The hippocampus is a brain area involved in many memory processes. This structure can also be affected in neurological diseases such as mesial temporal lobe epilepsy. A better understanding of its electrophysiological activity could benefit both the neuroscientific and clinical communities. We proposed, in a previous paper, a detailed bio-realistic conductance-based mathematical model of more than thirty thousand neurons to reproduce the main oscillatory features of the healthy hippocampus during slow-wave sleep and wakefulness, from slow to very fast frequencies. One big challenge of this mod

Objective.Hippocampal ripples are transient neuronal features observed in high-frequency oscillatory bands of local field potentials (LFPs), and they occur primarily during periods of behavioral immobility and slow-wave sleep. Ripples have been defined based on mathematically engineered features, such as magnitudes, durations, and cycles per event. However, the 'ripple' could vary from laboratory to laboratory because their definition is subject to human bias, including the arbitrary choice of parameters and thresholds. In addition, LFPs are often influenced by myoelectric noise arising from a

OBJECTIVE: The temporal evolution of periodic leg movements (PLM) and the relationship of their arousing effect on sleep episode has not been extensively investigated. We studied the nocturnal evolution of PLM associated or not with microarousal (MA) and associated with slow wave activity (PLM with slow wave activity) in 23 patients with PLM and/or restless legs syndrome (RLS).
METHODS: All subjects had PLM associated with MA or with slow wave activity as well as without MA and all slept for 4 sleep cycles. Spectral electroencephalogrpahic (EEG) analysis was done for the 4 sleep cycles to ass

STUDY OBJECTIVES: The changes in electroencephalogram (EEG) activity that characterize sleep and its sub-states-slow-wave sleep (SWS) and rapid eye movement (REM) sleep-are similar in mammals and birds. SWS is characterized by EEG slow waves resulting from the synchronous alternation of neuronal membrane potentials between hyperpolarized down-states with neuronal quiescence and depolarized up-states associated with action potentials. By contrast, studies of non-avian reptiles report the presence of high-voltage sharp waves (HShW) during sleep. How HShW relate to EEG phenomena occurring during

STUDY OBJECTIVES: Synchronization of neural activity within local networks and between brain regions is a major contributor to rhythmic field potentials such as the EEG. On the other hand, dynamic changes in microstructure and activity are reflected in the EEG, for instance slow oscillation (SO) slope can reflect synaptic strength. SO-spindle coupling is a measure for neural communication. It was previously associated with memory consolidation, but also shown to reveal strong interindividual differences. In studies, weak electric current stimulation has modulated brain rhythms and memory reten

BACKGROUND: Individuals with NREM parasomnias exhibit abnormal slow-wave activity and fragmented sleep. Sleep-state dissociation is the prevailing concept of NREM parasomnia-episodes; typically emerging from N3/N2 stages of NREM sleep's first cycle at the turning-point of deep sleep and arousal. While these relations provide a frame to understand these conditions, their mechanism and brain-topography remain unclear.
METHODS: We performed a systematic search of the literature (1/01/2015-20/06/2024) on brain-topographies and morphological changes based on neurophysiological and imaging studies

BACKGROUND: Initial observations with the human electroencephalogram (EEG) have interpreted slow oscillations (SOs) of the EEG during deep sleep (N3) as reflecting widespread surface-negative traveling waves that originate in frontal regions and propagate across the neocortex. However, mapping SOs with a high-density array shows the simultaneous appearance of posterior positive voltage fields in the EEG at the time of the frontal-negative fields, with the typical inversion point (apparent source) around the temporal lobe.
METHODS: Overnight 256-channel EEG recordings were gathered from 10 hea

The autonomic nervous system regulates cardiovascular activity during sleep, likely impacting cardiovascular health. Aging, a primary cardiovascular risk factor, is associated with cardiac autonomic disbalance and diminished sleep slow waves. Therefore, slow waves may be linked to aging, autonomic activity and cardiovascular health. However, it is unclear how sleep and slow waves are linked to cardiac autonomic profiles across multiple nights in older adults. We conducted a randomized, crossover trial involving healthy adults aged 62-78 years. Across 2 weeks, we applied auditory stimulation to

