The response of PRL to the oral administration of the dopamine receptor-blocking agent metoclopramide and the effect of metoclopramide on the TRH-induced release of PRL and TSH were measured in eight patients with hyperthyroidism and in eight age- and sex-matched euthyroid controls. As expected from the known direct inhibitory influence of thyroid hormones on pituitary TSH secretion, there was no TSH rise in response to metoclopramide in either group. A study was undertake to determine whether impotence in some diabetic patients might be due to a coincidental prolactinoma and therefore be potentially treatable. Of 83 consecutive men attending a diabetic clinic, three were found to have persistent mile to moderate hyperprolactinaemia. The administration of a dopamine antagonist, chlorpromazine, and two opiates, morphine and methadone, resulted in a significant rise in serum PRL within 90-150 min. Prior administration of dopamine receptor agonists (apomorphine, levodopa, aand bromocriptine) blocked this effect. In contrast, cyproheptadine, a serotonin antagonist, did not. We suggest that the opiates induce hyperprolactinemia in man via dopamine receptor blockade.

OBJECTIVE: Amylin is a 37-amino acid peptide that is secreted from the pancreatic beta cells. This peptide is cosecreted with insulin from the pancreas by glucose. Amylin plays a role in glucose homeostasis and in the regulation of lipid metabolism. Amylin receptors were identified in central nervous system of rats.
