Dental caries and vaccination strategy against the major cariogenic pathogen, Streptococcus mutans.

Although dental caries is a global problem in modern times, no vaccines are available for preventing these diseases. Among the bacterial pathogens that cause dental caries, including Streptococcus mutans, S. sobrinus, and Lactobacillus acidophilus, and Actinomyces viscosus, S. mutans is the most prominent and prevalent species. During the past, much effort has been focused on developing vaccines against S. mutans. Early attempts used fixed whole cells of S. mutans, but later it was found that serological cross-reactivity between heart tissue antigens and Streptococcus antigens occurs in patien…

Effect of Surface Modification of a Dental Composite on the Adhesion of Streptococcus mitis, Streptococcus mutans, and Candida albicans: An In Vitro Study.

Adhesion of different microorganisms to the surface of dental materials has generated significant interest since one of the most important requirements of biomaterials to be considered successful is their ability to withstand the damage caused by microorganisms that may lead to failure and the onset of different pathologies, such as caries. In vitro testing has demonstrated that surface modification is an alternative approach to reduce the adhesion of microorganisms to surfaces. The objective of this work was to assess the adhesion of Streptococcus mitis, Streptococcus mutans, and Candida albi…

Susceptibility of Streptococcus mutans and Streptococcus sobrinus to cell wall inhibitors and development of a novel selective medium for S. sobrinus.

Representative strains of Streptococcus mutans and Streptococcus sobrinus showed differences in susceptibility to members of the monobactam group of beta-lactam antibiotics: S. sobrinus was less sensitive than S. mutans. The minimum inhibitory concentrations of aztreonam (AZT) and carumonam, both of which belong to this group, were 2,000 microg/ml for S. sobrinus and 125 microg/ml for S. mutans. Further addition of fosfomycin, bacitracin and sodium chloride to Mitis Salivarius agar (MS) supplemented with AZT resulted in growth inhibition of S. mutans and oral streptococci other than S. sobrinu…

Anti-Cariogenic Potential of Lyophilized Cell-Free Supernatant of Probiotic Pediococcus pentosaceus GS4 against Streptococcus mutans and Streptococcus anginosus.

Streptococcus species, particularly Streptococcus mutans and Streptococcus anginosus, are recognized globally for their role in causing dental caries and other dental issues. Current treatments using fluoride products and conventional antibiotics are often ineffective due to the resilience of biofilms on tooth surfaces. To overcome these challenges, novel approaches that eliminate these pathogens are much needed. In this regard, we have prepared CFS (cell-free supernatant), a postbiotic preparation of our probiotic strain Pediococcus pentosaceus GS4, and investigated the anticariogenic propert…

Bacterial extracellular vesicles from Streptococcus mutans drive OSCC progression through β-catenin signaling.

Oral squamous cell carcinoma (OSCC) is an aggressive malignancy in which microbial dysbiosis is increasingly recognized as a contributing factor, yet the pathogenic potential of bacterial extracellular vesicles (EVs) remains poorly defined. To determine whether Streptococcus mutans-derived EVs (SmEVs) influence OSCC progression, we isolated and characterized SmEVs from ATCC 35,668 and examined their effects in CAL-27 cells and a nude-mouse xenograft model. SmEVs were readily internalized by OSCC cells, enhancing their proliferative, migratory, and invasive capacities in vitro. Transcriptomic a…

The CelB Gene Mediates Erythritol-Induced Inhibition of Exopolysaccharide Synthesis in Streptococcus mutans.

Streptococcus mutans is a primary contributor to dental caries due to its ability to form biofilms rich in extracellular polysaccharides (EPS). While erythritol has been recognized for its anti-cariogenic effects, the molecular pathways involved have remained unclear. In this study, we combined phenotypic and transcriptomic approaches to uncover the mechanism by which erythritol inhibits EPS synthesis. We found that erythritol treatment significantly reduces EPS production and biofilm density, and that these changes are accompanied by marked downregulation of the phosphotransferase system (PTS…

From adhesion to biofilm: strain-specific and environmental factors shaping Streptococcus mutans biofilms.

