[Changes in plasticity of rat hypothalamic neurons and astrocytes in humid and hot environment].

OBJECTIVE: To study the changes in the plasticity of the neurons and astrocytes in the paraventricular nucleus (PVN) and supraoptic nucleus (SON) of the hypothalamus of rats exposed to a humid and hot environment. METHODS: The rats were subjected to stimulation with a humid and hot environment for 120 min in a climate chamber (dry bulb temperature of 40.0-/+0.5 degrees C with relative humidity of 60-/+5%). During the exposure, the behavioral responses of the rats were observed, and the changes in the expressions of Fos and GFAP in the PVN and SON in response to the exposure evaluated using im…

Immunocytochemical study of the hypothalamo-neurohypophysial system. II. Distribution of neurophysin, vasopressin and oxytocin in the normal and osmotically stimulated rat.

Antisera, with cross reactive antibodies removed by affinity chromatography, were used in the immunoperoxidase-bridge technique to study the distribution of oxytocin and vasopressin together with neurophysin in the hypothalamo-neurohypophysial system of the rat. The hormones were demonstrated in different areas of the supraoptic nucleus (SON) and paraventricular nucleus (PVN), in neurosecretory fibres of the hypothalamo-neurohypophysial tract, median eminence, and in nerve terminals of the neurohypophysis. Intact normal and rats with hereditary hypothalamic diabetes insipidus (Brattleboro stra…

Excitability of oxytocin neurons in paraventricular nucleus is regulated by voltage-gated potassium channels Kv4.2 and Kv4.3.

Oxytocin is produced by neurons in the paraventricular nucleus (PVN) and the supraoptic nucleus in the hypothalamus. Various ion channels are considered to regulate the excitability of oxytocin neurons and its secretion. A-type currents of voltage-gated potassium channels (Kv channels), generated by Kv4.2/4.3 channels, are known to be involved in the regulation of neuron excitability. However, it is unclear whether the Kv4.2/4.3 channels participate in the regulation of excitability in PVN oxytocin neurons. Here, we investigated the contribution of the Kv4.2/4.3 channels to PVN oxytocin neuron…

A hypothalamic VMPO-supraoptic vasopressin circuit mediates procalcitonin-induced fluid imbalance.

Sepsis is a life-threatening condition characterized by infection-induced organ dysfunction, with fluid imbalance and cardiovascular instabi … that VMPOOprk1 neurons project to and activate arginine vasopressin (AVP)-expressing neurons in the supraoptic nucleus (SONAVP), leading to increased blood pressure. Together, our findings define a PCT-sensitive VMPOOprk1→SONAVP neural circuit that integrates fluid balance and cardiovascular regulation. Our data highlight critical role of the brain in coordinating o …

Hypothalamic Oxytocin Neuronal Activation Induces Bipolar-Like Mood Changes in Mice in a Sex- and Dosage-Dependent Manner.

Clinical studies have suggested that increased plasma oxytocin (OT) levels are a promising biomarker for bipolar disorder (BD), and our earlier post-mortem study found increased OT activity in the hypothalamic paraventricular nucleus (OTPVN) in BD. However, the potential contribution of the supraoptic nucleus (SON, OTSON), a major part of the central OT system, to BD remains unknown. We therefore systematically performed independent acute or chronic chemogenetic activation of OTPVN, OTSON, or OTPVN+SON experiments in OT-cre mice. We found that acute activation of OTPVN+SON neurons led to sligh…

Efferent projections from the parabrachial nucleus demonstrated with the anterograde tracer Phaseolus vulgaris leucoagglutinin.

Efferent projections from the parabrachial complex (PBN) were studied in the rat using the anterograde tracer, Phaseolus vulgaris leucoagglutinin (PHA-L). Projections to the hypothalamus (ventromedial, dorsomedial, paraventricular, and supraoptic nuclei) originate primarily in the lateral PBN (1PBN). The amygdalar central nucleus (ACE) receives strong projections from all parts of the PBN although the external 1PBN projects primarily to the lateral ACE. Whereas the projections to the lateral bed nucleus of the stria terminalis, median preoptic nucleus, diagonal band of Broca, and lateral preop

Sex-specific regulation of social play in juvenile rats by oxytocin neurons in the hypothalamus and oxytocin signaling in the nucleus accumbens.

Social play is a rewarding behavior shown across juvenile mammalian species and is important for the development of social competency throughout the lifespan. The neuropeptide oxytocin (OXT) regulates various social behaviors and is being used in clinical trials to improve social functioning. However, the role of OXT in juvenile social play is largely unknown. To address this gap, we determined the involvement of hypothalamic OXT-producing neurons in the supraoptic nucleus (SON) and paraventricular nucleus (PVN), PVNOXT projections to the nucleus accumbens (NAc), and OXT signaling within the N…

Therapeutic uses of oxytocin in stress-related neuropsychiatric disorders.

