Non-coding RNAs: cross-kingdom regulators of plant-virus-vector interactions in complex agroecosystems.

Plant viruses depend on insect vectors for dissemination in complex agroecosystems and often exploit plant and insect physiology to enhance transmission. This review synthesizes recent advances in understanding cross-kingdom exchange of non-coding RNAs (ncRNAs) as a central mechanism shaping plant-virus-vector interactions and broader agroecosystem dynamics. ncRNAs are exchanged among plants, insect vectors, and viruses via the phloem, insect saliva, and external plant surfaces through exudates. These molecules can suppress plant antiviral and anti-insect plant responses, modulate vector biolo…

Tissue-specific nuclear spin-spin relaxation anisotropy and water nano-confinement in celery stalks.

Recent magnetic resonance imaging (MRI) measurements have revealed proton spin-spin relaxation anisotropy in only one component of the celery stalk, even though water accounts for up to 95% of its weight and is present throughout all its tissues. Our analysis shows that this anisotropy arises from nanoconfined water within the collenchyma cell walls. In these regions, restricted molecular motion and hydrogen bonding between water molecules and cell-wall constituents give rise to residual anisotropic dipolar interactions. In contrast, water in the xylem, phloem, and parenchyma occupies larger c…

Screening and Functional Analysis of a Novel Salivary Effector DcE13 from the Asian Citrus Psyllid, Diaphorina citri.

The lack of sequence similarity among insect effectors necessitates the independent identification of effectors in the Asian citrus psyllid … e, DcE13 knockdown decreased survival rates and fecundity of D. citri, as well as interrupted their phloem feeding, indicating its role as an effector that facilitates psyllid feeding. These results enrich our current understanding of D. citri feeding behavior while simultaneously offering a potential target gene for controlling this pest.

Fire up Biosensor Technology to Assess the Vitality of Trees after Wildfires.

The development of tools to quickly identify the fate of damaged trees after a stress event such as a wildfire is of great importance. In th … the amount of starch and soluble sugars, while the accumulation of ethanol in the cambial cells and phloem is considered an alarm sign of cell death. However, their determination requires time-consuming laboratory protocols, making the approach impractical in the field. Biosensors hold considerable promise for substantially advancing this field. The gene …

Transport of chemical signals in systemic acquired resistance.

Systemic acquired resistance (SAR) is a form of broad-spectrum resistance induced in response to local infections that protects uninfected p … transported to distal uninfected leaves. SA is preferentially transported via the apoplast, whereas phloem loading of AzA and G3P occurs via the symplast. The symplastic transport of AzA and G3P is regulated by gating of the plasmodesmata (PD). The PD localizing proteins, PDLP1 and PDLP5, regulate SAR by regulating PD gating as well as the subcellular pa …

Morphological abnormalities and cell death in the Asian citrus psyllid (Diaphorina citri) midgut associated with Candidatus Liberibacter asiaticus.

Candidatus Liberibacter asiaticus (CLas) is a phloem-limited, gram-negative, fastidious bacterium that is associated with the development of citrus greening disease, also known as Huanglongbing (HLB). CLas is transmitted by the Asian citrus psyllid (ACP) Diaphorina citri, in a circulative manner. Two major barriers to transmission within the insect are the midgut and the salivary glands. We performed a thorough microscopic analysis within the insect midgut following exposure to CLas-infected citrus trees. We observed changes in nuclear architecture, including pyknosis and karyorrhexis as well …

[Zinc homeostasis in plants: the role of ZIP, MTP and HMA transporters and chelating agents].

Zinc (Zn) is an essential micronutrient involved in numerous physiological processes in plants, serving catalytic, structural, and regulator … uoles. Furthermore, they enhance its symplastic mobility and enable zinc transport in the xylem and phloem, thereby facilitating its distribution to shoots and seeds. Overall, this demonstrates the complex and multilevel nature of zinc homeostasis regulation in plants

Zinkicide Is a ZnO-Based Nanoformulation with Bactericidal Activity against Liberibacter crescens in Batch Cultures and in Microfluidic Chambers Simulating Plant Vascular Systems.

