Biomimetic exosomal delivery of ginsenoside Rd reduces post-stroke depression via microglial reprogramming in a pathway involving EGFR/JAK2/STAT3.

Post-stroke depression (PSD) represents a complex neuropsychiatric challenge characterized by persistent neuroinflammation and synaptic dysfunction, yet effective therapeutic interventions are constrained by the blood-brain barrier (BBB) and the lack of specific targets. Using a multidimensional screening strategy for Chaihu-Jia-Longgu-Muli Decoction (CLM), we identified ginsenoside Rd (Rd) as a key blood-absorbed bioactive constituent associated with Epidermal Growth Factor Receptor (EGFR) signaling. To overcome the bioavailability bottleneck, a biomimetic nanodelivery system is engineered by

Mechanical unloading promotes adult cardiomyocyte proliferation through epicardial NRG1-ERBB4 signaling.

In mammals, the postnatal increase in cardiac workload coincides with the loss of cardiomyocyte proliferative capacity, rendering adult cardiomyocytes permanently post-mitotic. Whether reducing load can restore regenerative potential in the adult heart remains unknown. Here we use a heterotopic heart transplantation model, in which the donor heart is vascularized but nonpumping, to show that mechanical unloading induces adult cardiomyocyte proliferation, revealed by Ki67-based and MADM (mosaic analysis with double markers) lineage tracing. Applying heterotopic transplantation to infarcted hear

ERBB4 as a therapeutic target in aortic dissection: Implications for cell-based therapies in vascular regeneration.

This study investigates the role of Erb-B2 receptor tyrosine kinase 4 (ERBB4) in aortic dissection (AD) pathogenesis and its potential as a therapeutic target in cell-based therapies. Using immunohistochemical (IHC) staining, we examined the phenotypic transition of vascular smooth muscle cells (VSMCs) in thoracic aortic tissue from AD patients. RNA sequencing identified differentially expressed genes (DEGs), highlighting ERBB4 as a key regulator. In vitro, ERBB4 knockdown in human aortic smooth muscle cells (HASMCs) was assessed using cell counting kit-8 (CCK-8 assays), wound healing, transwe

Nuclear Binding Protein 2/Nesfatin-1 Affects Trophoblast Cell Fusion during Placental Development via the EGFR-PLCG1-CAMK4 Pathway.

Previous studies have shown that nuclear binding protein 2 (NUCB2) is expressed in the human placenta and increases with an increase in the syncytialization of trophoblast cells. This study aimed to investigate the role of NUCB2 in the differentiation and fusion of trophectoderm cells. In this study, the expression levels of NUCB2 and E-cadherin in the placentas of rats at different gestation stages were investigated. The results showed that there was an opposite trend between the expression of placental NUCB2 and E-cadherin in rat placentas in different trimesters. When primary human trophobl

Reduced myocardial NRG-1/ErbB4 signaling is associated with STING-linked macrophage pyroptotic signaling in sepsis-induced cardiac injury.

Sepsis-induced cardiac injury lacks a mechanism-specific therapy, and loss of endogenous myocardial defense signaling may amplify inflammatory damage. We investigated whether recombinant neuregulin-1 (rNRG-1) protects against cecal ligation and puncture (CLP)-induced cardiac injury and whether stimulator of interferon genes (STING)-linked macrophage pyroptosis-associated signaling contributes to this response. Male mice underwent CLP or sham surgery. Myocardial NRG-1/ErbB4 signaling was assessed over time, rNRG-1 was administered intravenously at 25 μg/kg immediately before CLP, and cardiac fu

Occlusal disharmony attenuates inhibitory synaptic transmission in the medial prefrontal cortex and contributes to anxiety.

BACKGROUND: It has been well-validated that Occlusal Disharmony (OD) induces negative emotions, especially anxiety. While it leads to considerable frustration for both doctors and patients, its underlying mechanisms remain unclear, and effective treatment has been lacking. The present study aims to explore the pathological mechanisms of OD induced anxiety and to find a straightforward yet effective treatment strategy for this ostensibly intricate clinical phenomenon. MATERIAL AND METHODS: OD mice were established through binding a metal tube to their right mandibular incisor. Then, the mental

HER4 Affects Sensitivity to Tamoxifen and Abemaciclib in Luminal Breast Cancer Cells and Restricts Tumor Growth in MCF-7-Based Humanized Tumor Mice.