Oxytocin (OXT), produced and secreted in the paraventricular nucleus and supraoptic nucleus of magnocellular and parvocellular neurons. The diverse presence and activity of oxytocin suggests a potential for this neuropeptide in the pathogenesis and treatment of stress-related neuropsychiatric disorders (anxiety, depression and post-traumatic stress disorder (PTSD)). For a more comprehensive understanding of the mechanism of OXT's anti-stress action, the signaling cascade of OXT binding to targeting stress were summarized. Then the advance of OXT treatment in depression, anxiety, PTSD and the m…

Attenuation of the polypeptide 7B2, prohormone convertase PC2, and vasopressin in the hypothalamus of some Prader-Willi patients: indications for a processing defect.

7B2 is a neuroendocrine chaperone interacting with the prohormone convertase PC2 in the regulated secretory pathway. Its gene is located near the Prader-Willi syndrome (PWS) region on chromosome 15. In a previous study we were able to show 7B2 immunoreactivity in the supraoptic nucleus (SON) or the paraventricular nucleus (PVN) in only three of five PWS patients. Here we report that in contrast with five other PWS patients, the neurons in the hypothalamic SON and PVN of the two 7B2-immunonegative PWS patients also failed to show any reaction using two antibodies directed against processed vaso…

The subfornical organ's neural connections and their role in water balance.

The subfornical organ (SFO) has projections to specific sets of nuclei within the preoptic area and hypothalamus which enable it to influence behavioral and physiological controls of water balance. It projects to the nuclei of the anteroventral third ventricular area, to vasopressinergic (heavily) and oxytocinergic (moderately) magnocellular neurons of the supraoptic and paraventricular nucleus. It also projects to the parvocellular areas of the paraventricular nucleus which project to the median eminence and to all the motor nuclei of the autonomic nervous system. In addition the SFO projects

Lack of cross-tolerance between haloperidol and clozapine towards Fos-protein induction in rat forebrain regions.

We investigated whether the acute effects of haloperidol and clozapine on Fos expression in the rat forebrain are mediated by the same receptors through evaluation of cross-tolerance, particularly in the commonly affected areas. Acutely administered haloperidol (1 mg/kg. i.p.) and clozapine (20 mg/kg, i.p.) induce regionally different (e.g., the striatum, the hypothalamic paraventricular and supraoptic nuclei, and the central amygdala) and overlapping (e.g., the nucleus accumbens and the lateral septum) patterns of Fos-protein distribution in the rat forebrain. After long-term treatment, part

Chronic intermittent hypoxia modulates nNOS mRNA and protein expression in the rat hypothalamus.

Exposure to chronic intermittent hypoxia (CIH) as observed in obstructive sleep apnea (OSA) elicits a sustained elevation of sympathetic act … rgan and the periventricular hypothalamic nucleus of the CIH rats, but no significant change in the supraoptic nucleus or the lateral hypothalamic area. This work suggests that examination of central regulation of sympathetic activity may help elucidate the mechanisms of hypertension after CIH.

Lack of translation of normal 7B2 mRNA levels in hypothalamic mutant vasopressin cells of the homozygous Brattleboro rat.

The homozygous Brattleboro rat (di/di) synthesizes a vasopressin (VP) precursor with an aberrant C-terminus, which causes a hypothalamic form of diabetes insipidus. The neuroendocrine polypeptide 7B2 is present in VP and oxytocin (OT) neurons of the supraoptic and paraventricular nucleus of the hypothalamus in wild type rats. However, in the di/di rat 7B2 immunoreactivity is absent in the VP cell population, whereas 7B2 levels within the OT cells are unaffected. Remarkably, there is no obvious difference in 7B2 transcript levels between VP and OT neurons in the di/di rat hypothalamus. This stu

Hereditary diabetes insipidus: an immunohistochemical study of the hypothalamus and pituitary gland.

We report the histological findings in a case of hereditary diabetes insipidus (HDI) using vasopressin (VP) immunohistochemistry. The hypothalamus displayed a marked loss of magnocellular VP neurons, with preservation of the smaller cells. The neurohypophysis was severely atrophic with scanty immunoreactivity. Our results support the hypothesis that HDI results from a selective degeneration of VP neurons affecting chiefly the magnocellular elements projecting to the neurohypophysis. The sparing of the parvocellular component may reflect the projection of these neurons to non-pituitary targets.