In the rabbit, microinjections of DSIP (delta-sleep peptide) at optimal doses in the median thalamus, periaqueductal gray matter (22.5 nmol), and nucleus of the tractus solitarius (15.0 nmol) produce slow-wave sleep with abundant recruiting spindles. As is seen with morphine, this effect is blocked by naloxone (160 micrograms intracerebrally; i.c.). At least in the rabbit, DSIP is probably a neurotransmitter or a neuromodulator of the bulbothalamic system inducing slow-wave sleep and particularly recruiting spindles. It is likely to restore slow-wave sleep after supraoptimal awakening i.c. mic

STUDY OBJECTIVES: Sleep disorders are common non-motor symptoms of Parkinson's disease (PD) that significantly impact patients' quality of life. Specifically, alterations in sleep microstructure - such as reduced slow-wave activity and sleep spindles - are prevalent in PD. The locus coeruleus (LC), the brain's primary source of noradrenaline, plays a pivotal role in regulating sleep and wakefulness and is highly vulnerable to neurodegeneration in PD. This study explores whether disruptions in sleep microarchitecture in PD are linked to LC degeneration.
METHODS: We assessed polysomnography for

Classical transection studies suggest that, in addition to the hypothalamus, the brainstem is essential for non-rapid eye movement (NREM) sleep. The circuits underlying this function, however, have remained largely unknown. We identified a circuit distributed in the midbrain, pons, and medulla that promotes NREM sleep in mice. We focused on the sublaterodorsal tegmentum, an area implicated in dual regulation of REM and NREM sleep. Transcriptomic and genetic analyses revealed that neurons positive for the neuropeptide neurotensin promote NREM sleep. Further analyses identified downstream NREM s

Sleep slow waves are known to participate in memory consolidation, yet slow waves occurring under anesthesia present no positive effects on memory. Here, we shed light onto this paradox, based on a combination of extracellular recordings in vivo, in vitro, and computational models. We find two types of slow waves, based on analyzing the temporal patterns of successive slow-wave events. The first type is consistently observed in natural slow-wave sleep, while the second is shown to be ubiquitous under anesthesia. Network models of spiking neurons predict that the two slow wave types emerge due

Non-rapid eye movement (NREM) sleep is not uniform but characterized by brief intrusions of wake-like brain activity and increased muscle tonus known as micro-arousals or brief awakenings. Although micro-arousals are an inherent feature of human and animal sleep, knowledge about the neural correlates of micro-arousals is sparse. We here developed an algorithm for automatic detection of micro-arousals based on EMG activity and used it to analyse the associated laminar neural activity in the motor cortex in mice. Our analysis showed that short micro-arousals with a duration below 5 s were associ

Slow oscillations (SOs; ~ 0.5-1.5 Hz) are a hallmark of non-rapid eye movement sleep and are known to support memory consolidation. Although their dynamics are well characterized, processes that precede SO initiation and predict their spatial extent remain largely unknown, despite their potential utility for closed-loop intervention. Using high-density EEG from 29 healthy adults, we examined neural activity in the 2-s interval preceding the SO trough. SOs were classified into Global and Frontal subtypes, which differ in propagation. We quantified instantaneous spectral power, time-frequency dy

Obstructive sleep apnea may lead to disordered sleep architecture and impair the physiologic slow wave sleep related growth hormone release. Obstructive sleep apnea occurs with craniofacial syndromes and in children with airway narrowing, pharyngeal hypoplasia, tonsillar adenoidal hypertrophy, micrognathia and achondroplasia. To examine the relationship between disordered sleep and growth hormone release we studied a 9 year old male with achondroplasia, growth failure (3 cm/year) and obstructive sleep apnea. Polysomnography data and a 20 min sampling for sleep entrained growth hormone showed b

Patients with Alzheimer's disease (AD) exhibit non-rapid eye movement (NREM) sleep disturbances in addition to memory deficits. Disruption of NREM slow waves occurs early in the disease progression and is recapitulated in transgenic mouse models of beta-amyloidosis.
