Prenatal exposure to phenanthrene impairs spermatogenesis and fertility by elevating apoptosis, altering gene expression, and disrupting steroidogenesis in adult male mice across two generations.

BACKGROUND: Phenanthrene, a polycyclic aromatic hydrocarbon, can enter the human body via various routes and affect reproductive health. Its low molecular weight enables its transfer from the mother to the fetus. However, comprehensive research on its effects on reproductive system development is lacking. This study aimed to examine the impacts of prenatal phenanthrene exposure on the reproductive systems of both the immediate offspring and their progeny. METHODS: Pregnant mice were divided into three groups: control, sham, and phenanthrene. From the detection of the vaginal plug until pregna

Phenanthrene and its halogenated derivative inhibit adipogenesis and decrease peroxisome proliferator-activated receptor gamma (PPARγ) expression independently of aryl hydrocarbon receptor (AhR) activation.

Polycyclic aromatic hydrocarbons (PAHs) are natural by-products of incomplete combustion of fossil fuels, wood, incinerator waste, and are also used in man-made dyes, plastics, and pesticides. Humans are mostly exposed to PAHs through air (ex. smoke inhalation), drinking water, and foods. Phenanthrene (Phe) is the most abundant PAH found in the environment, however there are very few studies that have examined either its systemic health effects or its effects on metabolism and adipogenesis. Halogenated-polycyclic aromatic hydrocarbons (HPAHs) are a class of PAHs that have a halogen bound to an

Trehalose tetraester-producing Rhodococcus erythropolis KB1 enhances alfalfa-assisted rhizosphere remediation of phenanthrene-contaminated soil through plant metabolic reprogramming and microbiome assembly.

Biosurfactant-assisted rhizosphere remediation is a promising strategy for polycyclic aromatic hydrocarbon (PAH)-contaminated soils; however, the mechanisms by which biosurfactants regulate host plant responses to PAH stress and drive rhizosphere microbiome assembly remain unclear. Here, we constructed an alfalfa-assisted rhizosphere remediation system using novel trehalose tetraesters (TTEs) and TTE-producing Rhodococcus erythropolis KB1 to remediate phenanthrene-contaminated soil and decoded mechanisms via multi-omics. The colonization of KB1 and TTE-mediated rhizosphere regulation enhanced

Mechanistic insights into phenanthrene-triggered oxidative stress-associated neurotoxicity, genotoxicity, and behavioral disturbances toward the brandling worm (Eisenia fetida) brain: The need for an ecotoxicological evaluation.

In this study, earthworm (Eisenia fetida) brain was chosen as targeted receptors to probe the mechanisms of oxidative stress-related neurotoxicity, genotoxicity, and behavioral disturbances triggered by PHE. Results showed that PHE stress can initiate significant amounts of ROS, thus triggering oxidative stress in E. fetida brain. These effects were accompanied by a significant increase of damage to macromolecules DNA and lipids, resulting in severe oxidative effects. PHE exposure can induce AChE inhibition by ROS-induced injury and the accumulation of excess ACh at the nicotinic post-synaptic

Structure/reactivity relationships in the benzo[c]phenanthrene skeleton: stable ion and electrophilic substitution (nitration, bromination) study of substituted analogues, novel carbocations and substituted derivatives.

A series of novel carbocations were generated by low-temperature protonation of substituted benzo[c]phenanthrenes, B[c]Phs, and their charge delocalization pathways were elucidated by NMR on the basis of the magnitude of Deltadelta13C values. It has been shown that the protonation regioselectivity is strongly controlled by methoxy and hydroxyl substituents, whose directive effects override methyl substitution effects. Regiocontrol by -OMe and -OH substituents, and its stronger influence relative to methyl groups, was also observed in the nitration and bromination reactions. Charge distribution

Direct synthesis and characterization of site-specific adenosyl adducts derived from the binding of a 3,4-dihydroxy-1,2-epoxybenzo[c]phenanthrene stereoisomer to an 11-mer oligodeoxyribonucleotide.

