Biophysical and functional insights into YchP, a putative invasin outer membrane protein of Salmonella Typhimurium, and its role in virulence.

Salmonella enterica serovar Typhimurium is a Gram-negative pathogen responsible for gastrointestinal infections in humans and animals. Its ability to adhere to and invade intestinal epithelial cells is mediated by several outer membrane proteins (OMPs), including OmpC, OmpD, OmpA, and OmpF. YchP, a lesser-known OMP, has emerged as a putative invasin targeting β-integrins on host cells. In silico analysis revealed YchP as a 12-stranded β-barrel protein with a 41-residue signal peptide and a pore diameter that expands from 4 Å to 10 Å along an SU-shaped cavity, suggesting a potential role in sel

Cholera toxin: an intracellular journey into the cytosol by way of the endoplasmic reticulum.

Cholera toxin (CT), an AB(5)-subunit toxin, enters host cells by binding the ganglioside GM1 at the plasma membrane (PM) and travels retrograde through the trans-Golgi Network into the endoplasmic reticulum (ER). In the ER, a portion of CT, the enzymatic A1-chain, is unfolded by protein disulfide isomerase and retro-translocated to the cytosol by hijacking components of the ER associated degradation pathway for misfolded proteins. After crossing the ER membrane, the A1-chain refolds in the cytosol and escapes rapid degradation by the proteasome to induce disease by ADP-ribosylating the large G

[Expression, refolding and biological activity of recombinant type-I metalloproteinase acutolysin a from Agkistrodon acutus].

Type-I snake venom metalloproteinase acutolysin A gene was cloned into the prokaryotic expression vector pBAD/gIIIA and the resulting recombinant plasmid pDS was obtained. By the induction with 0.02% L-(+)-arabinose, the recombinant metalloproteinase was expressed in insoluble inclusion body in E. coli TOP10 and reached up to 5%--10% of total bacterial proteins. The recombinant metalloproteinase has an additional sequence of N-terminal 22 amino acids and C-terminal 8 amino acids (housing 6 histidines), both of which derived from the vector. The purified inclusion body was solubilized by 8 mol/

Expanding role of molecular chaperones in regulating α-synuclein misfolding; implications in Parkinson's disease.

Protein misfolding under stressful environmental conditions cause several cellular problems owing to the disturbed cellular protein homeostasis, which may further lead to neurological disorders like Parkinson's disease (PD), Alzheimer's disease (AD), Amyloid lateral sclerosis and Huntington disease (HD). The presence of cellular defense mechanisms like molecular chaperones and proteasomal degradation systems prevent protein misfolding and aggregation. Molecular chaperones plays primary role in preventing protein misfolding by mediating proper native folding, unfolding and refolding of the poly

Physiological Functions of Heat Shock Proteins.

Heat shock proteins (HSPs) are molecular chaperones involved in a variety of life activities. HSPs function in the refolding of misfolded proteins, thereby contributing to the maintenance of cellular homeostasis. Heat shock factor (HSF) is activated in response to environmental stresses and binds to heat shock elements (HSEs), promoting HSP translation and thus the production of high levels of HSPs to prevent damage to the organism. Here, we summarize the role of molecular chaperones as anti-heat stress molecules and their involvement in immune responses and the modulation of apoptosis. In add

Immunogenicity of a recombinant envelope domain III protein of dengue virus type-4 with various adjuvants in mice.

Dengue fever, a mosquito borne viral disease, has become a major public health problem with dramatic expansion in recent decades. Several dengue vaccines are at developing stage, none are yet available for humans. There is no vaccine or antiviral therapy available for dengue fever till date. Domain III of envelope protein is involved in binding to host receptors and it contains type and subtype-specific epitopes that elicit virus neutralizing antibodies. Hence domain III is an attractive vaccine candidate. In the present study we report the immunomodulatory potential of refolded D4EIII protein

Bacterial overexpression of recombinant heteroscorpine-1 (rHS-1), a toxin from Heterometrus laoticus scorpion venom: trends for antibacterial application and antivenom production.