The impact of the HER4 receptor on the growth and treatment of estrogen receptor-positive breast cancer is widely uncertain. Using CRISPR/Cas9 technology, we generated stable HER4 knockout variants derived from the HER4-positive MCF-7, T-47D, and ZR-75-1 breast cancer cell lines. We investigated tumor cell proliferation as well as the cellular and molecular mechanisms of tamoxifen, abemaciclib, AMG232, and NRG1 treatments as a function of HER4 in vitro. HER4 differentially affects the cellular response to tamoxifen and abemaciclib treatment. Most conspicuous is the increased sensitivity of MCF

Erbb4 regulates the oocyte microenvironment during folliculogenesis.

Polycystic ovary syndrome (PCOS) is one of the most common endocrine disorders leading to infertility in women affecting reproductive, endocrine and metabolic systems. Recent genomewide association studies on PCOS cohorts revealed a single nucleotide polymorphism (SNP) in the ERBB4 receptor tyrosine kinase 4 gene, but its role in ovary development or during folliculogenesis remains poorly understood. Since no genetic animal models mimicking all PCOS reproductive features are available, we conditionally deleted Erbb4 in murine granulosa cells (GCs) under the control of Amh promoter. While we ha

The guanine nucleotide exchange factor VAV3 participates in ERBB4-mediated cancer cell migration.

ERBB4 is a member of the epidermal growth factor receptor (EGFR)/ERBB subfamily of receptor tyrosine kinases that regulates cellular processes including proliferation, migration, and survival. ERBB4 signaling is involved in embryogenesis and homeostasis of healthy adult tissues, but also in human pathologies such as cancer, neurological disorders, and cardiovascular diseases. Here, an MS-based analysis revealed the Vav guanine nucleotide exchange factor 3 (VAV3), an activator of Rho family GTPases, as a critical ERBB4-interacting protein in breast cancer cells. We confirmed the ERBB4-VAV3 inte

Genome-wide association study identifies ERBB4 on 2q34 as a novel locus associated with sperm motility in Japanese men.

BACKGROUND: The decrease in sperm motility has a potent influence on fertilisation. Sperm motility, represented as the percentage of motile sperm in ejaculated sperms, is influenced by lifestyle habits or environmental factors and by inherited factors. However, genetic factors contributing to individual differences in sperm motility remain unclear. To identify genetic factors that influence human sperm motility, we performed a genome-wide association study (GWAS) of sperm motility. METHODS: A two-stage GWAS was conducted using 811 Japanese men in a discovery stage, followed by a replication s

Regulation of Chloride Channels by Epidermal Growth Factor Receptor-Tyrosine Kinase Inhibitor-Induced α-Defensin 5.

Epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs) are used to treat non-small cell lung cancer with EGFR mutations. However, first-generation erlotinib and second-generation afatinib often cause diarrhea, which may develop because of the association between EGFR-TKIs and the chloride channel or abnormalities in the intestinal microbiota due to disruption of the intestinal immune system. As reports on the effects of EGFR-TKIs on intestinal immunity are lacking, we aimed to determine whether the intestinal immune system is involved in the molecular effects of EGFR-TKIs on c

Whole-exome sequencing of duodenal adenocarcinoma identifies recurrent Wnt/β-catenin signaling pathway mutations.

BACKGROUND: Genomic alterations of small bowel cancers remain poorly understood due to the rarity of these diseases. In the current study, the authors report the identification of somatic mutations from patients with duodenal adenocarcinoma by whole-exome sequencing. METHODS: Whole-exome sequencing and follow-up analysis were conducted in 12 matched tumor-normal tissue duodenal adenocarcinoma tissue pairs to examine the genetic characteristics of this disease. Somatic mutations (single-nucleotide variants and short insertion/deletions) were obtained and filtered and then searched for recurren

Brain-iT Immunotherapy: The Neurotransmitters Dopamine, Glutamate, GnRH-II and Neuropeptide Y Revive, Activate and Empower Healthy T Cells, Exhausted T Cells of Cancer Patients, and Genetically Engineered CAR T Cells.

Neurotransmitters are critical signaling molecules of the nervous system that activate their receptors within milliseconds, and convey accurate, coordinated, tightly regulated and important messages from one nerve cell, across a synapse, to another nerve, muscle or gland cell. It is time to realize that specific neurotransmitters also activate T cells directly and rapidly via their receptors in T cells. Subsequently, they decrease immune checkpoint inhibitors, increase expression of critical receptors, and induce a kaleidoscope of effector T cell functions. This review summarizes our discoveri

Understanding tumor microenvironment dynamics and immune checkpoint inhibitor efficacy following mEGFR-targeted near-infrared photoimmunotherapy.

