Drug patch testing in Stevens-Johnson syndrome and toxic epidermal necrolysis: A systematic review.

BACKGROUND: The data on patch testing (PT) to identify culprit medications in Stevens-Johnson syndrome/toxic epidermal necrolysis (SJS/TEN) are limited to scattered case reports and small case series, without analysis of overall trends to inform clinicians of its utility, methodology, and safety. OBJECTIVE: To conduct a systematic review of the practice of PT in SJS/TEN, quantify the positivity rate of common drug classes, and assess safety during testing. METHODS: PubMed was searched from inception to 2021. Search terms included "patch testing" AND "SJS" OR "TEN" OR "Stevens-Johnson syndrom

Evaluation of in vitro rat and human airway epithelial models for acute inhalation toxicity testing.

In vivo models (mostly rodents) are currently accepted by regulatory authorities for assessing acute inhalation toxicity. Considerable efforts have been made in recent years to evaluate in vitro human airway epithelial models (HAEM) as replacements for in vivo testing. In the current work, an organotypic in vitro rat airway epithelial model (RAEM), rat EpiAirway, was developed and characterized to allow a direct comparison with the available HAEM, human EpiAirway, in order to address potential interspecies variability in responses to harmful agents. The rat and human models were evaluated in 2

SOBA: Development and testing of a soluble oligomer binding assay for detection of amyloidogenic toxic oligomers.

The formation of toxic Amyloid β-peptide (Aβ) oligomers is one of the earliest events in the molecular pathology of Alzheimer's Disease (AD). These oligomers lead to a variety of downstream effects, including impaired neuronal signaling, neuroinflammation, tau phosphorylation, and neurodegeneration, and it is estimated that these events begin 10 to 20 y before the presentation of symptoms. Toxic Aβ oligomers contain a nonstandard protein structure, termed α-sheet, and designed α-sheet peptides target this main-chain structure in toxic oligomers independent of sequence. Here we show that a desi

Zebrafish as an Animal Model for Ocular Toxicity Testing: A Review of Ocular Anatomy and Functional Assays.

Xenobiotics make their way into organisms from diverse sources including diet, medication, and pollution. Our understanding of ocular toxicities from xenobiotics in humans, livestock, and wildlife is growing thanks to laboratory animal models. Anatomy and physiology are conserved among vertebrate eyes, and studies with common mammalian preclinical species (rodent, dog) can predict human ocular toxicity. However, since the eye is susceptible to toxicities that may not involve a histological correlate, and these species rely heavily on smell and hearing to navigate their world, discovering visua

The use of multiwell culture plates in the duckweed toxicity test-A case study on Zn nanoparticles.

Extensive production of nanomaterials of various properties needs to be coupled with rapid toxicity testing in order to provide information about their potential risks to the environment and human health. Miniaturization of toxicity tests may accelerate economical testing of nanomaterials, but is not a common practice. We describe a case study to miniaturize a commonly used toxicity test with plant duckweed Lemna minor. 6-well, 12-well and 24-well culture plates were used to assess their potential use for the duckweed toxicity test with potassium chloride as reference material. The results wer…

Toxicity testing and drug screening using iPSC-derived hepatocytes, cardiomyocytes, and neural cells.

Unexpected toxicity in areas such as cardiotoxicity, hepatotoxicity, and neurotoxicity is a serious complication of clinical therapy and one of the key causes for failure of promising drug candidates in development. Animal studies have been widely used for toxicology research to provide preclinical security evaluation of various therapeutic agents under development. Species differences in drug penetration of the blood-brain barrier, drug metabolism, and related toxicity contribute to failure of drug trials from animal models to human. The existing system for drug discovery has relied on immort

Burrowing mayfly Ephemera orientalis (Ephemeroptera: Ephemeridae) as a new test species for pesticide toxicity.

The potential of mayfly Ephemera orientalis McLachlan eggs and first-instar larvae in ecotoxicological testing was investigated. Both stages of E. orientalis showed high tolerance to various environmental variables, such as water temperature, pH, water hardness, and dissolved organic carbon. Toxicological assays were conducted with three insecticides (emamectin benzoate, endosulfan, and cypermethrin), one fungicide (mancozeb), and one herbicide (paraquat dichloride). The two toxicity endpoints for the assay were the 14-day egg median hatching rate (EHC50) in static and renewal exposure systems

Visual function test for early detection of ethambutol induced ocular toxicity at the subclinical level.

CONTEXT: Tuberculosis in developed countries is on the rise, and the main treatment ethambutol is known to induce ocular toxicity. However, to date, there are unknown tests or protocols for detecting subclinical ethambutol-induced ocular toxicity, which is important as early detection is related to symptom reversibility. We defined the ethambutol induced ocular toxicity as stastically siginificant change of visual function which was induced by ethambutol. OBJECTIVE: We aimed to identify a visual function test for the early detection of subclinical ethambutol-induced ocular toxicity. Furthermo

Neuropsychological deficits in patients diagnosed with chronic toxic encephalopathy: which tests are most discriminative?

Chronic toxic encephalopathy (CTE), a syndrome affecting the central nervous system, has been associated with long-term occupational exposure to organic solvents. We studied the discriminative power of a series of cognitive tests in separating 20 patients exposed to organic solvents, diagnosed with CTE, from 27 non-CTE patients with similar symptoms and exposure, and 19 healthy referents. All subjects were examined with a test battery assessing various cognitive and psychomotor functions. The groups were comparable with regard to age and level of schooling. Stepwise discriminant analyses indic

[Value of the thyroliberin test in the evaluation of the results of surgical treatment of patients with diffuse toxic goiter].