Heteroscorpine-1 (HS-1) was identified as a member of the scorpine family. HS-1 shows insecticidal activities, exhibiting a low median lethal dose (LD50) in mealworm (Tenebrio molitor L.) and inhibitory activities against Bacillus subtilis, Klebsiella pneumoniae, and Pseudomonas aeruginosa. In this study, a recombinant HS-1 (rHS-1) was produced by overexpression in E. coli. A large yield of product was obtained. The structure of purified rHS-1 was confirmed through mass spectrometry. Both anti-crude venom and anti-rHS-1 antibodies specifically recognized rHS-1, suggesting its structural simila

Protein homeostasis, aging and Alzheimer's disease.

Alzheimer's disease (AD) is one key medical challenge of the aging society and despite a great amount of effort and a huge collection of acquired data on molecular mechanisms that are associated with the onset and progression of this devastating disorder, no causal therapy is in sight. The two main hypotheses of AD, the amyloid cascade hypothesis and the Tau hypothesis, are still in the focus of AD research. With aging as the accepted main risk factor of the most important non familial and late onset sporadic forms of AD, it is now mandatory to discuss more intensively aspects of cellular agin

Synthesis of the full-length hepatitis B virus core protein and its capsid formation.

Chronic infection with hepatitis B virus (HBV) is a major cause of cirrhosis and liver cancer. Capsid assembly modulators can induce error-prone assembly of HBV core proteins to prevent the formation of infectious virions, representing promising candidates for treating chronic HBV infections. To explore novel capsid assembly modulators from unexplored mirror-image libraries of natural products, we have investigated the synthetic process of the HBV core protein for preparing the mirror-image target protein. In this report, the chemical synthesis of full-length HBV core protein (Cp183) containin

The role of heat shock proteins in kidney disease.

Heat Shock Proteins (HSP) belong to the family of intracellular proteins that are constitutively expressed and are upregulated by various stressors including heat, oxidative and chemical stress. HSP helps in reparative processes, including the refolding of damaged proteins and the removal of irreparably damaged proteins that would initiate cellular death or apoptosis. A growing body of evidence has expanded the role of HSP and defined their role in diseases such as neurodegenerative disorders, cancer, ischemic heart disease and kidney diseases. The protective role of HSP in ischemic renal inju

Heat shock protein 40-Gok1 isolation from Toxoplasma gondii RH strain.

Toxoplasma gondii is ubiquitous obligate intracellular parasite and is one of the most important pathogen for humans and animals. In humans, T. gondii has two life forms: tachyzoites and bradyzoites. Tachyzoites form of T. gondii can cause acute infection, and it is called toxoplasmosis. The development of latent bradyzoites from rapidly growing tachyzoites has been linked to cellular and environmental stresses which are associated with heat shock proteins (Hsps). Hsps play a protective role against stressors. Hsp40 is an important member of Hsp family and T. gondii has 36 predicted Hsp40 fami

The Role of Disulfide Bridges in the Interaction of E. coli -Derived Recombinant SCARB2 and EV-A71's Capsid.

BACKGROUND: Hand, Foot, and Mouth disease is an acute infectious disease caused by a group of enteroviruses, including Coxsackievirus A16 and Enterovirus 71. EV-A71-causing disease can give rise to severe complications in children, leading to meningitis, encephalitis, and even death. A potential approach to prevent the virus spread is inhibiting the invasion of EV-A71 into target cells, thereby helping to prevent not only the spread of EV-A71 in the community but also lessen the risk of outbreaks. EV-A71 cell's receptor, human scavenger receptor class B member 2, SCARB2, was used as a trap to

[Mild solubilization, purification, and immunoreactivity characterization of VP1 inclusion body protein of serotype O foot-and-mouth disease virus based on sodium lauroyl sarcosinate].

To efficiently prepare the capsid protein VP1 of serotype O foot-and-mouth disease virus (FMDV) with favorable immunoreactivity and to address the challenges that VP1 is prone to form inclusion bodies in the Escherichia coli expression system and is difficult to refold, this study developed a novel process based on sodium lauroyl sarcosinate (LSN) for the mild solubilization and chromatographic purification of FMDV VP1 inclusion body protein. First, the recombinant SUMO-VP1 protein was expressed in E. coli by induction, and Western blotting results revealed that the recombinant SUMO-VP1 protei

Sequestration of Ribosome during Protein Aggregate Formation: Contribution of ribosomal RNA.

