The Inhibitory Effect of Essential Oils on Rhizopus stolonifer, Trichophyton mentagrophytes, and Microsporum gypseum.

BACKGROUND: Rhizopus stolonifer is an opportunistic fungus that causes respiratory infections, sinusitis, and otomycosis. Trichophyton mentagrophytes and Microsporum gypseum cause athlete's foot, ringworm, and nail infections. Previous research has shown that some essential oils inhibit fungal growth. METHODS: We investigated the effects of 0.5% and 1% concentrations of Rosmarinus officinalis, Cinnamomum verum, Citrus paradisi, and Capsicum annuum extracts on the growth of R. stolonifer and T. mentagrophytes. The combined effects also were determined at 0.5% concentration for these fungal spe…

Isolation and characterization of a novel strain of Rhizopus arrhizus for effective biodegradation and detoxification of zearalenone.

Zearalenone (ZEN) is a prevalent estrogenic mycotoxin in cereals and feed, posing significant risks to feed safety and animal health. In this study, a food-associated filamentous fungus, Rhizopus arrhizus MNQW, was isolated from traditional Xiaoqu and systematically evaluated for its ZEN biodegradation efficiency and application potential. Under mild fermentation conditions, the strain removed over 98% of 5 mg/L ZEN in minimal salt medium within 36 h. Subcellular fractionation and inhibitor assays indicated that ZEN degradation was mainly mediated by heat- and protease-sensitive intracellular …

Fatal Pharyngeal Cellulitis Caused by Rhizopus microsporus in a Patient with Acute Myeloid Leukemia.

INTRODUCTION: Patients with immunodeficiency are highly susceptible to life-threatening fungal infections. Pharyngeal cellulitis caused by Rhizopus microsporus are exceptionally rare. CASE PRESENTATION: We reported a case of relapsed acute myeloid leukemia (AML) complicated by post-chemotherapy Rhizopus microsporus pharyngeal cellulitis. This cellulitis resulted in severe tissue necrosis, pharyngeal obstruction, and sub-sequent suffocation. Emergency bedside tracheotomy was administered after acute respiratory distress. The metagenomic next-generation sequencing (mNGS) identified Rhizopus mic…

Pathways of Pathogenicity: Transcriptional Stages of Germination in the Fatal Fungal Pathogen Rhizopus delemar.

Rhizopus delemar is an invasive fungal pathogen responsible for the frequently fatal disease mucormycosis. Germination, a crucial mechanism by which infectious spores of Rhizopus delemar cause disease, is a key developmental process that transforms the dormant spore state into a vegetative one. The molecular mechanisms that underpin this transformation may be key to controlling mucormycosis; however, the regulation of germination remains poorly understood. This study describes the phenotypic and transcriptional changes that take place over the course of germination. This process is characteriz…

Primary Cutaneous Mucormycosis Caused by Rhizopus oryzae: A Case Report and Review of Literature.

Mucormycosis is an invasive infection caused by opportunistic fungi. Rhizopus, Lichtheimia, Mucor and Rhizomucor are the most common isolated genera. Primary cutaneous mucormycosis is usually related to traumatic injuries, but immunocompromised cases are associated with underlying conditions such as diabetes mellitus and malignancies. The treatment of choice is surgical debridement and liposomal amphotericin B. We present a 40-year-old male with fever and a painful necrotic lesion on the middle back and history of poorly controlled diabetes mellitus. Rhizopus oryzae was isolated and identified…

A noninvasive renal fungus ball caused by Rhizopus--a previously unreported manifestation of zygomycosis.

We describe the first reported case of renal zygomycosis presenting as an isolated fungus ball (bezoar) without renal parenchymal invasion. Since all previous descriptions of renal involvement have discussed tissue invasion, our case is unique in that the infection was confined solely in the renal pelvis and extended to the distal ureter without signs of contiguous renal infection. Our patient later developed renal insufficiency while receiving amphotericin B Lipid Complex (ABLC). The therapy was changed to posaconazole with subsequent clinical, mycologic, and radiographic improvement and the

Disseminated Rhizopus microsporus infection in a patient on oral corticosteroid treatment: a case report.

A 71-year-old male with mild steroid-induced hyperglycaemia was diagnosed with a lethal invasive Rhizopus microsporus infection. Disseminated zygomycosis is a rare entity and is most frequently found in neutropenic patients with haematological malignancies, post-transplants or in patients on deferoxamine therapy. Infection is characterised by tissue infarction and necrosis due to angioinvasive hyphae. Culture of Zygomycetes is necessary for species determination but histology is a must to prove the infection. Ante-mortem diagnosis and culture is challenging and therefore mortality approaches 1…

Rhizopus arrhizus in Italy as the causative agent of primary cerebral zygomycosis in a drug addict.