Site-specifically modified oligonucleotides were obtained in milligram quantities by reacting racemic 3t,4r-dihydroxy-1,2t-epoxy-1,2,3,4-tetrahydrobenzo[c]phenanthrene (B[c]PhDE-2, or anti-B[c]PhDE) with the single deoxyadenosine (dA) residue in the oligodeoxynucleotide d(CTCTCACTTCC). Enzyme digestion of the covalently modified oligonucleotides with the exonuclease spleen phosphodiesterase yielded covalently linked B[ca]PhDE-N6-deoxyadenosyl monophosphate (dAMP) adducts. Comparisons of the reverse phase HPLC retention times and CD spectra of these B[c]PhDE-3'-dAMP mononucleotide adducts, with

Batatasin I, a naturally occurring phenanthrene derivative, isolated from tuberous roots of Dioscorea batatas suppresses eicosanoids generation and degranulation in bone marrow derived-mast cells.

To find anti-inflammatory compounds from the tuberous roots of Dioscorea batatas, we isolated 6-hydroxy-2,4,7-trimethoxyphenanthrene (batatasin I) from the dichloromethane (CH(2)Cl(2)) fraction of this plant. Batatasin I inhibited both the generation of prostaglandin D(2) (PGD(2)), leukotriene C(4) (LTC(4)) and degranulation reaction in mouse bone marrow-derived mast cells (BMMCs). This compound inhibited cyclooxygenase-2 (COX-2) dependent PGD(2) generation in a dose dependent manner, with IC(50) values of 1.78 µM. Western blotting probed with specific anti-COX-2 antibodies showed that the dec

Purification and characterization of a salicylate hydroxylase involved in 1-hydroxy-2-naphthoic acid hydroxylation from the naphthalene and phenanthrene-degrading bacterial strain Pseudomonas putida BS202-P1.

1-Hydroxy-2-naphthoate is formed as an intermediate in the bacterial degradation of phenanthrene. A monooxygenase which catalyzed the oxidation of 1-hydroxy-2-naphthoate to 1,2-dihydroxynaphthalene was purified from the phenanthrene- and naphthalene-degrading Pseudomonas putida strain BS202-P1. The purified protein had a molecular weight of 45 kDa and required NAD(P)H and FAD as cofactors. The purified enzyme also catalysed the oxidation of salicylate and various substituted salicylates. The comparison of the Km and Vmax values for 1-hydroxy-2-naphthoate and salicylate demonstrated a higher ca

Insights into the genome and proteome of Sphingomonas paucimobilis strain 20006FA involved in the regulation of polycyclic aromatic hydrocarbon degradation.

In order to study the mechanisms regulating the phenanthrene degradation pathway and the intermediate-metabolite accumulation in strain S. paucimobilis 20006FA, we sequenced the genome and compared the genome-based predictions to experimental proteomic analyses. Physiological studies indicated that the degradation involved the salicylate and protocatechuate pathways, reaching 56.3% after 15 days. Furthermore, the strain degraded other polycyclic aromatic hydrocarbons (PAH) such as anthracene (13.1%), dibenzothiophene (76.3%), and fluoranthene. The intermediate metabolite 1-hydroxy-2-naphthoic

Rapid screen for bacteria degrading water-insoluble, solid hydrocarbons on agar plates.

A rapid procedure was devised for detecting on solid media bacteria able to degrade water-insoluble, solid hydrocarbons such as the polycyclic aromatic hydrocarbons phenanthrene, anthracene, and biphenyl. After Alcaligenes faecalis AFK2 was inoculated on a plate containing mineral salts agar, an ethereal solution of phenanthrene (about 10%, wt/vol) was sprayed on the surface of the plate, and the plate was incubated at 30 degrees C for 2 to 3 days. Colonies showing degradation were surrounded with clear zones on the opaque plate. A similar clear zone also was formed around colonies which had b

Probing the metal-to-ligand charge transfer first excited state in (η6-naphthalene)Cr(CO)3 and (η6-phenanthrene)Cr(CO)3 by resonance Raman spectroscopy and density functional theory calculations.

The nature of the lowest energy optical transition for the complexes (η(6)-naphthalene)Cr(CO)(3) and (η(6)-phenanthrene)Cr(CO)(3) in the solid state has been investigated by Raman spectroscopy using a range of different excitation wavelengths progressively approaching the resonant condition. Examination of the resonantly enhanced Raman modes confirms that the first absorption is attributed predominantly to a metal-to-arene charge transfer transition for both complexes. A notable difference in the photochemistry of the two complexes was observed. In the case of the phenanthrene complex, populat

Simultaneous removal of organic compounds and heavy metals from soils by electrokinetic remediation with a modified cyclodextrin.