Near-infrared photoimmunotherapy (NIR-PIT) is an innovative cancer treatment that provides both direct and selective killing of cancer cells and enhancement of the host's antitumor immune response. In Japan, EGFR-targeted NIR-PIT is already used clinically for head and neck cancer; however, there is limited understanding of the immune response kinetics of EGFR-targeted NIR-PIT. This limitation poses a significant barrier to further development of NIR-PIT, such as expanding its applications to other cancers and combination therapies. The purpose of this study is to establish a model incorporati

Impaired Neuregulin 1 Processing is Associated with Synaptic and Behavioral Abnormalities in a Prenatal Valproic Acid Model of Autism.

Autism spectrum disorder (ASD) is characterized by deficits in social communication and restricted/repetitive behaviors, yet the molecular mechanisms by which prenatal environmental insults lead to circuit dysfunction remain incompletely understood. Neuregulin 1 (NRG1)-ErbB4 signaling is a key regulator of synaptic development and excitation-inhibition (E/I) balance, but whether altered NRG1 processing contributes to ASD-related phenotypes remains unclear. Here, using a prenatal valproic acid (VPA) rat model, we examined the relationship between NRG1 processing, synaptic integrity, and behavio

TME-responsive nanoparticles co-targeting VCP, NETs, and dual immune checkpoints for immune revitalization in EGFR/PD-L1/CTLA-4-driven colorectal cancer.

Colorectal cancer (CRC) remains a formidable clinical challenge due to immune escape, metastasis, and resistance to conventional therapies. This study engineered a tumor microenvironment (TME)-responsive platform of orthogonally masked polyethylene glycol (omPEG)-coated solid lipid nanoparticles (SLNs) for targeted combinatorial immunotherapy in EGFR/PD-L1/CTLA-4-dysregulated CRC. The formulation co-delivered NMS-873 (NM), a VCP/p97 inhibitor, to induce endoplasmic reticulum stress (ERS) and proteostasis collapse, together with bispecific PD-L1/CTLA-4 aptamers (P1C4) for dual checkpoint blocka

Deprivation of EGFR signal causes senolysis in PDAC with CDK4/6 inhibition.

Approved KRAS inhibitors have shown limited therapeutic benefit over standard chemotherapy in PDAC and often encounter acquired resistance due to additional genetic alterations. RAS and RB1 functionally antagonize each other, which explains why RB1 is rarely mutated in KRAS-driven tumors. In PDAC cells, CDK4/6 inhibition induced cellular senescence accompanied by partial apoptosis. However, additional treatment with a senolytic agent or an ERK inhibitor promoted more efficient tumor cell elimination. While CDK4/6 inhibition downregulated KRAS activity, it concurrently upregulated EGFR signalin

ERBB4-Mediated Signaling Is a Mediator of Resistance to PI3K and BTK Inhibitors in B-cell Lymphoid Neoplasms.

BTK and PI3K inhibitors are among the drugs approved for the treatment of patients with lymphoid neoplasms. Although active, their ability to lead to long-lasting complete remission is rather limited, especially in the lymphoma setting. This indicates that tumor cells often develop resistance to the drugs. We started from a marginal zone lymphoma cell line, Karpas-1718, kept under prolonged exposure to the PI3Kδ inhibitor idelalisib until acquisition of resistance, or with no drug. Cells underwent transcriptome, miRNA and methylation profiling, whole-exome sequencing, and pharmacologic screeni

Synergistic cytotoxicity of the CDK4 inhibitor Fascaplysin in combination with EGFR inhibitor Afatinib against Non-small Cell Lung Cancer.

In the absence of suitable molecular markers, non-small cell lung cancer (NSCLC) patients have to be treated with chemotherapy with poor results at advanced stages. Therefore, the activity of the anticancer marine drug fascaplysin was tested against primary NSCLC cell lines established from pleural effusions. Cytotoxicity of the drug or combinations were determined using MTT assays and changes in intracellular phosphorylation by Western blot arrays. Fascaplysin revealed high cytotoxicity against NSCLC cells and exhibit an activity pattern different of the standard drug cisplatin. Furthermore,
