerae O1 El Tor biotype began. Nested PCR targeting the gene revealed that a total of eight strains were found to carry ctxAB. However, sequencing of the 16S rRNA genes of these isolates showed they were not V. cholerae but were either Klebsiella, Enterobacter, Pantoea, or Aeromonas. Subsequent nested PCR assays targeting all genes known to be encoded on the CTX phage (i.e., zot, …

The potential anti-herbivory role of microorganisms on plant thorns.

Thorns, spines and prickles are some of the anti-herbivore defenses that plants have evolved. They were recently found to be commonly aposematic (warning coloration). However, the physical anti-herbivore defense executed by these sharp structures seems to be only the tip of the iceberg. We show that thorns of various plant species commonly harbor an array of aerobic and anaerobic pathogenic bacteria including Clostridium perfringens the causative agent of the life-threatening gas gangrene, Bacillus anthracis, and Pantoea agglomerans. Septic inflammation caused by plant thorn injury can result …

A biosynthetic gene cluster for the acetyl-CoA carboxylase inhibitor andrimid.

Increasing bacterial resistance to antibiotics with conventional targets has focused attention on antibiotics with unconventional targets. One promising candidate, the acetyl-CoA carboxylase (ACC) inhibitor andrimid, is a potent, broad-spectrum antibiotic with high selectivity for prokaryotic ACC. Here, we report the use of a DNA-based approach to clone the andrimid biosynthetic gene cluster from Pantoea agglomerans, yielding a cosmid that confers robust andrimid production on Escherichia coli. This gene cluster encodes a hybrid nonribosomal peptide/polyketide (NRP/PK) synthase with several un…
