Species diversity of sand flies and ecological niche model of Phlebotomus papatasi in central Iran.

Cutaneous leishmaniasis (CL) is the most important vector-borne disease in Iran. Qom Province is a very important area in the case of CL tra … identified. The most abundant species were Sergentomyia sintoni, P. papatasi, P. sergenti s.l. and Phlebotomus alexandri. Two peaks of activity were found in May and August in lowlands; while in mountainous areas they were observed in June and September. Species diversity in mountainous areas was found to be higher than in lowlands. The environmental variabl …

Salivary antigen SP32 is the immunodominant target of the antibody response to Phlebotomus papatasi bites in humans.

BACKGROUND: Zoonotic cutaneous leishmaniasis (ZCL) due to Leishmania major is highly prevalent in Tunisia and is transmitted by a hematophagous vector Phlebotomus papatasi (P. papatasi). While probing for a blood meal, the sand fly injects saliva into the host's skin, which contains a variety of compounds that are highly immunogenic. We recently showed that the presence of anti-saliva antibodies was associated with an enhanced risk for leishmaniasis and identified the immunodominant salivary protein of Phlebotomus papatasi as a protein of approximately 30 kDa. METHODOLOGY/PRINCIPAL FINDINGS: …

Kinetics of antibody response in BALB/c and C57BL/6 mice bitten by Phlebotomus papatasi.

Источник пометил публикацию как отозванную

BACKGROUND: Phlebotomine sand flies are blood-sucking insects transmitting Leishmania parasites. In bitten hosts, sand fly saliva elicits specific immune response and the humoral immunity was shown to reflect the intensity of sand fly exposure. Thus, anti-saliva antibodies were suggested as the potential risk marker of Leishmania transmission. In this study, we examined the long-term kinetics and persistence of anti-Phlebotomus papatasi saliva antibody response in BALB/c and C57BL/6 mice. We also tested the reactivity of mice sera with P. papatasi salivary antigens and with the recombinant pro…

Behavioral evidence for the presence of a sex pheromone in male Phlebotomus papatasi scopoli (Diptera: Psychodidae).

Phlebotomus papatasi (Diptera: Psychodidae) is the Old World sand fly vector of zoonotic cutaneous leishmaniasis caused by Leishmania major (Trypanosomatidae: Kinetoplastida), a debilitating and disfiguring protist parasitic disease prevalent throughout southern Mediterranean countries, the Middle East, as well as southern and eastern European countries, where it is regarded as a serious public health problem. Little is known of the mating ecology of P. papatasi, and, in particular, the role (if any) of pheromones is not known. In this laboratory- and field-based study, we have shown that a ma…

Ecological niche model of Phlebotomus alexandri and P. papatasi (Diptera: Psychodidae) in the Middle East.

BACKGROUND: The purpose of this study is to create distribution models of two sand fly species, Phlebotomus papatasi (Scopoli) and P. alexandri (Sinton), across the Middle East. Phlebotomus alexandri is a vector of visceral leishmaniasis, while P. papatasi is a vector of cutaneous leishmaniasis and sand fly fever. Collection records were obtained from literature reports from 1950 through 2007 and unpublished field collection records. Environmental layers considered in the model were elevation, precipitation, land cover, and WorldClim bioclimatic variables. Models were evaluated using the thres…

Courtship and mating by the sandfly Phlebotomus duboscqi, a vector of zoonotic cutaneous leishmaniasis in the Afrotropical region.

Courtship behaviour of males of the Afrotropical sandfly Phlebotomus duboscqi Neveu-Lemaire (Diptera: Psychodidae) involved mounting the female and clasping her 'waist' with the male coxites placed between the female's thorax and abdomen. This behaviour, which we call 'piggy-backing', was preceded by male wing beating, perhaps involving mate recognition and contact pheromones. It did not seem to be pre- or postcopulatory mate guarding. Piggy-backing was attempted by P. duboscqi males on females of other species (P. papatasi and P. perniciosus) and even on other male P. duboscqi. The majority o…

The potential effect of global warming on the geographic and seasonal distribution of Phlebotomus papatasi in southwest Asia.