An understanding of the mechanisms underlying protein aggregation and cytotoxicity of the protein aggregates is crucial in the prevention of several diseases in humans. Ribosome, the cellular protein synthesis machine is capable of acting as a protein folding modulator. The peptidyltransferase center residing in the domain V of large ribosomal subunit 23S rRNA is the centre for the protein folding ability of the ribosome and is also the cellular target of several antiprion compounds. Our in vitro studies unexpectedly reveal that the partial unfolding or aggregation of lysozyme under reducing c

Common Features of Enveloped Viruses and Implications for Immunogen Design for Next-Generation Vaccines.

Enveloped viruses enter cells by inducing fusion of viral and cellular membranes, a process catalyzed by a specialized membrane-fusion protein expressed on their surface. This review focuses on recent structural studies of viral fusion proteins with an emphasis on their metastable prefusion form and on interactions with neutralizing antibodies. The fusion glycoproteins have been difficult to study because they are present in a labile, metastable form at the surface of infectious virions. Such metastability is a functional requirement, allowing these proteins to refold into a lower energy confo

Recombinant expression of margatoxin and agitoxin-2 in Pichia pastoris: an efficient method for production of KV1.3 channel blockers.

The K(v)1.3 voltage-gated potassium channel regulates membrane potential and calcium signaling in human effector memory T cells that are key mediators of autoimmune diseases such as multiple sclerosis, type 1 diabetes, and rheumatoid arthritis. Thus, subtype-specific K(v)1.3 blockers have potential for treatment of autoimmune diseases. Several K(v)1.3 channel blockers have been characterized from scorpion venom, all of which have an α/β scaffold stabilized by 3-4 intramolecular disulfide bridges. Chemical synthesis is commonly used for producing these disulfide-rich peptides but this approach

Stabilization of Bone Morphogenetic Protein-2 at Physiological pH: Contrasting Roles of CHAPS and Arginine in Aggregation Inhibition.

Bone morphogenetic protein-2 (BMP-2) is a key osteoinductive growth factor employed clinically in spinal fusion and fracture repair where bone regeneration is insufficient. However, its therapeutic efficacy is limited by low solubility and aggregation at physiological pH. This study investigates BMP-2 aggregation and identifies additives that stabilize its native, biologically active dimer form under physiological conditions. We demonstrate that 3-[(3-cholamidopropyl) dimethylammonio]-1-propanesulfonate (CHAPS) efficiently refolded monomeric BMP-2 from inclusion bodies into dimeric form but fa

Prion protein self-interactions: a gateway to novel therapeutic strategies?

Transmissible spongiform encephalopathies (TSEs) or prion diseases are fatal neurodegenerative disorders and include among others Creutzfeldt-Jakob disease in humans, bovine spongiform encephalopathy (BSE) in cattle, and scrapie in sheep. The central event in disease development in TSEs is the refolding of the normal host-encoded cellular prion protein (PrP) into abnormal and disease associated prion protein. The agent is thought to consist mainly or exclusively of these pathologically folded PrP molecules. The exact molecular mechanisms underlying this process and the role of normal PrP in th

2-Methyltetrahydrofuran and cyclopentylmethylether: two green solvents for efficient purification of membrane proteins like FhuA.

β-Barrel shaped membrane proteins are attractive hosts for hybrid catalysts in which reactions are controlled through space. Production and extraction of β-barrel shaped membrane proteins in gram scale is challenging due to their hydrophobicity. Solvent mixtures such as chloroform/methanol (CM) are widely used for membrane protein extraction but toxicity and mutagenicity were reported in several cases. 2-Methyltetrahydrofuran (2-MeTHF) and cyclopentylmethylether (CPME) are two green (reduction of solvent-related environmental damage in chemical production) and potentially efficient solvents fo