Streptococcus mutans is a key pathogen in dental caries and adheres to tooth surfaces via the surface protein SpaP, which binds salivary agglutinin (SAG). However, the extent to which adherence properties influence subsequent biofilm formation remains unclear. This study investigated biofilm formation of S. mutans strains with varying adherence properties on glass surfaces with or without a crude SAG (cSAG) coating prepared from human parotid saliva, under static or laminar flow conditions, and in mono- or multispecies conditions. Green-fluorescent protein-labeled S. mutans strains (V403, NG8,…

α2-3-Sialylated Glycoproteins Attenuate Streptococcus mutans Virulence and Associated Host Inflammatory Responses.

Glycans attached to host glycoproteins play an important role in regulating oral microbial colonization and maintaining community balance. Our previous studies revealed that children exhibit lower levels of α2-3 sialylated glycan structures (SAα2-3Gal) than adults, raising the possibility that this age-associated glycan deficiency contributes to the heightened cariogenicity of Streptococcus mutans. In this study, sialylated glycoproteins (Sia-GP) and corresponding desialylated controls (DeSia-GP and α2,3-DeSia-GP) were prepared from bovine milk-derived glycoproteins to investigate the function…

Natural compounds: new therapeutic approach for inhibition of Streptococcus mutans and dental caries.

Streptococcus mutans is recognized as one of the leading causes of dental caries, and biofilm formation by this bacterium plays a key role in dental plaque development and caries progression. Given the increasing resistance of bacteria to antibiotics and the adverse effects of some synthetic antimicrobials, the search for natural alternatives has received increasing attention. The recently published studies have demonstrated that natural compounds (NCs) such as curcumin, cinnamaldehyde, eugenol, thymol, carvacrol, epigallocatechin gallate, farnesol, catechin, inulin, menthol, apigenin, myricet…

Molecular insights into antibiofilm inhibitors of Streptococcus mutans glucosyltransferases through in silico approaches.

Streptococcus mutans, a primary cariogenic bacterium, plays a central role in dental caries, one of the most widespread chronic diseases globally. Glucosyltransferases (GTFs) are key virulence factors in this process, as they synthesize extracellular polysaccharides that contribute to biofilm formation and pathogenicity. Targeting GTFs has emerged as a promising strategy for preventing dental caries, with previous studies demonstrating its potential efficacy. This study builds on our prior work by providing detailed molecular insights into the binding modes of previously identified GTF inhibit…

Functional Characterization of Residues Affecting the Catalytic Activity of Glucosyltransferase from Streptococcus mutans.

Dental caries is a multifactorial, biofilm-mediated disease primarily caused by Streptococcus mutans, a key etiological agent. This bacterium secretes extracellular enzymes known as glucosyltransferases (Gtfs), also termed glucansucrases, which play a pivotal role in the synthesis of exopolysaccharides through the metabolism of dietary sucrose. These exopolysaccharides provide binding sites for the attachment and colonization of other microorganisms, contributing to the initiation and progression of dental caries. This study investigates the catalytic mechanisms of glucosyltransferases from S.…

Membrane vesicle production via cell-to-cell communication-induced autolysis in Streptococcus mutans.

Membrane vesicles (MVs) released from bacteria into the extracellular environment play various roles in bacterial and bacterial-host interactions. In Gram-positive bacteria, cell wall degradation or damage is crucial for cytoplasmic MV (CMV) release because the cytoplasmic membrane must pass through the thick cell wall to the extracellular space. Although Streptococcus mutans, a major causative agent of dental caries, is known to release CMVs that are involved in pathogenicity and biofilm formation, the mechanism by which CMV production is triggered is unknown. In this study, we demonstrate th…

Streptococcus mutans regulates ubiquitin modification of Candida albicans in the bacterial-fungal interaction.

The ecological interplay between Streptococcus mutans and Candida albicans within dental plaque biofilms is an important factor driving pathogenesis of dental caries. This study aimed to investigate S. mutans regulation of C. albicans growth and virulence through extracellular membrane vesicles (EMVs) and modulation of ubiquitination, a key protein post-translational modification. We established a transwell co-culture model to enable "contact-independent" interactions between S. mutans and C. albicans. S. mutans EMVs were found to directly associate with C. albicans cells and promote biofilm f…
