Enhanced bio-reduction of Cr(VI) using Shewanella putrefaciens CN32 mediated by Fe(III) minerals and riboflavin synergistically.

Iron minerals and the coupling of electron shuttle media can effectively overcome the problem of the insolubility of iron minerals and the higher cross-medium resistance consequently to enhance the bio-reduction rate of Cr(VI) by dissimilatory metal-reducing bacteria (DMRB). This study explored the potential synergistic enhancement of Cr(VI) bio-reduction by Shewanella putrefaciens CN32 in combination with three iron minerals (ferrihydrite, goethite and hematite) and riboflavin (RF). The addition of RF accelerates the transfer of electrons from bacterial cells to Fe minerals, which in turn pro…

Detection of SXT/R391 integrative conjugative elements carrying tigecycline resistance genes in Shewanella spp. isolated from retail seafood.

Tigecycline is a last resort antibiotic that is used to treat serious infections; however, some bacteria have developed tigecycline resistance by producing a tigecycline-inactivating enzyme or tigecycline resistance efflux pump, encoded by tet(X) and tmexCD-toprJ genes, respectively. Tons of seafood are consumed annually in China; however, whether seafood harbors tigecycline-resistant bacteria is not known. In this study, we isolated various tigecycline-resistant bacteria from retail seafood; among these, Shewanella was the predominant tigecycline-resistant genus (33/76, 43.4%). Genomic sequen…

Shewanella harboring antimicrobial and copper resistance genes in sea urchins (Paracentrotus lividus) from the Crozon peninsula (Brittany, France).

Shewanella is a genus of aquatic non-fermenting Gram-negative bacteria with increasing numbers of reports of infections in humans and appearance of antimicrobial resistant strains. Cases of infection show a relatively strong association with seafood consumption or exposure to seawater. This study aimed to analyze Shewanella spp. isolated from the sea urchin Paracentrotus lividus collected from the Crozon peninsula (France) with the intention of obtaining insights into the role of this genus as a reservoir of antimicrobial and heavy metal resistance genes. Five among seven Shewanella isolates w…

Fate of Fe and Cd upon microbial reduction of Cd-loaded polyferric flocs by Shewanella oneidensis MR-1.

Polyferric sulphate has been widely used for emergent control on incidental release of heavy metals such as Cd to surface water, causing precipitation of Cd-loaded polyferric flocs to the sediment. To date, little is known about whether the dissolution of the flocs in the presence of dissimilatory iron reducing bacteria (DIRB) can occur and how the dissolution influences the fate of Fe and Cd in the sediment. Here, we demonstrated that Shewanella oneidensis MR-1, as representative DIRB, has the ability to reduce the flocs, resulting in the release of Fe(2+) and Cd(2+) to the solution. Batch ex…

S(-II) reactivates Cd2+-stressed Shewanella oneidensis via promoting low-molecular-weight thiols synthesis and activating antioxidant defense.

Efficient remedies for living organisms including bacteria to counteract cadmium (Cd2+) toxicity are still highly needed. Plant toxicity studies have showed that exogenous S(-II) (including hydrogen sulfide and its ionic forms, i.e., H2S, HS-, and S2-) application can effectively alleviate adverse effects of Cd stress, but whether S(-II) could mitigate bacterial Cd toxicity remains unclear. In this study, S(-II) was applied exogenously to Cd-stressed Shewanella oneidensis MR-1 and the results showed that S(-II) can significantly reactivate impaired physiological processes including growth arre…

The role of electron shuttle enhances Fe(III)-mediated reduction of Cr(VI) by Shewanella oneidensis MR-1.