The time course of the basal level of the thyrotropic hormone (b TSH) and its concentration after thyroliberin (TRH) stimulation was studied in 28 patients with diffuse toxic goiter suffering from severe thyrotoxicosis before the onset of drug therapy, directly before operation, early and long after the operation. The TRH test was shown as a diagnostic test of efficacy of drug therapy. A higher level of b TTH with a lowered reaction to TRH in the first 6 mos. after operation was of compensatory nature requiring no substitution therapy. The restoration of the endocrine axis hypothalamus-pituita

Staphylococcus aureus associated with toxic shock syndrome: phage typing and toxin capability testing.

Phage type 29 Staphylococcus aureus was identified singly or with type 52 in 64.1% of 248 coded isolates from patients with toxic shock syndrome. These phage types also have a high capability of producing pyrogenic exotoxin C and enterotoxin F. The origin and development of these toxigenic strains were explored by studying 25,220 isolates of S. aureus stored in a staphylococcal bank between 1960 and 1979. A small percentage of phage types 29, 52 were found in 1960, but their prevalence increased between 1961 and 1970, and continued at elevated levels through 1979. The toxigenic capabilities of

Rapid Turnaround Time for Toxic Alcohol Testing Is Associated with Shorter Hospital Length of Stay.

INTRODUCTION: Toxic alcohol poisoning (methanol or ethylene glycol) can cause severe metabolic disturbances, end-organ damage, and death. Timely diagnosis is paramount and predicated on history, lab markers, and confirmatory laboratory testing, which inform antidote use and/or hemodialysis. Confirmatory toxic alcohol testing capabilities and turnaround times vary by geographic region. Our objective in this study was to evaluate how differences in toxic alcohol testing turnaround times impact our primary outcome of hospital length of stay and secondary outcomes of antidote (fomepizole) administ

Integrated in vitro/in silico test battery for the prediction of chemical-induced cholestatic liver toxicity.

Cholestatic liver toxicity induced by chemicals is a multisectoral concern and remains a challenge to predict, partly due to a lack of mechanistic understanding of chemical-induced cholestasis. In the context of modern chemical safety, there has been a demand to step away from animal testing and promote alternative mechanistically-anchored approaches. Based on a previously introduced adverse outcome pathway network and using 7 (non-)cholestatic reference test chemicals, the present study built the initial iteration of a test battery integrating in vitro and in silico approaches. The former inv

Application to developmental toxicity testing of a novel method for whole-brain imaging of microglia in zebrafish.

Microglia, parenchymal macrophages resident in the central nervous system, regulate brain development by dynamically changing their functional and morphological states in a spatiotemporal-dependent manner. Because the function of microglia may differ depending on their location, their status should ideally be assessed within discrete brain regions. In this study, we developed a novel whole-brain imaging method to visualize microglial morphology in the forebrain, midbrain, and hindbrain of live zebrafish larvae at 5-6days post-fertilization, and quantified various morphological parameters using

A study protocol for chemical analysis and toxicity testing of virgin and recycled microplastics and associated chemicals.

Plastics can contain a variety of different chemicals, which are either intentionally (IAS) or non-intentionally (NIAS) added substances. Recycled plastics can contain especially NIAS, which might vary in amount and characteristics, possibly compromising the applicability of the plastics. As plastics can eventually degrade into microplastics, these substances can be released into their environment and upon human exposure, pose a threat to human health. Therefore, simple methods for the comprehensive monitoring of these chemicals are needed to guarantee the safe use of plastics. The purpose of

Case Report: Persistent toxic reactions in a toddler with a negative blood cantharidin toxicology test.

Cantharidin is a potent natural toxin and has been used in traditional Chinese medicine to treat cancers and various ailments such as rabies or psoriasis. However, improper use of cantharidin can easily lead to poisoning. Despite there is an increasing amount of literature on the toxic mechanisms of cantharidin, there is still limited knowledge about the disease progression, toxic reactions and toxic blood concentrations in children with cantharidin poisoning. Caregivers of children suffering from cantharidin poisoning might fail to provide an accurate exposure dose when the children exhibit s

Thamnophis sirtalis and their toxic relationship: Testing for intraspecific venom variation in Common Garter Snakes.

Intraspecific phenotypic variation can be used as a window into the ecological differences among individuals of a species and lead to a better understanding of adaptive evolution. Adaptive traits, such as venom, that play an important ecological role for a species are useful models for understanding the sources of intraspecific variation. Intraspecific studies on front-fanged venomous snakes have offered deeper insights into the diverse mechanisms and adaptations that support the effectiveness of venom across species. Despite the extensive research on front-fanged venomous snakes, rear-fanged

Comparative investigation of toxicity induced by UV-A and UV-C radiation using Allium test.

Organisms are increasingly exposed to ultraviolet (UV) rays of sunlight, due to the thinning of the ozone layer and its widespread use in sterilization processes, especially against the SARS-CoV-2 virus. The present study was conducted with the purpose of evaluating the damages of UV-A and UV-C radiations in Allium cepa L. roots. The effects of two different types of UV on some physiological, biochemical, cytogenotoxic, and anatomical parameters were investigated in a multifaceted study. Three groups were formed from Allium bulbs, one of which was the control group. One of the other groups was