Phloem-limited bacterial "Candidatus Liberibacter" species are associated with incurable plant diseases worldwide. Antimicrobial treatments for these pathogens are challenging due to the difficulty of reaching the vascular tissue they occupy at bactericidal concentrations. Here, in vitro antimicrobial mechanisms of Zinkicide TMN110 (ZnK), a nonphytotoxic zinc oxide (ZnO)-based nanoformulation, were compared to those of bulk ZnO (b-ZnO) using as a model the only culturable species of the genus, Liberibacter crescens Minimum bactericidal concentration (MBC) determination and time-kill assays sho…

Identification and Analysis of Graft-Responsive miRNAs in Mulberry Rootstock-Scion Interactions.

Grafting profoundly influences fruit tree performance, yet the molecular mechanisms underlying rootstock-scion interactions in mulberry (Morus multicaulis) remain poorly understood. To investigate the regulatory networks linking grafting to scion physiology, we performed an integrated analysis combining small RNA sequencing, transcriptome-based KEGG pathway assessment, and qRT-PCR validation of key target genes across five rootstock-scion combinations. High-throughput miRNA profiling of phloem tissues identified 71 conserved and 156 novel miRNAs, which exhibited distinct, genotype-dependent ex…

Ligand-Based Scaffold Hopping of VU730 Identifies Natural Product-Inspired Analogs with Insecticidal and Antifeedant Activity against Asian Citrus Psyllid, Diaphorina citri.

Huanglongbing (HLB), caused by the phloem-restricted bacterium Candidatus Liberibacter (CLas) vectored by the Asian Citrus Psyllid (Hemiptera: Psyllidae; ACP), has led to billion-dollar losses in the global citrus industry. Pharmacological inhibition of inward rectifier potassium (Kir) channels leads to acute mortality and inhibits CLas acquisition by ACP through salivary gland-directed antifeedant activity. Here, we designed a series of analogs that shifted synthetic Kir inhibitors toward a more biogenic chemical space through systematic modification of VU730 (C17H19N3O2). Analog 7c, which co…

Plant Sugar Transporters (SWEET): An Emerging Role in Carbon-Lipid Homeostasis.

In plants, the directed distribution of photosynthetic products from source organs to heterotrophic sink organs forms the material basis for biomass accumulation and crop yield. SWEETs (sugars will eventually be exported transporters) facilitate passive sucrose efflux to link source and sink transport during phloem loading for plant carbon allocation. However, it remains unclear how SWEET-mediated sugar transport coordinates carbon allocation and lipid metabolism. Therefore, this Review integrates the structural evolution, transport mechanisms, and physiological functions of SWEETs. It investi…

The Greenhouse Whitefly Trialeurodes vaporariorum Management in Africa and Beyond.

Whiteflies (Hemiptera: Aleyrodidae) are among the most destructive pests of vegetables and ornamental crops, causing severe yield losses through phloem feeding and virus transmission. Although historically most research has centered on Bemisia tabaci, the greenhouse whitefly Trialeurodes vaporariorum is rapidly emerging as a major pest across sub-Saharan Africa. Its spread in both open-field and protected cropping systems is linked to shifts in land use, climate variability, and intensified production systems. Furthermore, growing insecticide resistance to commonly used insecticides threatens …

Is photosynthetic transcriptional regulation in Triticum aestivum L. cv. 'TugelaDN' a contributing factor for tolerance to Diuraphis noxia (Homoptera: Aphididae)?

Diuraphis noxia (Russian wheat aphid, RWA) is a major pest on wheat in South Africa and most other wheat growing countries. Being a probing-sucking insect, RWAs insert their stylets into the phloem sieve elements and feed on the phloem sap. This feeding causes necrotic lesions in resistant varieties, or decoloration of leaves and death in susceptible varieties. In an effort to broaden our understanding on the response of the plant to RWA feeding, we synthesized and analyzed expressed sequence tags (ESTs) from suppression subtractive hybridization (SSH) libraries. These libraries were construct…

An LRR co-receptor kinase essential for systemic nitrogen-demand signalling.