Thousands of sites are contaminated with both heavy metals and organic compounds and these sites pose a major threat to public health and the environment. Previous studies have shown that electrokinetic remediation has potential to remove heavy metals and organic compounds when they exist individually in low permeability soils. This paper presents the feasibility of using cyclodextrins in electrokinetic remediation for the simultaneous removal of heavy metals and polycyclic aromatic hydrocarbons (PAHs) from low permeability soils. Kaolin was selected as a model low permeability soil and it was

The ATP-independent dihydro-2-phenanthroyl-CoA reductase AprC catalyzes two consecutive two-electron reduction steps of dihydro-2-phenanthroyl-CoA to hexahydro-2-phenanthroyl-CoA in anaerobic phenanthrene degradation.

Polycyclic aromatic hydrocarbons (PAHs) are widespread pollutants and especially difficult to degrade under anoxic conditions. The non-substituted three-ring PAH phenanthrene is activated through carboxylation to 2-phenanthroic acid which is then ligated to Co-enzyme A to produce 2-phenanthroyl-CoA, followed by step-wise reduction of the aromatic rings. The first reduction reaction is catalyzed by 2-phenanthroyl-CoA reductase producing dihydro-2-phenanthroyl-CoA. In this study, we elucidated the second reductase enzyme in the pathway, catalyzing the reduction of dihydro-2-phenanthroyl-CoA to h

Methanotrophic bacteria and facilitated transport of pollutants in aquifer material.

In situ stimulation of methanotrophic bacteria has been considered as a methodology for aquifer remediation. Chlorinated aliphatic hydrocarbons such as trichloroethylene are fortuitously oxidized by the methane monooxygenase produced by methanotrophic bacteria. Experimental results are presented that indicate that both colloidal suspensions containing methanotrophic cells and the soluble extracellular polymers produced by methanotrophic cells have the potential to enhance the transport and removal of other environmental contaminants such as polynuclear aromatic hydrocarbons and transition meta

Role of biochar pyrolysis temperature on intracellular and extracellular biodegradation of biochar-adsorbed organic compounds.

Immobilizing organic pollutants by adsorption of biochar in farmland soil is a cost-effective remediation method for contaminated soil. As the adsorption capacity of biochar is limited, biodegradation of biochar-adsorbed organic pollutants was a potential way to regenerate biochars and maintain the adsorption performance of biochars to lower the cost. It could be affected by the biochar pyrolysis temperature, but was not evaluated yet. In this study, biodegradation of adsorbed phenanthrene on a series of biochars with pyrolysis temperatures from 150 to 700 °C by Sphingobium yanoikuyae B1 was i

Inhibitory monoclonal antibody to human cytochrome P450 2B6.

The human cytochrome P450 2B6 metabolizes, among numerous other substrates, diazepam, 7-ethoxycoumarin, testosterone, and phenanthrene. A recombinant baculovirus containing the human 2B6 cDNA was constructed and used to express 2B6 in Sf9 insect cells. The 2B6 was present at 1.8 +/- 0.4% of the total cellular protein and was purified to a specific content of 13.3 nmol/mg protein. Mice were immunized with the purified 2B6, and a total of 811 hybridomas were obtained from the fusion of NS-1 myeloma cells and spleen cells of the immunized mice. Monoclonal antibodies (MAbs) from 24 of the hybrids

Sediment-air equilibrium partitioning of semi-volatile hydrophobic organic compounds part 2. Saturated vapor pressures, and the effects of sediment moisture content and temperature on the partitioning of polyaromatic hydrocarbons.

A gas saturation methodology was used to determine the sediment/air partition coefficient (K(SA)) for phenanthrene and dibenzofuran on a local sediment from the Campus Lake, Baton Rouge. The effects of sediment moisture content, air relative humidity and temperature on K(SA) for phenanthrene were ascertained. The saturated vapor pressures of the compounds were also measured. The sediment moisture content had a striking effect on K(SA); increasing sediment moisture content decreased K(SA). Temperature also had a dramatic effect on K(SA). The variation with temperature was used to evaluate the h

Construction of helical structures with planar chiral [2.2]paracyclophane: fusing helical and planar chiralities.

Optically active conjugated molecules bearing one-handed helical structures, in which two [4]helicenes or two phenanthrenes are stacked at terminal benzene moieties, were prepared using enantiopure [2.2]paracyclophane as a chiral scaffold. The obtained molecules exhibited excellent chiroptical properties such as circularly polarized luminescence with high disymmetry factors. Planar chiral pseudo-meta-disubstituted [2.2]paracyclophane was also optically resolved on the sub-gram-scale.