The distribution of Phlebotomus papatasi in Southwest Asia is thought to be highly dependent on temperature and relative humidity. A discriminant analysis model based on weather data and reported vector surveys was developed to predict the seasonal and geographic distribution of P. papatasi in this region. To simulate global warming, temperature values for 115 weather stations were increased by 1 degree C, 3 degrees C, and 5 degrees C, and the outcome variable coded as unknown in the model. Probability of occurrence values were then predicted for each location with a weather station. Stations …

Refractory barriers in the sand fly Phlebotomus papatasi (Diptera: Psychodidae) to infection with Leishmania panamensis.

The life cycle of Leishmania panamensis in Phlebotomus papatasi was studied to characterize barriers limiting parasite colonization, differentiation, migration, and attachment in an unnatural sand fly host. The insects were fed a suspension of L. panamensis-infected macrophages and human erythrocytes, and were examined up to 16 days post-infection by light and electron microscopy. Histologic examination of 401 flies showed the peritrophic membrane to be the first important barrier to parasite establishment in the gut lumen. In most flies, parasites were unable to escape from the closed peritro…

[The spread of Phlebotomus papatasi Scop., 1786 through the territory of Central Asia and southern Kazakhstan].

The data on distribution of the main vector of zoonotic cutaneous leishmaniasis (ZCL) Phlebotomus papatasi through Central Asia and southern Kazakhstan are presented. The highest indices of density of this species (100 and more specimen per stick trap) were observed only in the Tejen oasis, rather high ones (1-10, 10-100) in the river valleys and at the foothill, and the lowest one (0.01-0.1)--over the desert territory where ZCL was not practically found. Using these data 2 maps were prepared. The first map shows the pattern of Ph. papatasi geographical distribution with the use of a relative …

First report using near-infrared spectroscopy to age-grade Phlebotomus papatasi [Scopoli, 1786] sand flies.

Vector age and survival directly influence vectorial capacity and disease transmission potential. Current age-grading techniques for phlebotomine sand flies are time-intensive and destructive as they require dissection by highly skilled laboratory personnel that groups hematophagous females as young or old depending on parity status. This study examined the potential of near-infrared spectroscopy (NIRS) to age-grade Phlebotomus papatasi sand flies, a primary vector of Leishmania major, a causative agent of cutaneous leishmaniasis. Female (n = 746) and male (n = 497) adult P. papatasi were coll…

The optimization of PpSP15 purification from salivary glands in Iranian wild Phlebotomus papatasi (Diptera: Psychodidae).

BACKGROUND & OBJECTIVES: Sand fly saliva contains proteins that modulate the host immune system and it plays an important role in both blood feeding and the outcome of Leishmania infections. The profile of the salivary proteins was examined and analyzed from an endemic focus of zoonotic cutaneous leishmaniasis by wild P. papatasi to find local and suitable antigens as potential proteins for developing Leishmania vaccine alongside the development of a new extraction technique. METHODS: Specimens were caught from Bojnord, using funnel and CDC traps. Different methods of protein extraction were

Monitoring Leishmania infection and exposure to Phlebotomus perniciosus using minimal and non-invasive canine samples.

BACKGROUND: In endemic areas of zoonotic leishmaniosis caused by L. infantum, early detection of Leishmania infection in dogs is essential to control the dissemination of the parasite to humans. The aim of this study was to evaluate the serological and/or molecular diagnostic performance of minimally and non-invasive samples (conjunctiva cells (CS) and peripheral blood (PB)) for monitoring Leishmania infection/exposure to Phlebotomus perniciosus salivary antigens in dogs at the beginning and the end of sand fly seasonal activity (May and October, respectively) and to assess associated risks fa…

Acetylcholinesterase of the sand fly, Phlebotomus papatasi (Scopoli): cDNA sequence, baculovirus expression, and biochemical properties.

BACKGROUND: Millions of people and domestic animals around the world are affected by leishmaniasis, a disease caused by various species of flagellated protozoans in the genus Leishmania that are transmitted by several sand fly species. Insecticides are widely used for sand fly population control to try to reduce or interrupt Leishmania transmission. Zoonotic cutaneous leishmaniasis caused by L. major is vectored mainly by Phlebotomus papatasi (Scopoli) in Asia and Africa. Organophosphates comprise a class of insecticides used for sand fly control, which act through the inhibition of acetylchol…