Plants have evolved a signalling pathway in which, when one root senses local nitrogen (N) deficiency, nitrate uptake by other roots is enhanced in a complementary manner. This long-range communication, known as systemic N-demand signalling, is triggered when the root-to-shoot mobile signal, C-TERMINALLY ENCODED PEPTIDE (CEP), which is induced in roots under N starvation, is perceived by CEP RECEPTOR 1 (CEPR1) expressed in the leaf phloem. However, the molecular components required for CEP-dependent CEPR1 activation remain unknown. Here we identified a leucine-rich repeat receptor kinase that …

Inward Rectifier Potassium (Kir) Channel Inhibitors Protect Citrus from the Asian Citrus Psyllid by Inducing Toxicity and Inhibition of Feeding.

Huanglongbing (HLB), caused by the phloem-restricted bacterium Candidatus Liberibacter asiaticus (C. Las), is vectored by the Asian citrus psyllid (ACP) and threatens global citrus production. We demonstrate that VU041 (C19H20F3N3O), a small-molecule inhibitor of inward rectifier potassium (Kir) channels, provides a dual-mechanism approach to management of Huanglongbing (HLB) disease by combining acute toxicity with antifeedant activity. Topical and tarsal contact bioassays revealed that VU041 induced rapid mortality to ACP (24 h LD50 = 25 ng/insect) and reduced salivary gland secretion to sup…

The MYB transcription factor APL is a rational target for base editing to engineer flowering time.

Achieving optimal flowering time is a pivotal factor in precision plant breeding. Here, in maize, we characterized an EMS-induced additive delayed-flowering mutant, edf1. edf1 and its heterozygotes are late-flowering under both long- and short-day photoperiods in the field, and harbor a splice-donor mutation in ALTERED PHLOEM DEVELOPMENT (APL). This mutation leads to the accumulation of the minor APL-β transcript, a splice variant expressed across a large natural maize population. Expression of the major splice variant APL-α is associated with flowering initiation; APL knockdown delays floweri…

Iron deficiency changes regulatory mechanisms governing sieve element cell differentiation.

Plant cell differentiation incorporates environmental cues over time to optimize overall growth. Iron deficiency influences development, suc … element differentiation, particularly enucleation and cell wall thickening, and consequently delays phloem sap unloading to roots. Using Dynamic Bayesian modeling we also characterize how iron deficiency changes the fundamental structure of the gene regulatory network associated with sieve element differentiation. We identify DOF1.5 as a positive regulat …

Alkaloid binding to opium poppy major latex proteins triggers structural modification and functional aggregation.

Opium poppy accumulates copious amounts of several benzylisoquinoline alkaloids including morphine, noscapine, and papaverine, in the specialized cytoplasm of laticifers, which compose an internal secretory system associated with phloem throughout the plant. The contiguous latex includes an abundance of related proteins belonging to the pathogenesis-related (PR)10 family known collectively as major latex proteins (MLPs) and representing at least 35% of the total cellular protein content. Two latex MLP/PR10 proteins, thebaine synthase and neopione isomerase, have recently been shown to catalyze…

Sulfur dioxide inhibition of translocation in bean plants.

Exposure of the source leaf of bean (Phaseolus vulgaris L. cv. Black Valentine) for 2 hours to 2.9 microliters per liter SO(2) inhibited the net photosynthetic rate an average of 75% and, simultaneously, the translocation rate an average of 45%. Calculations indicated that the experimentally determined translocation rates from SO(2)-stressed leaves were lower than were the rates expected on the basis of the observed reductions in photosynthesis. It is inferred that, under SO(2) stress, the phloem-loading system becomes a major limiting step in controlling the translocation rate.Following remov…