A rare case of primary cerebral zygomycosis in an Italian drug addict is described. The diagnosis was based on the histologic detection of broad, aseptate, hyaline mycelium in fluid aspirated from a brain mass detected by computerized axial tomography. The zygomycete isolated from the clinical specimen was identified as Rhizopus arrhizus var arrhizus. The patient had no known predisposing condition that would have suppressed his immunological defenses. Once the diagnosis was established, treatment with amphotericin B was initiated, but the patient died during the first day of treatment.

Cutaneous Rhizopus infection. Occurrence as a postoperative complication associated with an elasticized adhesive dressing.

A 29-year-0ld woman in good health except for scoliosis suffered severe sequelae during the postoperative course for placement of a Harrington rod. A cutaneous Rhizopus infection in and about the incision site was attributed to the use of a contaminated elasticized adhesive (Elastoplast) dressing. The comtamination was established as a nosocomial problem, which is extremely difficult to control. The extent of the infection, subsequent long recovery course, and remarkable sequelae make this case unusual.

Structure-activity relationship studies of tanzawaic acid A, an antifungal agent against Rhizopus oryzae.

Through structure-activity relationship studies of tanzawaic acid A (TZAA), this research aims to contribute the development of potent antifungal drugs for treating mucormycosis infections. To this end, new TZAA analogs bearing chemical modifications in the side chain and/or C8-methyl group moieties were synthesized. These analogs are not accessible from natural TZAA and therefore required total synthesis. As a result, four of the newly synthesized TZAA analogs exhibited more potent and selective inhibitory activity against R. oryzae compared to TZAA.

Stable and reproducible homologous recombination enables CRISPR-based engineering in the fungus Rhizopus microsporus.

Mucormycosis is a lethal and emerging disease that has lacked a genetic model fulfilling both high virulence and the possibility of performing stable and reproducible gene manipulation by homologous recombination (HR). Here, we developed a new methodology to successfully perform HR in Rhizopus microsporus. We isolated an uracil auxotrophic recipient strain and optimized the critical steps in the genetic transformation of this fungus. This was followed by an adaptation of a plasmid-free CRISPR-Cas9 system coupled with microhomology repair templates. We reproducibly generated stable mutants in t…

Rhizopus nigricans polysaccharide activated macrophages and suppressed tumor growth in CT26 tumor-bearing mice.

In this study, a homogeneous polysaccharide (RPS-1) was extracted from liquid-cultured mycelia of Rhizopus nigricans. The weight-average molecular weight of RPS-1 was 1.617 × 107 g/mol and structural characterization indicated that RPS-1 was a non-starch glucan which consisted of a backbone structure of (1→4)-linked α-d-glucopyranosyl residues substituted at the O-6 position with α-d-glucopyranosyl branches. RPS-1 stimulated the production of nitric oxide and tumor necrosis factor-α by triggering phosphorylation of mitogen-activated protein kinases and nuclear translocation of nuclear factor k…

Genomic analysis of the basal lineage fungus Rhizopus oryzae reveals a whole-genome duplication.

Rhizopus oryzae is the primary cause of mucormycosis, an emerging, life-threatening infection characterized by rapid angioinvasive growth with an overall mortality rate that exceeds 50%. As a representative of the paraphyletic basal group of the fungal kingdom called "zygomycetes," R. oryzae is also used as a model to study fungal evolution. Here we report the genome sequence of R. oryzae strain 99-880, isolated from a fatal case of mucormycosis. The highly repetitive 45.3 Mb genome assembly contains abundant transposable elements (TEs), comprising approximately 20% of the genome. We predicted…

Cloning and functional characterization of the Rhizopus oryzae high affinity iron permease (rFTR1) gene.

Rhizopus oryzae is the most common etiologic agent of mucormycosis. Clinical and animal model data clearly demonstrate that the presence of elevated available serum iron predisposes the host to develop mucormycosis. Therefore, the high affinity iron permease (rFTR1) which encodes a protein required to scavenge iron from the environment, is highly likely to be a critical determinant of virulence for R. oryzae. We have cloned rFTR1 by using a PCR approach relying on degenerate primers designed from the conserved regions of Saccharomyces cerevisiae high affinity iron permease. Sequence analysis o…