Chromate is one of the hazardous toxic pollutants. Reduction of Cr(VI) to Cr(III) has shown to reduce the toxicity of chromate. This work examined the reduction of Cr(VI) using an anaerobic batch cultures of Shewanella oneidensis MR-1 containing Fe(III). To do so, 10 mg/L Cr(VI) was reduced to Cr(III) within 3 days along with the oxidization of Fe(II) to Fe(III). The removal rate of Cr(VI) increased with increasing the concentration of Fe(III). In the absence of Cr(VI), the Fe(II) concentration of the batch culture increased with the growth of S. oneidensis MR-1. These data showed that S. onei…

Facilitated extracellular electron transfer of Shewanella loihica PV-4 by antimony-doped tin oxide nanoparticles as active microelectrodes.

Dissimilatory metal reducing bacteria are capable of extracellular electron transfer (EET) to insoluble metal oxides as external electron acceptors for their anaerobic respiration, which is recognized as an important energy-conversion process in natural and engineered environments, such as in mineral cycling, bioremediation, and microbial fuel/electrolysis cells. However, the low EET efficiency remains one of the major bottlenecks for its practical application. We report firstly that the microbial current generated by Shewanella loihica PV-4 (S. loihica PV-4) could be greatly improved that is …

Synergistic integration of cadmium sulphide and Mn3O4 nanozymes on Shewanella oneidensis for photocatalytic hydrogen production with reduced ROS toxicity.

The integration of semiconductor nanomaterials with electroactive bacteria offers a promising strategy for enhancing light-driven biohybrid systems. However, the generation of reactive oxygen species (ROS) during photocatalysis poses a significant challenge, impairing microbial viability and reducing process efficiency. In this study, we developed a novel biohybrid system by sequentially biosynthesizing cadmium sulphide (CdS) nanoparticles and manganese oxide (Mn3O4) nanozyme on the surface of Shewanella oneidensis MR-1, creating an S. oneidensis-CdS@Mn3O4 composite. The CdS nanoparticles faci…

Antibacterial activity and mechanism of astaxanthin crystals against Escherichia coli, Shewanella putrefaciens and its application in cooked pork preservation.

The objective of this work was to evaluate the antibacterial activity and mechanism of astaxanthin (AXT) crystals against Escherichia coli (E. coli) and Shewanella putrefaciens (S. putrefaciens). Scanning electron microscopy (SEM) showed that the bacteria after AXT action began to shrink, adhere, sag and other phenomena. The decrease of extracellular polysaccharide, the increase of protein content and conductivity, and FTIR analysis indicated that the action of AXT made the membrane protein structures change. Propidium iodide (PI) staining and fluorescence images revealed that the permeability…

Iron(III) minerals and anthraquinone-2,6-disulfonate (AQDS) synergistically enhance bioreduction of hexavalent chromium by Shewanella oneidensis MR-1.

Bioreduction of hexavalent chromium (Cr(VI)) to sparingly soluble trivalent chromium (Cr(III)) is a strategy for the remediation of Cr(VI) contaminated sites. However, its application is limited due to the slow bioreduction process. Here we explored the potential synergistic enhancement of iron(III) minerals (nontronite NAu-2, ferrihydrite, and goethite) and electron shuttle anthraquinone-2,6-disulfonate (AQDS) on the bioreduction of Cr(VI) by Shewanella oneidensis MR-1. AQDS alone increased the bioreduction rate of Cr(VI) by accelerating electron transfer from MR-1 to Cr(VI). Iron minerals al…

A plasmid-borne Shewanella algae Gene, qnrA3, and its possible transfer in vivo between Kluyvera ascorbata and Klebsiella pneumoniae.

The plasmid-borne quinolone resistance gene qnrA1 is prevalent in multidrug-resistant Enterobacteriaceae. A chromosomally encoded homologue in Shewanella algae, qnrA3, has been described. We isolated two qnrA3-positive strains, one of Klebsiella pneumoniae (He96) and one of Kluyvera ascorbata (Kas96), from the feces of an immunocompromised outpatient. The qnrA3 allele was identical to that of S. algae except for 5 nucleotides and differed from qnrA1 by 29 nucleotides affecting three amino acids. The analysis of the qnrA3 genetic environment showed that qnrA3 was inserted downstream from an ISC…
