Anti-Inflammatory, Anti-Oxidative and Anti-Apoptotic Effects of Thymol and 24-Epibrassinolide in Zebrafish Larvae.

Thymol (THY) and 24-epibrassinolide (24-EPI) are two examples of plant-based products with promising therapeutic effects. In this study, we investigated the anti-inflammatory, antioxidant and anti-apoptotic effects of the THY and 24-EPI. We used zebrafish (Danio rerio) larvae transgenic line (Tg(mpxGFP)i114) to evaluate the recruitment of neutrophils as an inflammatory marker to the site of injury after tail fin amputation. In another experiment, wild-type AB larvae were exposed to a well known pro-inflammatory substance, copper (CuSO4), and then exposed for 4 h to THY, 24-EPI or diclofenac (D…

The biosynthesis of thymol, carvacrol, and thymohydroquinone in Lamiaceae proceeds via cytochrome P450s and a short-chain dehydrogenase.

Thymol and carvacrol are phenolic monoterpenes found in thyme, oregano, and several other species of the Lamiaceae. Long valued for their smell and taste, these substances also have antibacterial and anti-spasmolytic properties. They are also suggested to be precursors of thymohydroquinone and thymoquinone, monoterpenes with anti-inflammatory, antioxidant, and antitumor activities. Thymol and carvacrol biosynthesis has been proposed to proceed by the cyclization of geranyl diphosphate to γ-terpinene, followed by a series of oxidations via p-cymene. Here, we show that γ-terpinene is oxidized by…

Impact of Moderate Heat, Carvacrol, and Thymol Treatments on the Viability, Injury, and Stress Response of Listeria monocytogenes.

The microbial safety and stability of minimally processed foods are based on the application of combined preservative factors. Since microorganisms are able to develop adaptive networks to survive under conditions of stress, food safety may be affected, and therefore understanding of stress adaptive mechanisms plays a key role in designing safe food processing conditions. In the present study, the viability and the sublethal injury of Listeria monocytogenes exposed to moderate heat (55 °C) and/or essential oil compounds (carvacrol and thymol, 0.3 mM) treatments were studied. Synergistic effect…

Ecotoxicity evaluation of thymol in tunneller dung beetles (Coleoptera: Geotrupidae) as a natural alternative to ecotoxic anthelmintics.

Synthetic anthelmintics widely used in livestock production are largely excreted unchanged in faeces, where they can exert sublethal and lethal effects on non-target organisms such as dung beetles. Phytochemical compounds, including thymol (THY), have been proposed as environmentally compatible alternatives; however, information on their ecotoxicological safety across different dung beetle taxa remains limited. We assessed the sublethal effects of dietary thymol on Thorectes lusitanicus (Coleoptera: Geotrupidae), a flightless tunneller dung beetle considered functionally sensitive to veterinar…

Preparation of controlled-released starch-alginate microspheres loaded with thymol nanoemulsions and their therapeutic effects on salmonellosis.

The rise of antimicrobial resistance has spurred interest in natural alternatives such as thymol, a potent broad-spectrum antibacterial agent. However, its clinical potential for treating enteric infections is severely limited by poor stability, low bioavailability, inherent cytotoxicity, and non-targeted release. To overcome these challenges, thymol nanoemulsion was firstly prepared and then incorporated into starch‑sodium alginate microspheres (TMS). This system was comprehensively characterized and evaluated for its antibacterial efficacy, safety, and therapeutic performance both in vitro a…

Thymol-Functionalized Hyaluronic Acid as Promising Preservative Biomaterial for the Inhibition of Candida albicans Biofilm Formation.

Hyaluronic acid (HA) is a naturally occurring biopolymer that has been employed for a plethora of medicinal applications. Nevertheless, as HA is a natural polysaccharide, it can be a substrate able to promote microbial growth and proliferation. Biopolymer-drug conjugates have gained attention over the years to overcome drawbacks of each single component. Within this context, thymol (Thy), a phenolic compound occurring in essential oils (EOs) extracted from Thymus and Origanum, has been largely studied for its antimycotic applications. However, it is characterized by a low water solubility and …
