A primary mucinous cystoadenocarcinoma of the retroperitoneum.

A case of primary retroperitoneal mucinous cystoadenocarcinoma of the ovarian type in the presence of normal ovaries is reported. The histogenesis of this rare tumor has been uncertain. Special immunohistochemical stains done on our specimen show that the histogenesis of this tumor is most likely from mucinous metaplasia of coelomic mesothelium. Three cases of primary retroperitoneal mucinous cystoadenocarcinoma of the ovarian type have previously been reported in the English literature and are reviewed.

Simplified staining in situ of neurosecretory cells of the supraoesophageal ganglion of the female Culex pipiens molestus (Forskal) (Diptera: Culicidae).

The author describes the morphology and distribution of the neurosecretory cells in the supraoesophageal ganglion of the adult female Culex pipiens molestus, using paraldehyde fuchsin and paraldehyde thionine-paraldehyde fuchsin as vital staining techniques. The brain of Culex pipiens molestus has three regions (the proto-, deuto- and tritocerebrum) in which principally two types of neurosecretory cells (A and B) can be detected. Both A (dark) and B (light) cells are to be found in the protocerebrum, where they are termed medial neurosecretory cells, as they are distributed in the pars interce

Utility of semithin sections in the diagnosis of donovanosis.

The relative merits of tissue smears, paraffin sections and semithin plastic embedded sections in the diagnosis of donovanosis were assessed. In three cases reported negative or doubtful for the organisms by the former two methods, semithin plastic sections provided definite diagnosis. The latter method revealed in addition the organisms in epidermis and perivascular locations. From amongst the three stains: toluidine blue, Leishman's and thionine azure II basic fuchsin, the last mentioned was found to be the method of choice for demonstrating the organisms differently from the host cells.

Synaptic density on the axonal tree of a pyramidal cell in the cortex of the mouse.

The aim of this study was to investigate the density and distribution of synapses on the axonal tree of a pyramidal cell in the mouse cortex. The method used was a new variation of Golgi-electron microscopy involving the modification of Colonnier, the photochemical method of Blackstad and the use of phosphotungstic acid. A segment of axon collateral of 87 microns carried 17 synapses, the proximal 27 microns being free of synapses. The main axon was postsynaptic down to a length of 30 microns. On the following 163 microns there were six presynaptic elements. On the proximal parts of four other

Biological, chemical, immunological and staining properties of bacteria isolated from tissues of leprosy patients.

Two kinds of microorganisms are found in tissue of leprosy patients: Mycobacterium leprae (ML) and leprosy derived corynebacteria (LDC). ML from untreated patients has an alcohol-acid-fastness, which is lost upon treatment with antibiotics and immune response (tuberculoid leprosy). Vulnerable ML thus produced can be reversibly de-stained by organic solvent: in tissue sections from tuberculoid and treated patients, more bacteria are, thus, revealed by the Wade-Fite than by the Ziehl-Neelsen procedure. Organisms of genera Corynebacterium, Mycobacterium and Nocardia (CMN group), have DNA with %GC

Cytochemical comparison of immunologically characterized human leukaemia/lymphoma cell lines representing different levels of maturation.

Forty-seven human leukaemia/lymphoma cell lines belonging to myelocytic, monocytic, non-T/non-B, T-, and B-lineage and representing different levels of maturation as well as fresh cells from normal and leukaemic subjects were examined for immunological markers and cytochemically for acid phosphatase, alkaline phosphatase, alpha-naphthyl acetate esterase (pH 5.8 and 8.0), alpha-naphthyl butyrate esterase (pH 5.8 and 8.0), non-specific esterase, chloroacetate esterase, chymotrypsin-like protease, deoxyribonuclease II, beta-glucuronidase, sudan black, and periodic acid Schiff's staining. Spectral characteristics of metachromatically stained cartilage: effects of enzymatic degradation and dehydration.

When a microspectrophotometer was used to study cartilage, stained metachromatically with Azure A and Safranin O, the following 3 points were made: 1. the absorption maximum of stained cartilage matrix is identical to that reported for solutions of these dyes containing chondroitin sulfate; 2. partial enzymatic degradation of the cartilage matrix has no effect on the position of the absorption peak (although the intensity is greatly diminished); 3. ethanolic dehydration shifts the absorption peak somewhat toward the orthochromatic position, and decreases the intensity of the stain.

The identification of eosinophil colonies in soft-agar cultures by differential staining for peroxidase.

There has been a need to easily quantitate the incidence of eosinophil colonies within soft agar cultures. This has been realized by layering of the agar with benzidine dihydrochloride that permits detection of peroxidase activity in cells. Eosinophil colonies can be specifically identified by the addition to the substrate of potassium cyanide, an inhibitor of enzyme activity in neutrophils and monocytes. The enumeration of eosinophil colonies can be accomplished by scanning fresh or embedded cultures with low power magnification.
